PRL-8-53 powder (51352-87-5) History

PRL-8-53 (also known as Methyl 3-(2-(benzyl(methyl)amino)ethyl)benzoate or PRL-8-53 HCl) is a Nootropic research compound. It was first synthesized by a medical chemistry professor, Nikolaus Hansl at Creighton University in the 1970s, during research into amino ethyl meta benzoic acid esters. [1] The compound is derived from benzoic acid and phenylmethylamine (Benzylamine) and has been shown to have memory-enhancing effects.

The exact mechanism of action for PRL-8-53 is not yet fully understood. However, in the original patent research, it was indicated that dosages of up to 200 mg/kg did not possess stimulant properties. Furthermore, dosages of 20 mg/kg did not potentiate the effects of dextroamphetamine.

PRL-8-53 (51352-87-5) Mechanism Of Action

Research has shown that PRL-8-53 exerts influence on the cholinergic system – by increasing dopamine signal transduction (“potentiates dopamine”) and partially inhibiting serotonin release. PRL-8-53 was also able to reverse the catatonic and hypnotic effects of reserpine.

Other than this, there is still room for further research into the mechanism of action by which PRL-8-53 exerts its effects.

Further Scientific research: Unfortunately, there is a very limited amount of scientific research that has been conducted on PRL-8-53 to this date. Independent replication of experiments conducted during the patent application would go a long way toward confirming PRL-8-53’s mechanisms and efficacy.

Researchers found that oral supplementation of PRL-8-53 was able to improve avoidance learning in rats. One human study has also been done on PRL-8-53. Each participant ingested a placebo and PRL-8-53, at separate times, in a double blind crossover study. Participants then took a memory test, consisting of twelve monosyllabic words in a particular order. The test was conducted three times, immediately after supplementation, one day after supplementation, and four days after supplementation.

People that remembered more than eight words after taking a placebo experienced minor benefits, but people over 30 or with poor short term memory benefitted greatly from supplementation. In some cases, their score on the word test doubled.

The effect of 3-(2-benzylmethylaminoethyl) benzoic acid methyl ester hydrochloride (PRL-8-53) on learning and on retention of verbal information in human subjects was investigated. Using the serial anticipation method under double-blind conditions it was found that PRL-8-53 causes slight improvement of acquisition. Retinetion of verbal information was found improved to a statistically significant degree (most P values better than 0.01, some better than 0.001). No significant changes were found for either visual reaction time or motor control after drug when compared with placebo values.
