However, penetration below the horny layer was not observed. These results indicate the high permeability and stability of the MF liposome solution in the horny layer. Suppressive and Cytoprotective Effect of MF on UVA-induced ROS Intracellular oxidative stress levels of the human skin keratinocytes, which were administered with MF or hydrogenated lecithin and phytosterol without fullerene (Lpsm), were measured using the nitroblue tetrazolium (NBT) reduction assay. MF exhibited potent ROS scavenging activity, and suppressed UVA-induced intracellular ROS in keratinocytes. In the control sample, UVA-induced intracellular ROS level increased to 125%, as compared to that in the non-UVA-irradiated cells. When the keratinocytes were treated with MF during UVA-irradiation,

the absorbance by NBT-formazan decreased to the same level as that by non-UVA-irradiated cells (96%). The MF treatment was more effective than the Lpsm treatment, as shown in figure 3. Clinical Efficacy of MF The clinical efficacy of MF is evaluated with respect to barrier function recovery. The clinical study was performed on 10 subjects (4 men and 6 women) with an average age of 40.9 years. The subjects applied MF serum (C60: 2 ppm) or serum (without MF) to the inner side of their forearm (approximately 0.1g), and MF serum to half of their face (approximately 0.3-0.4g), twice a day for 28 days. Treatment sites were randomized, and subjects were blinded to the treatment during the forearm measurement. After the 28day trial, application of MF serum significantly increased the stratum

corneum hydration values on the subjects’ forearm, as compared with application of serum (figure 4-(a), P=0.052). Moreover, L-values and spectral reflectance increased (Figure 4-(b)) after application of MF serum, as compared with those pre-application. The results revealed that MF serum exhibits a high moisturizing efficiency on stratum corneum. In addition, applying MF serum improved brightness and transparency of skin. MF and its active functional ingredients help in reducing oxidative stress in keratinocytes and in restoring skin barrier function with their potent antioxidative properties. MF is also a simple liposome formulation to synthesize and the original liposome using MF has excellent skin penetration property. MF has potential application as a multifunctional active ingredient in cosmetic. .
