n aquesta edició de l’El· lipse presentem el genoma del gat domèstic comparat amb el del gat salvatge, un estudi publicat a PNAS, amb la participació de Tomàs Marquès Bonet de l’IBE (CSIC-UPF). També la comparació detallada del genoma del ratolí i l’humà, publicada a Nature, on va participar l’equip de Roderic Guigó (CRG). Pensant en els ratolins i gats, em ve a la ment una petita faula de Franz Kafka.

«Ah», va dir el ratolí, «el món es fa cada dia més estret. De primer era tan ample que vaig tenir por, i vaig continuar corrent i vaig ser feliç quan per fi vaig veure en la llunyania murs a dreta i esquerra, però aquests murs allargats s’acosten tan de pressa l’un a l’altre que jo ja sóc a la darrera cambra, on hi ha el racó amb la trampa cap a la qual corro». «Només has de canviar de direcció», va dir el gat, i se’l va cruspir.

Igual que en molts dels contes de Kafka, el tema és l’error en jutjar la realitat i el fracàs. Els investigadors a Espanya estan paralitzats davant un parany o haurien d’intentar, amb confiança, evitar quedar-se tancats entre les parets? Potser el gat té la resposta.

n this issue of El·lipse we present the genome of the domestic cat compared to the wild cat. This is based on a study published in PNAS by a team including Tomàs Marqués Bonet from the IBE (CSIC-UPF). We also show the results of a detailed comparison of the mouse and human genomes, published in the latest issue of Nature, involving Roderic Guigó’s team at the CRG. Thinking of mice and cats puts me in mind of “A little fable” by Franz Kafka.

“Alas”, said the mouse, “the whole world is growing smaller every day. At the beginning it was so big that I was afraid, I kept running and running, and I was glad when I saw walls far away to the right and left, but these long walls have narrowed so quickly that I am in the last chamber already, and there in the corner stands the trap I am running into.” “You only need to change your direction”, said the cat, and ate it up.

As in many of Kafka’s stories the theme is the misjudgement of reality and failure. Are researchers in Spain staring paralyzed at a trap or should they be confidently avoiding becoming caught between the walls?

The cat may know the answer.

ls centres de recerca del Parc tenen la capacitat d’atraure el millor talent científic per l’excel·lència de les seves investigacions, però també pel fet de compartir un espai únic com és l’edifici PRBB. La seva ubicació i arquitectura el converteixen en un edifici emblemàtic de la ciutat, que atrau fins i tot realitzadors a la recerca d’escenaris visualment atractius per rodar pel·lícules.

Així doncs, el PRBB ha estat escollit com a localització de diverses produccions cinematogràfiques. Algunes s’han quedat com a mer projecte, com és el cas de l’última pel·lícula de Sean Penn, The Gunman (2015). D’altres s’han fet realitat, i el PRBB apareix, per exemple, com part del

paisatge apocalíptic que José Coronado y Quim Gutiérrez veuen a Los últimos días (2013). També ha estat un hospital futurista a Circuit (2010), amb Nacho Duato, i una clínica de tractaments de fertilització in vitro a Manuale d’amore 2 (2006).

L’última pel·lícula rodada al PRBB i estrenada recentment als cinemes ha estat Rastres de sàn· dal (2014), de la directora Maria Ripoll, on la protagonista interpreta una investigadora en cèl· lules mare que treballa al CMRB. També s’està a punt d’estrenar l’últim llargmetratge en 3D del director Manuel Huerga, Barcelo· na, rosa de foc (2014), on el PRBB apareix com a espai referent de la recerca biomèdica amb imatges espectaculars. L’atractiu de l’edifici PRBB es fa palès fins i tot a la gran pantalla.

T

he Park’s research centres have the capacity to attract top scientific talent thanks to the excellence of its research, but also for the opportunity of sharing the unique PRBB building. Its location and architecture make it one of the city’s iconic buildings, and it even attracts filmmakers in search of visually appearing settings for films.

So the PRBB has been chosen as a shooting location for se veral cinematographic productions. Some have remained on the drawing board, like for Sean Penn’s latest film, The Gunman (2015). Others have become reality, and the PRBB appears, for example, as part of the apocalyptic landscape that José Coronado and Quim Gutiérrez

see in Los últimos días (The Last Days, 2013). It was also a futuristic hospital in Circuit (2010) with Nacho Duato, and an in vitro fertility clinic in Manuale d’amo· re 2 (Manual of Love 2, 2006).

The latest film shot at the PRBB, which recently premièred in cinemas, is Traces of sandalwood (2014) by director Maria Ripoll, where the main character is a stem cell researcher who works at the CMRB. Also about to be released is the latest 3D feature film directed by Manuel Huerga, Bar· celona, rosa de foc (Barcelona, fire rose; 2014) where the PRBB appears as a benchmark biological research centre, with spectacular images. DESTACAT / HIGHLIGHT Setmana de la Ciència Els centres del PRBB van participar activament a la 19a Setmana de la Ciència del 14 al 23 de novembre. L’IBE (CSIC-UPF) i la plataforma La Ciència al Teu Món van organitzar una competició per descobrir els camins dels humans des del seu origen comú a l’Àfrica. El CREAL es va instal·lar a la plaça de la Universitat per explicar els darrers resultats dels estudis de contaminació i també va rebre una visita d’escolars i una altra de 20

professors d’Europa interessats a millorar l’educació mediambiental dels estudiants. L’IMIM va rebre la visita d’alumnes de batxillerat de dos instituts. El CRG va acollir 129 estudiants de secundària, i 200 estudiants més van fer un taller a les seves escoles gràcies al kit d’electroforesi del CRG. A més, dos investigadors del centre van anar a escoles a explicar la vida d’un científic. En total, unes 150.000 persones, entre elles 14.000 escolars, van participar en aquesta iniciativa a Catalunya.

Science week 2014 The PRBB centres played an active role in the 19th Science Week held from 14-23 November. The IBE (CSIC-UPF) and the platform “La ciència al teu món” (Science in your world) organised a competition to discover the paths followed by humans since their common origin in Africa. The CREAL set up in Plaça Universitat to explain the latest pollution study results, and also received one visit from a school and another of 20 teachers from

around Europe interested in improving students’ environmental education. The IMIM was visited by A-level students from two institutes. The CRG hosted 129 secondary school students, and a further 200 did a workshop in their own classrooms thanks to the CRG’s electrophoresis kit.

On top of this, two researchers from the centre went to schools to explain the life of a scientist.

Estadística, onades de calor i salut Analysing the prevention messages

avier Basagaña va arribar al CREAL el 2008, just després de fer un doctorat en bioestadística a la Universitat de Harvard. Des d’aleshores el seu principal interès ha estat estudiar els efectes en la salut de temperatures extremes.

Quan hi ha una onada de calor —dos o tres dies seguits a una temperatura supe­ rior a un cert llindar, 30 ºC en el cas de Barcelona— el nombre d’hospitalitzacions i la mortalitat augmenten, sobretot entre la gent gran i els malalts crònics. «L’onada de calor del 2003 va causar 8.000 morts més del que és normal a Espanya i 70.000 a tot Europa», explica Basagaña. Segons les projeccions, i tenint en compte el canvi climàtic, s’esperen onades de calor més sovint i més duradores. «A finals de segle podríem tenir una onada com la de 2003 cada quatre anys», pronostica el científic. Per això el seu grup està fent prediccions sobre què passarà quan hi hagi més onades de calor. Tenint en compte l’envelliment de la població, preveuen que el 2050 a Catalunya hi podria haver 10 vegades més morts relacionades amb la calor que la mitjana dels últims 20 anys. Això, però, no té en compte una possible adaptació fisiològica a un temps més calorós, que evitaria alguns dels efectes.

«De fet, cada estiu ens adaptem a la calor: per exemple, comencem a suar abans i la composició de la nostra suor canvia», explica Basagaña. A més, també hi ha l’adaptació tecnològica, amb aires condicionats, i de comportament, com hidratar-se bé o no fer esport en les hores més caloroses.

Analitzant els missatges preventius De fet, a partir del 2003 es van posar en marxa plans de prevenció amb aquests tipus de missatges i el grup n’està analitzant l’efectivitat. Aquests plans inclouen tant campanyes informatives com una major coordinació entre hospitals, residències de gent gran, protecció civil i el servei meteorològic. Avui dia es pot predir l’arribada d’una onada de calor a una setmana vista i activar els plans d’emergència. El grup de Basagaña estudia si el missatge arriba a tota la població amb la mateixa efectivitat i han fet un mapa de vulnerabilitat que mostra

que, dins de Barcelona, el risc de tenir efectes negatius en la salut a causa de la calor és el doble en algunes àrees que en d’altres.

Estadística, una ciència creativa El grup dirigit per Basagaña també es dedica a desenvolupar nous mètodes estadístics. En particular, l’interessa veure com afecta la informació que no tenim als resultats per tal de poder millorar la precisió de les inferències que es fan en els estudis. Per fer això, reanalitza estudis observacionals afegint errors aleatoris o eliminant algunes de les dades recollides, a fi de veure com això afecta els resultats obtinguts. Així doncs, han quantificat que els estudis epidemiològics que utilitzen mapes de contaminació basats en poques mesures poden infraestimar els efectes en la salut en un 50%.

La inferència és essencial en els estudis epidemiològics. Per exemple, per saber a quina contaminació estem exposats es fan servir uns monitors que els voluntaris duen a sobre, però «no els poden dur tot el dia. A més, els aparells mesuren la contaminació ambient, però no sabem quina proporció d’aquesta contaminació el voluntari ha inhalat realment... Sempre hi ha un cert grau d’incertesa», explica l’investigador.

L’estadística és una ciència molt oberta; hi ha molt espai per a creativitat, diu Basagaña. I ho explica: «Quan reps les dades d’un estudi, hi ha moltes característiques que cal tenir en compte a l’hora d’analitzar-les: segons com s’hagi dissenyat l’estudi, si falten algunes mesures, etc., t’has d’adaptar a cada situació, modificar lleugerament els tests que fas servir».

Since then, his main interest has been the health effects of extreme temperatures.

When there is a heat wave — two or three consecutive days with a top temperature above a certain threshold, 30ºC in the case of Barcelona — the number of hospitalisations and deaths increase, especially amongst old people and the chronically ill. “The heat wave in 2003 resulted in 8,000 more deaths than usual in Spain, and 70,000 across Europe”, explains Basagaña.

According to projections, and taking climate change into account, more frequent and longer heat waves are expected. “At the end of the century we will have a heat wave like that in 2003 every four years”, predicts the scientist. For this reason his group is ma­king forecasts about what will happen when there are more heat waves. Considering the ageing population, it is foreseen that in Catalonia, the year 2050 could see 10 times more heat-related deaths than the average for the last 20 years. This, however, does not take into account possible physio­logical adaptations to a hotter period, which would cancel out some of the effects. “In fact, each summer we adapt to the heat, for example, we begin to perspire earlier and the composition of our sweat changes”, explains Basagaña. As well as this there are technological adaptations, like air-conditioning, and behavioural changes, such as good hydration and not doing sport at the hottest times.

In fact, since 2003 prevention plans with these types of messages have been launched by the government, and the group is analysing their effectiveness. The plans include both informative campaigns and greater coordination between hospitals, old folks’ homes, civil defence and the meteorological service. Nowadays a heat wave can be predicted a week before it arrives, and emergency plans can be put into operation. Basagaña’s group looks at whether the message reaches the entire population with the same effectiveness, and they have made a vulnerability map showing that, in Barcelona, the risk of negative health effects due to heat is twice as high in some areas compared to others.

Statistics, a creative science Basagaña and his group also devote their time to developing new statistical me­thods. In particular, it interests them to see how missing information affects results, in order to improve the accuracy of the inferences made in the studies. To do this, they re-analyse observational studies adding random errors, or eliminating some of the data collected, to see how this alters the results obtained. In this way, they have found that epidemiological studies which use pollution maps based on few measurements can underestimate health effects by 50%.

Inference is crucial to epidemiological studies. For example, to find out which pol­lutants we are exposed to, monitors are given to volunteers to carry around, but “they do not have them with them all day, and on top of this the devices measure the environmental pollution, but we do not know what proportion of this the volunteer has really inhaled... There is always a degree of uncertainty”, explains the researcher.

Statistics is a very open science, there is a lot room for creativity, says Basagaña.

And he explains: “When you receive study data there are many characteristics to take into account in the analysis: according to how the study was designed, if some measu­rements are missing, and so on. You have to adapt to each situation, slightly modifying the tests that you use”

Analitzen la producció biomèdica d’alt impacte a Espanya Núria Pérez

n estudi bibliomètric publicat via web (http://www.fundaciorecerca.cat/ topcited) i dirigit per Jordi Camí, catedràtic de Farmacologia de la UPF i director del PRBB, permet explorar els articles biomèdics de científics residents a Espanya que han rebut més notorietat entre el 1997 i el 2011.

Els autors, entre els quals hi ha Raül Méndez i Eduard Suñén, de la Fundació Catala-

na per a la Recerca i la Innovació (FCRI), i Joan Pons, de l’AQuAS, han analitzat els prop de 190.000 articles biomèdics publicats durant aquests 15 anys signats per almenys un autor d’Espanya. D’aquests articles, 2.514 (1,3%) formaven part de l’1% dels treballs més citats a tot el món entre els del mateix any de publicació i disciplina.

Un 46,8% dels més citats són d’autors que treballen a Catalunya i un 27,8% a Madrid.

L’estudi confirma, doncs, el lideratge de Catalunya en recerca biomèdica, així com el gran pes que encara té la recerca clínica.

Analysing high impact biomedical production in Spain

bibliometric study published on the web (http://www.fundaciorecerca. cat/topcited) and directed by Jordi Camí, a pharmacology lecturer at the UPF and director of the PRBB, explores the most renowned biomedical articles from 1997 to 2011 by scientists resident in Spain.

The authors, who include Raül Méndez and Eduard Suñén of the Catalan Founda-

tion for Research and Innovation (FCRI), and Joan Pons from AQuAS, have analysed the almost 190,000 biomedical articles published over this fifteen year period by authors in Spain. Of these, 2,514 (1.3%) were amongst the 1% most cited works in the world from the same publication year and discipline. 46.8% of these most cited papers are by authors working in Catalonia, with 27.8% based in Madrid. The study therefore confirms Catalonia’s leadership in biomedical research, as well as the continued importance of clinical research

n consorci internacional que inclou investigadors del CRG liderats per Roderic Guigó ha presentat una comparació exhaustiva dels elements fun­ cionals del genoma del ratolí amb els de l’humà. Els resultats donen pistes de per què certs processos, com el sistema immunitari, el metabolisme i la resposta a l’estrès, són tan diferents entre els ratolins i els humans.

Les dades permetran una comprensió millor de la biologia i l’evolució dels mamífers, i oferiran nova informació sobre l’ús de ratolins com a models animals de malalties humanes.

Comparant els processos que regulen l’activitat del genoma en diferents teixits d’humans i ratolins, els científics han trobat que, en general, els sistemes en les dues espècies són molt semblants i s’han conservat a través del temps, validant la utilitat d’aquest model animal. by Roderic Guigó has presented an exhaustive description of the mouse’s functional genome elements and their comparison with the human genome. The results shed light on why certain processes in mice, like the immune system, metabolism and stress response, are so different to those of humans. The data will enable a better understanding of mammalian biology and evolution, as well as contribute

new information on the use of mice as animal models for human diseases.

By comparing the processes regulating genome activity in different human and mice tissues, the scientists have found that, in general, the systems in the two species are very alike, and have been preserved through time, validating the usefulness of this animal model. However, they have also detected certain differences in the DNA and certain gene expression patterns, explains Alessandra Breschi, one of the first co-authors of the main work and a PhD student in Guigó’s lab.

“This study is part of ENCODE, a living project that generates constantly updated and improved maps. We hope that the project can keep providing this data, making it available to everyone and treating it in a systematic way”, concludes Guigó, the only principal investigator in Europe involved in this work. The results have been publi­shed in four articles in Nature, as well as in other scientific journals including Genome Biology, Genome Research, PNAS, Blood and Nature Communications

n estudi internacional, en el qual han participat investigadors de l’IMIM, ha identificat 15 muta­ cions en el gen NPC1L1 associades a una reducció dels nivells de colesterol LDL o «colesterol dolent». A més, la presència d’aquestes mutacions es va associar a una reducció d’un 50% del risc de patir un infart de miocardi.

La proteïna NPC1L1 s’encarrega d’absorbir, a l’intestí, el colesterol dels aliments. Una reducció d’aquesta absorció provoca una disminució del colesterol LDL en sang. En l’estudi publicat a la revista The New England Journal of Medi· cine es va analitzar aquest gen en unes 21.000 persones (7.000 de les quals havien patit un infart) i es van identificar les 15 mutacions esmentades. Posteriorment, es va analitzar la seva presència en 90.000

persones més i es va establir la freqüència d’aquestes mutacions en una de cada 650 persones.

Actualment hi ha un fàrmac per reduir els nivells de colesterol, l’ezetimiba, que disminueix l’activitat de l’NPC1L1, però fins ara cap estudi no ha demostrat que redueixi el risc de patir un infart. «Una de les diferències entre el tractament i la mutació és que les mutacions exerceixen la seva acció des del naixement, mentre que el fàrmac s’utilitza només en cas de necessitat», comenta Roberto Elosúa, investigador a l’IMIM i un dels autors de l’estudi.

Genetic mutations found that reduce heart attack risk by 50%

A

n international study in which researchers from the IMIM have taken part has identified 15 mutations in the NPC1L1 gene associated to

reduced levels of LDL, or “bad”, cholesterol. As well as this, the presence of these mutations is associated with the risk of suffering myocardial infarction being reduced by 50%.

The protein NPC1L1 is responsible for absorbing, in the intestines, cholesterol from food. A reduction in this absorption leads to decreased LDL cholesterol in the blood. The study published in The New England Journal of Medicine looked at this gene in some 21,000 people (7,000 of which had suffered a heart attack) and identified the 15 mutations mentioned.

Subsequently, the presence of this gene was analysed in a further 90,000 people and the frequency of the mutation was established as one person in every 650.

There is currently a drug for reducing cholesterol levels, ezetimibe, that diminishes NPC1L1 activity, but until now no study had shown that it reduced heart attack risk. “One of the differences bet-

uasi 40 anys després que la majoria de països prohibissin la producció i l’ús de compostos organoclorats (COs), encara es detecten restes d’aquests compostos químics tòxics en l’organisme dels joves. Així ho conclou un estudi realitzat per investigadors del CREAL, centre de recerca ISGlobal, en col·laboració amb l’Institut de Diagnosi Ambiental i Estudis de l’Aigua del CSIC. L’estudi va analitzar uns 100 nens de Ribera d’Ebre i 480 de Menorca, tots part d’INMA, un projecte estatal que pretén avaluar l’impacte de l’entorn en els infants. Es van obtenir mostres de sang en

el moment de néixer, als 4 i als 14 anys i s’hi van mesurar diversos COs. L’estudi demostra que les càrregues totals de DDE (subproducte del pesticida DDT) i PCB en sèrum són superiors als 14 anys que en néixer. Això és degut sobretot a la dieta, la font principal d’exposició a aquests compostos. L’estudi també mostra nivells superiors de COs en els nens alletats durant els primers mesos, tot i que els autors recorden que la llet materna proporciona molts més beneficis que els danys potencials d’aquestes substàncies.

Mireia Gascón, autora principal de l’estudi, alerta de la situació en països en vies de desenvolupament on encara s’utilitza el DDT com a insecticida per controlar el mosquit transmissor de la malària.

Years after being banned, organochlorine compounds persist in the body

A

lmost 40 years after the majority of countries banned the production and use of organochlorine compounds (OCs), remnants of these toxic chemical compounds can still be detected in the bodies of young people. This is the conclusion of a study carried out by researchers at the CREAL, an ISGlobal research centre, in collaboration with the CSIC’s Institute of Environmental Asses­ sment and Water Research.

The study analysed 100 children from Ribera d’Ebre and 480 from Menorca, all part of the INMA Project, a state project

that aims to assess the impact of the environment on children. They obtained blood samples at birth, ages 4 and 14, and measu­ red various OCs. The study shows that the total DDE (a by-product of the pesticide DDT) and PCB loads in serum are higher at 14 years of age than at birth. This is principally due to diet, the main source of exposure to these compounds. The study also shows higher levels of OCs in children who were breastfed in the first months, even though the authors point out that the benefits of a mother’s milk outweigh any potential harm from these substances.

Conference Programme financed by the CRG and the PRBB AIDA ANDRÉS, Friday December 19. Andrés, from the Department of Evolutionary Genetics at the Max Planck Institute for Evolutionar y Anthropology in Leipzig, Germany, aims to understand the influence that natural selection has played in the evolution of humans and other primates. In particular she is interested in how adaptation has shaped the evolution of relevant phenotypes, how natural selection has influenced the acquisition of species-specific traits, what selective forces are responsible for phenotypic diversity within populations and how past selection affects our present-day phenotypes. She has been invited by Jaume Bertranpetit (UPF).

GREGORY WELLENIUS, Monday January 12. Wellenius, from the Department of Epidemiology at Brown University in Rhode Island, US, is interested in studying environmental determinants of cardiovascular disease. His work has primarily focused on studying the effects of ambient air pollution on the risk of cardiovascular events and its effects on cardiovascular phy­ siology. He uses tools from the fields of epidemiology and toxicology to evaluate the association between environmental exposures and disease, identify subgroups of the population that may be particularly susceptible, and elucidate the physiologic mechanisms for the observed effects. He has been invited by Mark Nieuwenhuijsen (CREAL).

Steitz, from the Molecular Biophysics and Biochemistry & HHMI Boyer Center for Molecular Medicine at Yale University School of Medicine, New Haven, US, is studying noncoding RNA-protein complexes (ncRNPs), which are ubiquitous in eukaryotic cells and inhabit specific cellular compartments.

Her lab discovered the most famous noncoding nuclear RNPs (snRNPs), which participate in pre-mRNA splicing by recognizing important intron signals and assembling to form an active splicing complex called a spliceosome. She is also trying to decipher the roles of viral noncoding RNPs, and is also interested in translational control and miRNAs. She has been invited by Juan Valcárcel and Fátima Gebauer (CRG).

LAURE BALLY-CUIF, Monday February 9. Bally-Cuif, from the CNRS Research Unit “Neurobiology & Development” and the Institut de Neurobiologie Alfred Fessard in Gifsur-Yvette Cedex, France, focuses on zebrafish neurogenetics. Her goal is to identify some of the mechanisms involved in controlling neurogenesis and neuronal specification in the embryonic and adult brain, and their influence on behaviour. She uses the model system zebrafish (Danio rerio), which offers unprecedented advantages for manipula­ ting gene function both during development and in adulthood, where adult neural stem cells are abundant, and which is also amenable to behavioral assays.

«La diversitat de temes del simposi va ser clau pel seu èxit» Maruxa Martínez-Campos D’on provenien els assistents? a majoria dels participants provenien del CRG i d’altres centres del PRBB, així com de molts instituts de Barcelona. Però també vam tenir gent de Singapur, Alemanya, França, Itàlia...

L

Com es va organitzar el simposi?

Cada any organitza el simposi un programa diferent del CRG. Vam començar a organitzar el d’enguany fa un any i mig, quan cada cap de grup va convidar el seu «ponent perfecte», el científic que els encantaria tenir a la vora. La majoria dels convocats va acceptar perquè ells també volien reunir-se amb la diversitat d’experts mundials aplegats.

Com va ser el format de la conferència?

A part de les xerrades dels setze oradors convidats, i d’altres d’uns 10 minuts realitzades per científics joves seleccionats d’entre els estudiants de doctorat i els postdocs que van presentar abstracts, vam experimentar amb la «poster speed presentation», que va ser un gran èxit. Vam fer tres sessions d’uns 20 minuts en què tothom que havia presentat un pòster —al voltant de 60— va tenir exactament un minut per explicar-lo, acompanyat d’una sola diapositiva. Òbviament no podien dir gaire, però la idea era enviar el missatge principal i que la gent interessada anés més tard a veure el pòster, a discutir-lo amb més detall amb l’autor, etc.

Aquestes sessions van ser molt dinàmiques, gràcies al fet que vam ser molt estrictes amb el temps! Però va funcionar i la gent va participar molt més del que és habitual en les posteriors sessions de pòsters.

Quins van ser els temes més destacats?

Un dels èxits del simposi va ser la seva diversitat. Vam estar exposats a ciència de primera qualitat i a idees sorprenents, veritablement inspiradores. Els temes van variar des d’una vista detallada de l’organització estructural dels complexos de transcripció fins a una visió general dels canvis en l’organització tridimensional d’un cromosoma sencer durant el procés d’inactivació del cromosoma X; des de les funcions dels nous RNA circulars fins als rols de les secrecions de cèl·lules senescents en la promoció de la metàstasi del càncer; de la conversió directa d’hepatòcits en epitelis del conducte biliar o de fibroblasts en neurones, a una visió de la lògica genètica sobre com el control transcripcional de llarg abast dels gens Hox determina la formació de les extremitats.

Cap a on van ara aquests camps?

Una tendència general comuna és l’anàlisi de cèl·lules individuals, tant dels seus comportaments com dels seus components.

Les tecnologies ja estan preparades per fer això i els resultats revelen que l’anàlisi de les poblacions sovint amaga la diversitat de les cèl·lules individuals, que és necessària per explicar les propietats fonamentals dels sistemes biològics. Una altra idea clau és la comprensió que els circuits genètics que determinen la pluripotència cel·lular,

PERFIL / PROFILE El 13è Simposi Anual del CRG va tenir lloc el 6 i el 7 de novembre a l’Auditori del PRBB. Coorganitzat pels 11 caps de grup del programa «Regulació gènica, cèl·lules mare i càncer» del CRG, així com per Salvador Aznar-Benitah, exmembre del programa, el simposi va reunir més de 250 científics per discutir el progrés i els reptes de futur en aquestes àrees diverses però relacionades. Juan Valcárcel, coordinador del programa, ens parla de com va anar.

The 13th CRG Annual Symposium took place on the 6th and 7th of November in the PRBB Auditorium. Co-organised by the eleven group leaders of the “Gene regulation, stem cells and cancer” programme at the CRG, as well as former group leader from the programme Salvador Aznar-Benitah, the meeting brought together more than 250 scientists to discuss progress and future challenges at the cutting edge of these diverse but related fields.

Juan Valcárcel, coordinator of the programme, tells us how it went.

“The diversity of subjects at the symposium was key to its success” Where were the attendees from?

M

ost of the participants were from the CRG and other PRBB centres, as well as various institutes in Barcelona. But people also came from places like Singapore, Germany, France, and Italy.

How was the meeting organised?

Each year a different CRG programme organises the symposium. We started organi­ sing this year’s meeting a year and a half ago, when each PI sent out an invitation to their “dream speaker”. Most of the guest speakers accepted because they, too, wanted to meet up with other experts from diverse fields.

You had a “Speed poster presentation” – what is that?

Apart from the main talks by the 16 guest speakers, and shorter 10-minute talks by younger scientists selected from amongst the PhD students and postdocs who submitted abstracts, we experimented with the “Speed poster presentation”, which was a huge success. We had three sessions of around 20 minutes each in which everyone who had submitted a poster — about 60 people — had exactly one minute to explain it with the help of a single slide. Obviously they could not say much, but the idea was to put across their main message and

get people interested, so later they would go and see the poster and discuss it in more depth with the author. The sessions were very dynamic, we were very strict with the timings and it worked well: the poster sessions were much better attended than usual.

What were the highlights?

One of the successes of the me­eting was its diversity. We were treated to top quality science and amazing, truly ins­piring insights. These ranged from a detailed view of the structural organisation of transcription complexes to an overview of the changes in the three-dimensional organisation of an entire chromosome during X chromosome inactivation; from the roles of novel circular RNAs to the part senescent cell secretions play in promoting cancer metastasis; from the direct conversion of hepatocytes into bile duct epithelium and fibroblasts into neurones, to a glimpse into the genetic logic of how long-range transcriptional control of Hox genes determines limb bud formation.

How are these fields moving forwards?

A general trend is the analysis of single cells, both of their individual behaviour and of their components. Technologies are now ripe for this and the results reveal that analysing populations does not reveal the diversity of individual cells, which is ne­ cessary for explaining the key properties of biological systems. Another important rea­ lisation is that genetic circuits determining cell pluripotency, differentiation, transformation and senescence are interconnected in networks of epigenetic, transcriptional and post-transcriptional gene regulation

El crom de l’aigua i els nivells en sang i cabells

U

n estudi epidemiològic realitzat a Grècia en el qual han participat investigadors del CREAL, centre de recerca de l’aliança ISGlobal, ha investigat els efectes sobre la salut associats amb l’exposició a llarg termini a crom ingerit a través de l’aigua. Mitjançant una entrevista personal es va recollir informació sociodemogràfica, sobre l’estil de vida, consum d’aigua potable, hàbits dietètics i l’historial professional i mèdic de 304 participants.

Els investigadors van mesurar les concentracions de crom en sang i en el cabell dels subjectes, així com diversos paràmetres bioquímics, hematològics i inflamatoris en sang. També els van fer uns tests motors.

La presència de crom en l’aigua potable consumida pels participants va variar en-

tre 0,5 i 90 micrograms per litre —el límit establert per l’OMS és de 50. Els investigadors van trobar que els nivells de crom en la sang i als cabells, així com certs paràmetres hematològics i bioquímics, es correlacionaven amb l’exposició al crom, calculada a partir dels nivells en aigua i la ingestió individual. Tot i així, els nivells detectats són considerats dins la normalitat, segons la literatura científica. «A més, hem comprovat que el deteriorament motor no s’associava amb l’exposició a crom», explica Cristina Villanueva, del CREAL.

ce research centre, has investigated the health outcomes associated with longterm exposure to chromium via drinking water. Socio-demographics, lifestyle, drinking water intake, and dietary habits, as well as occupational and medical history data from 304 participants were recorded in personal interviews. The researchers

then measured the total chromium concentration in the blood and hair of the study subjects, and looked at certain haematological, biochemical and inflammatory parameters in their blood. A motor test was also car­ried out on the individuals.

The subjects were exposed to chromium in drinking water ranging from <0.5 to 90μg/l — where 50μg/l is the maximum recommended by the WHO. The researchers found that chromium levels in blood and hair, as well as some of the haematological and biochemical parameters, were associated to lifetime chromium exposure via drinking water, calculated from the concentrations in the water and individual ingestion ascertained though the questionnaires. Nevertheless, the levels detected were within “normal range” according to the literature. “Also, no motor impairment was associated with exposure to chromium”, says Cristina Villanueva at the CREAL

El «hikikomori» també és un problema a Europa Rosa Manaut

etges de l’Hospital del Mar han publicat a la revista Internatio· nal Journal of Social Psychiatry el primer estudi europeu sobre la síndrome de hikikomori o aïllament social, que inicialment es creia vinculada únicament a la cultura japonesa.

Les persones amb hikikomori es re­ clouen a casa evitant qualsevol compromís social. Fins ara a Espanya només se n’havien reportat casos puntuals. «La creació a Barcelona d’un servei d’atenció domiciliària per a persones amb trastorns mentals greus ha tret a la llum la veritable dimensió del problema», explica el responsable de l’estudi, Víctor Pérez-Solà, director de l’Institut de Neuropsiquiatria i Addic­

cions de l’Hospital del Mar i investigador a l’IMIM. Un 73,8% dels 164 casos detectats a Espanya eren homes de 36 anys de mitjana, mentre que la mitjana d’edat de les dones era de 51. La majoria vivien amb la família i la meitat tenien estudis superiors. El pe­­­­ río­de mitjà d’aïllament era de 40 mesos i el màxim era de 30 anys. «El hikikomori sovint va acompanyat de trastorns psicòtics (35%), ansietat (22%) o trastorns afectius (75%)», afegeix Pérez-Solà. Gairebé un 60% dels casos tenia historial de hikikomori a la família.

«Cal oferir psicoeducació familiar per facilitar l’acceptació del diagnòstic i el tractament del hikikomori, per poder accedir als pacients i aconseguir la seva col·laboració.

Així mateix, ha d’haver-hi una xarxa terapèutica per garantir la continuïtat del tractament», conclou Ángeles Malagón-Amor, primera signant de l’estudi.

Hikikomori syndrome is also a problem in Europe

n the International Journal of Social Psychiatry, doctors from the Hospital del Mar have published the first European study on the hikikomori syndrome or social isolation, which was initially believed to be linked only to Japanese culture.

People with hikikomori shut themselves away at home and avoid any social commitments. Up to now only a few isolated cases had been reported in Spain. “The creation of a home care service in Barcelona for people with serious mental disorders has shed light on the true size of the problem” explains the scientist who led the study, Víctor Pérez-Solà, director of the Ins­ titute of Neuropsychiatry and Addictions

at the Hospital del Mar and a researcher at the IMIM. 73.8% of the 164 cases detected in Spain were young men with an average age of 36, while women had an average of 51. The majority of cases lived with their family and half were university educated. The average period of isolation was 40 months, up to a maximum of 30 years. “Hikikomori is often accompanied by psychotic disorders (35%), anxiety (22%) or affective disorders (75%)”, adds Pérez-Solà. Almost 60% of the cases had a history of hikikomori in the family.

“It is necessary to offer family counselling to help people accept the diagnosis and treatment of hikikomori, to gain access to the patients and get their collaboration.

There must also be a therapeutic network to guarantee the continuity of the treatment” concludes Ángeles Malagón-Amor, first author of the study

El genoma felí revela variants genètiques exclusives del gat domèstic animals territorials que han de reconèixer vastes extensions de terreny i sorprendre les preses, fins i tot en la foscor.

ls gats salvatges del paleolític van evolucionar cap als actuals gats domèstics gràcies a canvis genètics que els van fer més dòcils per obtenir aliment, dotats de molt bona memòria i més espantadissos. Aquestes són algunes de les conclusions de l’anàlisi comparativa publicada a la revista PNAS sobre els gats domèstics. Liderat per científics nordamericans i amb participació del laboratori de Genòmica Comparada que coordina Tomàs Marqués-Bonet a l’IBE, l’estudi ha comparat el genoma del gat domèstic (Fe· lis silvestris catus), d’uns 20.000 gens, amb el del gat salvatge, el tigre, el gos, la vaca i l’humà, i ha identificat 281 variants genètiques exclusives del gat domèstic.

«S’han seleccionat evolutivament gens que tenen a veure amb els mecanismes de recompensa», ha manifestat Marqués-

Bonet. «Això s’explica perquè els gats han après a fer bondat a canvi d’obtenir aliment». A més, els investigadors han trobat en el seu genoma evidència de la bona memòria i de l’excel·lent oïda i visió dels gats.

The feline genome reveals exclusive genetic variants in the domestic cat

alaeolithic wildcats evolved into our current domestic cats thanks to genetic changes that led them to be tamer when getting food, gave them a very good memory and made them more skittish. These are some of the conclusions of the comparative analysis of domestic cats published in the journal PNAS. Led by North American scientists and involving the Comparative Genomics lab coordina­

ted by Tomàs Marqués-Bonet at the IBE in Barcelona, the study compared the genome of the domestic cat (Felis silvestris catus), with around 20,000 genes, to that of the wildcat, tiger, dog, cow and human, identifying 281 genetic variants exclusive to the domestic cat.

“There has been evolutionary selection of genes related to reward mechanisms”, states Marqués-Bonet. “This is because cats have learnt to exchange good beha­viour for food.” Additionally, the resear­ chers have found evidence of good memory in the cat genome, as well as of excellent hearing and vision. This is in line with the fact that felines are territorial animals that have to recognise rough expanses of terrain and surprise prey, even in the darkness.

Encara que és lluny de ser el model més utilitzat, avui en dia el pollastre és crucial per estudiar el desen­volupament de les extremitats, el sistema nerviós, els òrgans sensorials i, fins i tot, malalties. uè va ser primer: l’ou o la gallina?

Aquesta pregunta és tan antiga com l’ús d’ous en la nostra dieta. Però els ous de gallina també es poden utilitzar en la ciència. Les grans dimensions de l’embrió de pollastre van permetre als científics de temps antics (350 aC) fer observacions significatives sense l’ajuda d’un microscopi.

Aristòtil va obrir ous de gallina en diferents moments d’incubació, va observar com l’organisme canviava amb el temps i va reconèixer similituds entre el desenvolupament humà i el de pollastre. Al segle xvi els científics europeus utilitzaren l’embrió de pollet per demostrar la diferenciació de teixits, desmentint la creença generalitzada en aquell moment que els organismes estan «preformats» en la seva versió adulta i que durant el desenvolupament tan sols creixen en mida. Més tard, l’ús del microscopi va permetre als científics observar directament l’òvul al llarg del seu desen­ volupament, convertint el posllastre en un important sistema model per a la biologia

hat came first, the chicken or the egg? This question is as old as the use of eggs for food.

However, hen eggs can be also used in science. The large size of the chick embryo allowed ancient scientists (350 BCE) to make significant observations without the need for a microscope. Aristotle opened up chicken eggs at various stages of incubation, noted how the organism changed over time and recognised significant similarities between human and chicken development. In the 16th century European

scientists used chicks to demonstrate tissue differentiation, disproving the widely held belief at the time that organisms are “preformed” in their adult version and merely grow larger during development.

Later on, microscopes enabled scientists to look directly into the egg during its development, making the chicken an important model system for developmental biology. An example of this is the discovery of the three embryonic germ layers that give rise to all tissues in adults: ectoderm, mesoderm and endoderm. The adult chicken has also made significant contributions to immunology, virology and cancer research, including the work that led Nobel laureate Peyton Rous to discover that viral infection could induce sarcoma.

Recently, the publication of the chicken genome revealed that it shares 65% of its genes with humans, enabling the expansion of transgenic techniques. Although far from being the most utilised model system nowadays, the hen is crucial for studying the development of limbs, the neural system, sensory organs and disease. Long live the chicken!

n cop passades les festes de Nadal, què fer amb els turrons sobrants que s’acumulen a les cases? Us proposo un flam!

Ingredients: 150 g de torró de Xixona, 500 ml de llet, 5 ous grans, 4 cullerades soperes de sucre, caramel líquid.

Preescalfar el forn. Triturar amb la batedora tots els ingredients (excepte el caramel líquid). Abocar-ho dins un motlle caramel·litzat i coure-ho al bany maria (posar el motllo dins un recipient una mica més gran amb aigua) al forn a 190 oC durant una hora. Per saber si ja està cuit, podeu punxarlo amb un bastonet dels que s’utilitza per als pinxos, si surt net ja està, si no deixar coure uns minuts més. Deixar refredar i desemmotllar.

Ingredients: 150 g of Turró Xixona (or similar), 500ml milk, 5 large eggs, 4 tablespoons sugar, liquid caramel.

Preheat the oven. Blend all the ingredients together (except the liquid caramel). Cover the bottom and sides of a mould with the caramel and pour in the mixture. Heat this in a bain-marie (by putting the mould in a slightly larger container filled with water) in the oven at 190oC for one hour. To test whether it is cooked, prick it with a skewer: if it comes out clean, it is done, if not, cook it for a few more minutes. Let it cool down and turn it out of the mould.

Note: If you are brave enough to make the liquid caramel yourself, here’s how: put 100 g of sugar in a saucepan with 3 tablespoons of water and heat it gently over a low heat stirring with a wooden spoon until the sugar becomes a runny golden-coloured paste.

Es busca: recerca reproduïble El juny passat, líders científics es va reunir amb representants de revistes científiques i dels organismes de finançament per abordar solucions a la manca de reproductibilitat dels resultats de la recerca biomèdica, un problema creixent. La reunió, centrada en el paper de les revistes, va acabar amb suggeriments com ara tenir polítiques específiques per a l’anàlisi estadística, o recomanar als autors que dipositin les dades a dipòsits públics. Serà suficient per garantir una major transparència i reproductibilitat dels resultats de la investigació? El·lipse ho ha demanat a alguns investigadors dels centres del PRBB. In search of reproducible research This past June, scientific leaders met with representatives from journals and funding agencies to address their concerns about a lack of reproducibility of biomedical research results. The meeting focused on the role of journals, and suggestions ranged from having specific policies for statistical analysis to recommending housing data in public repositories. Will this be enough to ensure the greatest transparency and reproducibility of research results? El·lipse has asked some researchers at the PRBB centres.

“Rigorous peer-review by expert in the field, including a forensic evaluation of the raw data, is the best way to ensure and reproducibility of research. Revision of numerical data and analyses by expert statisticians is a crucial step especially in clinical studies, where results may affect medical practice. On the other hand, scientific misconduct could be prevented by giving young scientists training on ethical issues. Specific software to detect fake images and plagiarism also helps.”

D’altra banda, tant editors com revisors han de fomentar que els autors proporcionin tanta informació com sigui possible sobre els procediments, els controls i totes les anàlisis addi­cionals, incloent-hi resultats contradictoris, ja que aquests són clau per a la transparència i amb el temps poden ser significatius per a la reproductibilitat. Personalment, crec que fer públics els comentaris dels revisors, juntament amb les respostes dels autors, és un bon pas.»

“Major changes improving reproducibility and transparency ought to come from policy makers and funding agencies. In any case, journals should always require that data be housed in public repositories. Moreover, both editors and reviewers should encourage authors to provide as much information as possible on the procedures, controls, and any extra analyses, including contradictory results, as these are the key to transparency and may eventually be very significant for reproducibility issues. Personally, I think publishing the reviewer’s comments together with the ans­ wers from the authors is a good step as well.”

“When I read papers from the 1970s, I see how far we have advanced in reporting our studies in terms of standardization and completeness. These guidelines go further by promoting transparency and data sharing. Opacity has no place in science, and policies for open access to results and data should be promoted. The availability of online resources has dramatically improved possibilities, but guidelines are needed to control how this is managed, to limit misuse due to vested interests and to secure the privacy of subjects.”

La comunitat s’està adonant que el diàleg té lloc en els blocs, en qualsevol lloc on els investigadors poden parlar entre si directament. La por, la incertesa i el dubte: aquestes són les raons per les quals encara estem esperant que les revistes estableixin les normes, quan és la comunitat científica mateixa qui ho hauria de fer. La pregunta no és què poden fer les revistes, sinó què podem fer nosaltres: negar-nos a revisar els papers quan no se’ns dóna accés a les dades, o fer que els casos de frau que coneguem surtin a la llum.»

“The culture of debate is in a worldwide state of apathy. Most private journals fear open debate, because of the legal implications in case of defamation, as well as other issues.

The community is coming to realise that dialogue is taking place on social media, blogs, and anywhere else researchers can talk to each other directly. Fear, uncertainty and doubt are the reasons we are still waiting for journals to set standards, while these should be addressed by the community.

The ques­tion is not what journals can do, but what you can do: refusing to review papers wi­thout access to the data or bringing known cases of fraud to light.”

Finançament de La Marató al PRBB Maruxa Martínez-Campos

l 30 d’octubre passat van ser lliurats els ajuts als 44 projectes de recerca seleccionats per rebre els més d’11 milions d’euros recollits en l’edició 2013 de La Marató de TV3 dedicada a les malalties neurodegeneratives. Cinc investigadors dels centres del PRBB coordinen algun dels projectes seleccionats o hi participen: Cristina Pujades i Antoni Serrano

del CEXS-UPF, Gian Gaetano Tartaglia del CRG i Jordi Peña Casanova i Patricia Robledo de l’IMIM. Cadascun dels projectes rebrà entre 300.000 i 400.000 euros, al llarg dels propers tres anys.

euros from the 2013 edition of La Marató de TV3 dedicated to neurodegenerative diseases. What is the strangest place you have ever been?

I went on a trip to Myanmar (previously Burma) with my wife, to a little village in the interior of the country where there was a wedding being celebrated and, I still do not know how, we ended up eating in the house of the happy couple... Can you imagine this happening here? No way.

What annoys you most?

Hypocrisy, injustice, dishonesty, arrogance... It annoys me that people forget their principles in order to achieve their personal goals, especially if this happens at the expense of other people.

Who is your favourite person?

I have a wonderful two-year-old daughter with whom I am madly in love.

What is your worst defect?

As a proper Taurus, I am very headstrong.

When I get something into my head, I have to do it...

What mistakes do you forgive easily?

Everybody has the right to make a mistake and ask for forgiveness.

Which characteristics do you value most in a person?

Good manners and respect, intelligence and humility, but above all, kindness.

Un projecte per millorar la qualitat de l’assistència

Manolis Kogevinas, nou director del Programa Educatiu Europeu en Epidemiologia.

Aquest programa d’aprenentatge intensiu té lloc a Florència cada estiu. Dirigit a epidemiòlegs, professionals de la salut pública, estadístics i metges, l’any vinent tindrà lloc del 22 de juny al 10 de juliol i estarà coordinat pel CREAL.

A project to improve quality medical care

A

ith the aim of promoting patient safety and quality medical care, the European Commission organised a meeting in Rome on the 2nd and 3rd of December involving health experts, researchers, politicians and journalists. During the meeting they announced the results of eight projects funded by the EU Health Programme. One of these was the EURHOBOP Project (EURopean Hospital Benchmarking

by Outcomes in acute coronary syndrome Process), coordinated by Jaume Marrugat, a researcher at the IMIM. “The purpose of EURHOBOP is to define certain objective indicators for measuring the quality of medical care, specifically for coronary illnesses. This will help European hospitals to compare themselves, confidentially, with others that share the same characteristics, and will enable a ranking of European hospitals providing the best medical care in this field” explains Marrugat.

In order to define the indicators, the researchers have used patient registers from 70 European hospitals, including more than 27,000 cases of cardiovascular illnesses, and have analysed the procedures that are typically employed, like angiographs, thrombolysis and percutaneous interventions

mb l’objectiu de promoure la seguretat del pacient i la qualitat de l’assistència mèdica, la Comissió Europea va organitzar una reunió a Roma els dies 2 i 3 de desembre amb experts en salut, investigadors, polítics i periodistes.

Durant la trobada es van donar a conèixer els resultats de vuit projectes finançats pel Programa de Salut de la UE. Un d’ells va ser el Projecte EURHOBOP (EURope· an Hospital Benchmarking by Outcomes in acute coronary syndrome Processes), coordinat per Jaume Marrugat, investigador de l’IMIM. «EURHOBOP s’encarrega de definir uns indicadors objectius per mesurar la qualitat de l’atenció mèdica, en concret en les malalties coronàries. Això ajudarà els hospitals europeus a comparar-se, de manera confidencial, amb d’altres amb les seves mateixes característiques i a fer un rànquing europeu d’hospitals amb la millor atenció mèdica en aquest àmbit» explica Marrugat.

Per tal de definir els indicadors, els investigadors han utilitzat registres de pa­ cients de 70 hospitals europeus, amb més de 27.000 casos de malalties cardiovasculars, i han analitzat els procediments que s’utilitzen habitualment com ara angiogra­fies, trombòlisis o intervencions percutànies.

La Societat Respiratòria Europea premia Josep M. Antó. La Societat Respiratòria Europea (ERS) ha posat en marxa un prestigiós premi que reconeix l’excel·lència en la investigació, l’educació i el lideratge clínic en medicina respiratòria, i va anunciar els primers membres guardonats durant el seu Congrés Internacional de 2014 a Munic. El director del CREAL és un dels pocs espanyols d’entre els 129 escollits. Uns 50 membres addicionals seran seleccionats cada any. Segons l’organització, la comunitat de l’ERS reunirà els membres que han destacat en el seu camp per formar un consell assessor d’elit que serà consultat sobre diversos assumptes en els propers anys.

The European Respiratory Society recognises Josep M. Antó. The European Respiratory Society (ERS) has launched a prestigious award to recognize excellence in research, education and clinical leadership in the ­field

of respiratory medicine. It announced its first awarded members during its 2014 interna­tional meeting in Munich. The director of the CREAL was one of the few Spaniards amongst the 129 selected. About 50 new members will be chosen each year. According to the organization, the Fellowship of the ERS will include members who have excelled in their field, forming an elite advisory board to be called upon by the society.

Glòria Mas Martín rep un premi de la Fundació BBVA. La investigadora postdoctoral del laboratori de Luciano Di Croce al CRG, ha estat guardonada amb aquest ajut com a reconeixement a la seva carrera. La Fundació BBVA ha concedit 56 beques de fins a 40.000 euros cadascuna per un període d’entre sis i dotze mesos, selec­cionades entre més de 1.664 aspirants. «Estic al final del meu període postdoctoral i necessito un altre any per acabar un projecte. Aquesta beca em permetrà publicar el que em falta per ser competitiva, i així poder establir-me com a investigadora independent», diu Mas Martín.
