Phenethyl Caffeate powder (104594-70-9) is an alkyl caffeate ester in which 2-phenylethyl is the alkyl component. It has a role as an antineoplastic agent, an anti-inflammatory agent, an immunomodulator, a metabolite, an antioxidant, a neuroprotective agent, an antiviral agent and an antibacterial agent.

Phenethyl caffeate powder is the phenethyl alcohol ester of caffeic acid and a bioactive component of honeybee hive propolis, with antineoplastic, cytoprotective and immunomodulating activities. Upon administration, Phenethyl caffeate inhibits the activation of nuclear transcription factor NF-kappa B and may suppress p70S6K and Akt-driven signaling pathways. In addition, Phenethyl caffeate inhibits PDGF-induced proliferation of vascular smooth muscle cells through the activation of p38 mitogen-activated protein kinase (MAPK) and hypoxia-inducible factor (HIF)-1alpha and subsequent induction of heme oxygenase-1 (HO-1).

Phenethyl Caffeate Mechanism Of Action

Phenethyl caffeate is a constituent of the propolis of honeybee hives and has been shown to contain inhibitory effects on induced papilloma promotion in mice. Studies have shown that Phenethyl caffeate acts as an anti-proliferative agent in over-productive cell growth. It is also shown that Phenethyl caffeate can cause DNA fragmentation and apoptosis which can have toxic results towards cloned rat embryo fibroblast (CREF) cells. In addition, CREF cells depleted of cellular glutathione (GSH) seemed to be more sensitive to Phenethyl caffeate-induced cell death.

Phenethyl caffeate has been reported to have anti-carcinogenic, anti-inflammatory, and immunomodulatory properties. Phenethyl caffeate also inhibits HIV-1 integrase.

In other animal models, this compound inhibits 5-lipoxygenase and decreases blood pressure, collagen deposition, and other atherosclerotic biomarkers. In animal and cellular models of colorectal cancer, phenethyl caffeate induces apoptosis and inhibits cellular and tumor growth.
