E

l nou Institut d’Inves­ tigació Sanitària Hospital del Mar es troba en el procés final d’acreditació per integrar-se en la xarxa d’instituts de recerca vinculats al Servei Nacional de Salut.

Aquests instituts s’estan creant per fomentar l’associació dels hospitals amb universitats i altres organismes de recerca, amb l’objectiu d’apropar la recerca bàsica, epidemiològica i clínica, i potenciar el concepte d’hospital com a centre de recerca. La constitució de l’IISHospital del Mar reforçarà el seu reconeixement extern i obrirà una nova via d’obtenció de recursos competitius, preferentment en el camp de la investigació translacional.

L’IIS-Hospital del Mar agrupa professionals de la UAB, la UPF, l’IMIM, el CRG, el CMRB, el CREAL i el PRBB al voltant de l’Hospital del Mar, i el seu ens de gestió és la FIMIM. La seva organització científica s’estructura en vuit programes de recerca i el seu marc organitzatiu s’ha establert en un conveni aprovat per les institucions participants.

Cinc anys, cinc kilòmetres Maruxa Martínez-Campos

A

mb el lema «Salut i kilòmetres» gravat a les samarretes de color taronja, quasi 100 residents del PRBB van sortir el 30 de maig de la plaça interior del parc per córrer plegats cinc kilòmetres al llarg del Passeig Marítim. La iniciativa, sorgida d’un grup de corredors del PRBB, tenia com a objectiu simbòlic celebrar el 5è aniversari del parc: cinc anys, cinc kilòmetres.

Durant aquests cinc anys, més de 1.300 residents han treballat junts per fer avenços en la recerca biomèdica i trobar formes de millorar la salut de la població. I ho han fet tot compartint el seu dia a dia: moltes hores de feina, però també alguns moments d’oci, que han servit perquè tant els científics com el personal de suport del PRBB s’hagin conegut millor. S’han trobat, fora de

l’horari laboral, per fer activitats que els uneixen, tant esportives com culturals, solidàries o festives; massivament o en petit comitè. El teatre, el cant coral, la capoeira, el ioga, les danses africanes, el futbol, el bàsquet, els esmorzars i mercats solidaris, les sortides dels runners, les festes que cada any reuneixen un gran nombre de residents o el campionat de vòlei platja amb més de 500 participants en són alguns exemples.

Totes aquestes activitats, moltes d’elles iniciatives dels residents i organitzades per ells mateixos, així com les relacions i amistats que hi han sorgit, han facilitat la convivència entre tots els residents del parc, i han generat oportunitats i ganes de col·laborar en projectes comuns. El PRBB ha avançat molt en aquests cinc anys, i l’empenta dels seus residents hi ha tingut molt a veure.

Five years, five kilometres

W

ith the slogan "Health and kilometres" emblazoned on their orange shirts, on May 30, nearly 100 PRBB residents assembled in the inner square of the park to run five kilometres together along the sea front. The initiative, which a group of runners of the PRBB came up with, symbolises the celebration of the 5th anniversary of the park: five years, five kilometres.

Over these past five years more than 1,300 residents have worked together, making advances in biomedical research and finding ways of improving people's health. They have done it all sharing, day in and day out, not only many hours of work, but also leisure time. These more relaxed moments have helped both the scientists and support staff of the PRBB to get to know one another better.

They have got together, outside working hours, for joint sporting, cultural, supportive or festive activities, in big groups and small. The theatre, choir, capoeira, yoga, African dancing, football, basketball, charitable breakfasts and markets, the outings of runners, the parties which each year bring together a large number of residents and the beach volleyball championship which has over 500 participants are some examples.

All of these activities, many of them initiatives of the residents which they organised themselves, and the relationships and friendships that have been formed, have made the coexistence of all the residents of the park easier, and have led to opportunities and the desire to cooperate in joint projects. The PRBB has advanced considerably in five years, and the drive and energy of the residents has been intrinsic

DESTACAT / HIGHLIGHT Bon posicionament de l’IMIM i el CRG Segons l’informe Indicadores bi­ bliométricos de la actividad cientí­ fica española 2008, realitzat pel grup SCImago per encàrrec de la Fundación Española para la Ciencia y la Tecnología (FECYT), el CRG i l’IMIM es troben en primera i quarta posició, respectivament, entre les institucions sa­nitàries espanyoles amb una major ràtio de citacions per document.

Ambdues institucions comparteixen, a més, llocs destacats

en el grau d’internacionalització de la recerca que desenvolupen.

L’informe destaca la novena posició d’Espanya en el rànquing mundial per nombre de publicacions i el fet que les institucions catalanes segueixen sobresortint tant en producció com en visibilitat respecte de la resta de l’Estat.

IMIM and CRG rank highly According to the report entitled Bibliometric indicators of Span­ ish scientific activity 2008, by the SCIMago group, the CRG and the

IMIM are in first and fourth positions respectively amongst the Spanish health institutions with high ratios of citations per document.

Both institutions also do well with respect to the degree of internationalisation of their research.

The report highlights that Spain is ninth in the world rankings by number of publications, as well as the fact that the Catalan institutions continue to excel, both in production and visibility, with respect to the rest of the country

IIS-Hospital del Mar

T

he new Hospital del Mar Health Research Institute is in the final process of being accredited to join the network of research institutes linked to the National Health Service. Such institutes are being created by the Ministry of Science and Innovation to encourage the association of hospitals with universities and research organisations, with the aim of bringing together basic, epidemiological and clinical research, and promoting the concept of a hospital as a research centre. The constitution of the IIS-Hospital del Mar will reinforce its external recognition and will open a new avenue for obtaining competitive resources, fostering translational research.

The IIS-Hospital del Mar gathers professionals from the UAB, UPF, IMIM, CRG, CMRB, CREAL and PRBB around the Hospital del Mar, and is managed by the FIMIM. It includes eight research programmes and the organisational framework has been established in an agreement approved by the participating institutions.

TONI GABALDÓN – GRUP DE GENÒMICA COMPARATIVA DEL CRG

«Som a l’edat d’or de la genòmica comparativa» such as of the aphid, published last year, and the lynx, sequenced a few months ago.

The group studies very diverse topics, always using the same methodology: the comparison of sequences and the generation and analysis of trees. One of the interests of the group is to understand what happens after gene duplication: whether the new gene is lost or differentiated, and how fast this process is.

l valencià Toni Gabaldón va arribar al PRBB el 2008. D’aleshores ençà ha creat un grup amb una formació mixta de biologia i informàtica que es dedica a comparar genomes des d’un punt de vista evolutiu. Dues preguntes són a la base de la seva recerca: quins processos evolutius tenen lloc i com afecten la funció dels gens.

El grup té diverses línies de recerca prò­ pies i moltes col·laboracions. La diversitat dels seus estudis el porta a col·laborar amb gent d’arreu del món, des dels Estats Units i Europa fins al Brasil o Malàisia.

La seva recerca es basa en els arbres filogenètics. Aquests arbres mostren les relacions evolutives entre els gens de diferents espècies i estan basats en la comparació de seqüències d’ADN. L’anàlisi dels arbres dóna informació sobre quins gens s’han duplicat o perdut o quins han acumulat més canvis.

«Abans es creava un arbre comparant un sol gen en unes quantes espècies diferents.

Ara mirem tot el genoma, uns 22.000 gens; o sigui, que hem d’analitzar 22.000 arbres alhora», explica Gabaldón. Per això, una de les seves línies de recerca és el desenvolupament d’eines informàtiques amb capacitat per analitzar aquest gran nombre d’arbres alhora i per detectar patrons. Aquests programes han permès al grup participar en l’anotació d’alguns genomes, com el del pugó, publicat l’any passat, o el del linx, que s’acaba de seqüenciar fa uns mesos.

El grup estudia qüestions molt diverses, sempre amb la mateixa metodologia: comparar seqüències i generar i analitzar arbres. Un dels interessos del grup és entendre què passa després de la duplicació de gens: si el nou gen es perd o es diferencia i com és de ràpid aquest procés.

La transferència horitzontal de gens Una altra àrea d'estudi és la transferència horitzontal de gens (HGT), és a dir, el pas de gens d’una espècie a una altra: «Es creia que només passava a nivell de bacteris, però nosaltres hem demostrat que en fongs és molt freqüent, i també s’ha vist en insectes».

L’arbre de la vida és un altre dels seus temes d’interès, el qual intenta plasmar les relacions evolutives entre totes les espècies existents. Fins ara s’havia fet amb uns quants gens, però «ara tenim el genoma complet de

quasi 3.000 espècies i la tendència és que cada any es dupliqui –comenta el cap del grup–. Tenim una quantitat d’informació genètica inimaginable fa tres anys. Sento que estem a l’edat d’or de la genòmica comparativa. Ens calen, però, eines capaces d’aprofitar aquesta informació. Ara mateix, el nombre de seqüències disponibles augmenta molt més ràpid que la potència dels ordinadors». Encara que l’arbre de la vida és un concepte bàsic en qualsevol estudi evolutiu, no està lliure de controvèrsies. Actualment es discuteix si realment hi ha un arbre de la vida o si és més aviat una xarxa. I en això hi té molt a veure la HGT, perquè si els gens no es passen només de generació en generació, sinó entre espècies, això modifica la forma de l’arbre i fa que s’assembli a una xarxa.

Finalment, el grup estudia com fongs que normalment són part de la flora intestinal es converteixen en patògens oportunistes en individus immunodeprimits, com ara persones amb sida o pacients que han patit quimioteràpia. «Hi ha milers d’espècies que viuen al nostre cos i només una petita proporció es converteix en patògens. Per entendre el perquè, estem seqüenciant fongs d’espècies patògenes i no patògenes, i de soques més i menys virulentes», explica Gabaldón.

El grup està catalogant tots els canvis que troben al genoma d’aquestes espècies i en fa arbres filogenètics. En col·laboració amb experts experimentals, han introduït les diferents soques en macròfags humans per estudiar quins gens s’expressen i entendre com reaccionen els fongs quan es troben amb la primera línia de defensa humana, i com respon aquesta defensa a cada tipus de fong.

"We're in the golden age of comparative genomics"

V

alencian Toni Gabaldón arrived at the PRBB in 2008. Since then he has created a group with a mixed education in biology and computer science which is devoted to the comparison of genomes from an evolutionary stand­point.

Two main questions underlie their research: which evolutionary processes take place and how these affect gene function.

The group has several of their own lines of research as well as many collaborative projects. The diversity of studies leads to collaboration with people from around the world, from the United States and Europe to Brazil and Malaysia.

Their research is based on phylogenetic trees. These reveal the evolutionary relationships between genes of different species and are based on the comparison of DNA sequences. Analysis of the trees provides information about which genes have been duplicated or lost and which have accumulated the most changes.

"Previously, we created a tree by comparing a single gene in several different species. Now we look at the entire genome, some 22,000 genes, meaning we have to analyse 22,000 trees at the same time", explains Gabaldón. For this reason, one of his lines of research is the development of information technology tools capable of analysing this huge number of trees together, and detecting patterns. These programs have enabled the group to participate in the annotation of some genomes,

Another issue is horizontal gene transfer (HGT), i.e., the passage of genes from one species to another: "It was thought that this only happened on a bacterial level, but we have shown that in fungi it is very frequent, and it has also been seen in insects".

The Tree of Life is another of their areas of interest. It attempts to reflect the evolutionary relationships between all existing species. So far this has been done with only a few genes, but "we now have the complete genome of almost 3,000 species and the trend is to double this amount every year –says the head of the group–.

I feel we’re in the golden age of comparative genomics. But we need tools to take advantage of this information. Right now, the number of available sequences is increasing much faster than computing power".

Although the tree of life is a basic concept, it is not without controversy. Currently under discussion is whether the tree of life is actually a network. And HGT is important for this issue: if genes are not only passed from one generation to the next, but also between species, the shape of the tree is modified and looks more like a network.

Finally, the group is looking at how fungi which normally are part of the intestinal flora become opportunistic pathogens in immunocompromised individuals, such as people with AIDS. "There are thousands of species that live in our body and only a small proportion of these become pathogens. To understand why, we are sequencing pathogenic and non-pathogenic species of fungi, and strains which are more and less virulent", says Gabaldón. The group is cataloguing all of the genomic changes they find and constructing phylogenetic trees. In collaboration with experimental experts, they have introduced the different strains into human macrophages to study how the fungi react to them

Les propietats de l’oli d’oliva, reconegudes per Europa Rosa Manaut/Marta Calsina

L

’Autoritat Europea de Seguretat Alimentària (EFSA) ha realitzat un dictamen científic sobre les propietats saludables de l’oli d’oliva. Aquesta certificació ha estat possible gràcies a l’aportació del grup de recerca en risc cardiovascular i nutrició de l’IMIM (Institut de Recerca Hospital del Mar), dirigit per Maribel Covas.

«Amb els nostres estudis, hem demostrat que el consum d’oli d’oliva augmenta els nivells de lipoproteïna d’alta densitat (HDL), el colesterol bo, i que això és directament proporcional al contingut de polifenols que conté. També hem constatat la implicació dels polifenols en la disminució de l’oxidació lipídica, un dels principals factors de risc de malaltia cardiovascular», afirma Covas. La coordinadora del grup ha estat guardonada recentment amb un premi atorgat pels productors d’oli d’oliva de Catalunya.

The properties of olive oil, recognised by Europe

T

he European Food Safety Authority (EFSA) has conducted a scientific report on the healthy properties of olive oil. This certification was made possible thanks to the contribution of the cardiovascular risk and nutrition research group of the IMIM (Hospital del Mar Research Institute), directed by Maribel Covas.

"Our studies have demonstrated that the consumption of olive oil increases the levels of high density lipoprotein (HDL) and good cholesterol, and that this is directly proportional to its polyphenol content. We have also noted the involvement of polyphenols in the reduction of lipid oxidation, a leading risk factor for cardiovascular disease", says Covas. The head of the group was recently awarded a prize by the Catalan olive oil producers

Com troben el camí les proteïnes Laia Cendrós

nvestigadors del CRG han descobert un nou mecanisme per a la classificació i distribució de les proteïnes que han d’abandonar la cèl·lula en mamífers.

El laboratori de Vivek Malhotra ja havia descrit anteriorment el paper d’una proteïna del citosquelet, l’actina, en la distribució de les proteïnes durant la via de secreció.

Ara els investigadors descriuen, en un article a Developmental Cell, la implicació de l’actina en el manteniment de l’equilibri del calci a la regió trans del Golgi, la principal estació de classificació de proteïnes de la via de secreció.

Com que les proteïnes de secreció, com ara la insulina, els neurotransmissors o els

anticossos, tenen diversos destins, fins ara es creia que per ser distribuïdes necessitaven receptors específics. «Hem descrit un mecanisme que no implica receptors específics, sinó que és la concentració de calci a la regió trans del Golgi, crucial per a una bona distribució de les proteïnes», explica Julia von Blume, autora del treball. «És un subtil equilibri que es manté gràcies a la interacció del citosquelet d’actina, proteïnes de remodelació de l’actina i un transportador d’ions de calci. Si aquest equilibri es trenca, un gran nombre de proteïnes no arribaran al seu destí», afegeix.

Segons els autors, aquest descobriment ajudarà a entendre millor patologies com ara la diabetis o l’asma, així com la morfogènesi de la pell i dels ossos.

How proteins find their way

R

esearchers from the CRG have discovered a new mechanism for the classification and distribution of the proteins that have to leave the cell, in mammals.

The lab led by Vivek Malhotra had already described the role of a cytoskeleton protein, actin, in the distribution of proteins in the secretory pathway. Now the researchers, in an article in Developmen­ tal Cell, have described the involvement of actin in maintaining the balance of calcium in the Trans-Golgi Network, the main sorting station for protein secretory pathway.

Since the secreted proteins, like insulin, neurotransmitters and antibodies,

have various destinations, it was believed until now that their distribution needed specific receptors. "We have described a mechanism that does not involve specific receptors. Instead it is the concentration of calcium in the Trans-Golgi Network which is crucial for a good distribution of proteins", explains Julia von Blume, one of the authors. "It's a subtle balance that is maintained by the interaction of the actin cytoskeleton, actin remodelling proteins and a calcium ion transporter. If this balance is broken, a large number of proteins will not reach their destination", she adds.

According to the authors, this discovery will enable a better understanding of diseases such as diabetes and asthma, as well as skin and bone morphogenesis

ientífics suïssos i catalans han demostrat que la diversitat de resposta a un estímul extern en una població de cèl·lules depèn del grau de compactació de l’ADN de cada cèl·lula.

Aquest descobriment, publicat a la revista Science, proporciona el mecanisme molecular a través del qual les cèl·lules fixen un llindar que determinarà de quina manera respondran davant d’una situació d’estrès ambiental.

Totes les cèl·lules han de ser capaces d’adaptar-se als canvis que es pro­ dueixen en el seu entorn per sobreviure a les situacions d’estrès cel·lular. Aquesta resposta d’adaptació es basa en una reprogramació de l’expressió dels gens de cada cèl·lula. L’equip ha descobert que, davant d’un determinat estímul d’estrès, unes cèl·lules responen i d’altres no, generant una variabilitat entre la població en la seva resposta a l’estrès. Així, malgrat que totes les cèl·lules tenen el mateix ADN, l’expressió dels seus gens és

diferent. En el seu treball, els investigadors descriuen el grau de compactació de l’ADN com la causa principal d’aquesta variabilitat.

El treball s’ha dut a terme gràcies a la col·laboració entre un grup de Zuric dirigit per Matthias Peter i el grup dirigit per Francesc Posas i Eulàlia de Nadal a la UPF.

Cells respond differently to the same stresses

S

wiss and Catalan scientists have demonstrated that the diversity of response to external stimuli in a population of cells depends on the degree of DNA compaction in each cell. The discovery, published in the journal Science,

provides the molecular mechanism by which cells set a threshold which determines how they respond to a situation of environmental stress.

All cells must be capable of adapting to changes in their environment in order to survive situations of cell stress. This adaptive response is coordinated by specialised proteins and based on a reprogramming of the genetic expression of each cell. The team has discovered that faced with a determined stress stimulus, some cells respond while others do not, generating variability among the population in their response to the stress. Thus, although all of the cells have the same DNA, their genetic expression is different. In their work, the researchers describe the degree of compaction of DNA as the principal cause of this variability.

The work was carried out thanks to the collaboration between a group from the Institute of Biochemistry (IBC) at the ETH Institute of Technology (ETH) in Zurich, led by Matthias Peter, and the cell signaling research group at the UPF led by Francesc Posas and Eulàlia Nadal

Relacionen la progesterona amb el càncer de mama hereditari Laia Cendrós

D

os investigadors del CRG han descobert un nou mecanisme pel qual mutacions en el gen BRCA1 poden induir càncer de mama. El treball, que publica la revista Cancer Research, explica el creixement de cèl·lules canceroses amb defectes de BRCA1 a dos nivells: per l’increment de la quantitat de receptor de progesterona a les cèl·lules i pel seu efecte sobre l’expressió de gens que activen la proliferació cel·lular.

Mutacions al gen BRCA1 poden causar càncer de mama en el 80% dels casos, així com càncer d’ovari en un 54%. Una de les explicacions d’aquesta especificitat de

BRCA1 per desenvolupar càncer en teixits amb gran acció d’hormones femenines és la funció del gen en la regulació de l’activitat d’aquestes hormones.

Miguel Beato i Verónica Calvo han demostrat ara que, quan el gen BRCA1 està mutat, la cèl·lula té més receptors de progesterona, de manera que augmenta el seu efecte sobre la proliferació cel·lular. Els descobriments posen de manifest la relació directa entre un excés en els efectes de progesterona i el risc de desenvolupar càncer de mama. Això podria ajudar a dissenyar millors terà­pies que actuïn directament sobre les molècules implicades. Aquesta aportació també podria comportar nous canvis en la prescripció de tractaments anticonceptius hormonals.

Progesterone linked to hereditary breast cancer

T

wo researchers from the CRG have discovered a new mechanism by which mutations in the gene BRCA1 can induce breast cancer. The study, published in the journal Cancer Re­ search, explains the growth of cancerous cells with BRCA1 defects on two levels: through an increase in the quantity of progesterone receptor in the cells and by its effect on the expression of genes which trigger cell proliferation.

Mutations in the gene BRCA1 may cause breast cancer in 80% of cases, and ovarian cancer in 54% of cases. One of the explanations for this specificity of BRCA1 to develop cancer in tissues with

high activity of female hormones is the function of the gene in regulating the activity of these hormones.

Miguel Beato and Verónica Calvo have now demonstrated that when the BRCA1 gene is mutated, the cell has more progesterone receptors, which increases its effect on cell proliferation.

The findings reveal the direct relation­ ship between an excess of the effects of progesterone and the risk of developing breast cancer. This could help in the design of new therapies to act directly on the molecules involved. It could also lead to changes in the prescription of hormonal contraceptives, as it happened when the relationship between oestrogen levels and the risk of developing breast cancer was demonstrated

«Hem estat els primers a fer servir smart phones en un estudi real amb voluntaris» pe that the models will serve as a tool to assist policy makers with their decisions.

Maruxa Martínez-Campos Qui participa a TAPAS i quin és el seu objectiu? osaltres en som els coordinadors, però hi ha implicats grups de recerca de sis països europeus. Volem entendre l’impacte de les polítiques de transport actiu, com caminar o anar en bicicleta, en la salut de les persones.

Estem fent models quantitatius per tal de mesurar aquest impacte en sis ciutats: Barcelona, Basilea, Copenhaguen, París, Praga i Varsòvia. Per a això utilitzem tant dades de la bibliografia existent com dades locals.

N

Per a què serviran aquests models?

La idea és que es podrà escollir una política específica de transport actiu, per exemple, la introducció d’una xarxa de carrils bici o d’una tarifa de congestió com la de Londres, i preveure els efectes en la salut que es deriven de la seva aplicació. Esperem que serveixi com una eina que ajudi els polítics a l’hora de prendre decisions.

De quina mena d’efectes estem parlant?

Bàsicament, un increment del transport actiu comporta un augment de l’activitat física en la població. A més, la reducció de transports motoritzats implica menys gasos emesos i, per tant, una millora en la qualitat de l’aire i un fre al canvi climàtic. Però en els nostres estudis no oblidem els possibles efectes adversos, com ara un augment d’accidents o una major inhalació de contaminants per part dels qui caminen o van en bici.

Quins estudis heu fet fins ara?

Hem fet un primer model teòric sobre el sistema del Bicing i estudis experimentals que ens permetran ampliar el nostre coneixement. Un és una enquesta a 800 persones per entendre el seu comportament respecte al transport actiu. Hem complementat l’enquesta amb un estudi pilot amb 35 voluntaris per obtenir mesuraments més objectius. Durant cinc dies van dur a sobre uns aparells que recollien la seva localització amb un GPS i el seu nivell d’activitat física amb un software incorporat a un smart phone.

PERFIL / PROFILE Audrey de Nazelle va arribar fa tres anys i mig de la University of North Carolina Chapel Hill, on va fer un doctorat en Ciències Ambientals.

Aquesta matemàtica francesa, activista ecologista des de jove, sempre ha tingut clara la importància de les polítiques en salut pública. Actualment està fent un postdoc al grup de Mark Nieuwenhuijsen del CREAL, i juntament amb ell lidera el projecte TAPAS, que va començar el 2009 i finalitzarà el 2012.

Audrey de Nazelle arrived here three years ago from the University of North Carolina Chapel Hill where she had done a PhD in Environmental Science. This French mathematician, an ecological activist from a young age, has always understood the importance of politics in public health. She is currently doing a postdoc in Mark Nieuwenhuijsen’s group at CREAL, and together with him is leading the TAPAS project, which started in 2009 and which will run until 2012.

Per a què serveix aquest tipus d’informació?

Per exemple, per analitzar el tipus de ruta que la gent fa servir i de què depèn que triïn un mitjà de transport o un altre. També podem comparar les seves rutes amb els mapes de contaminació que tenim i analitzar la seva exposició.

Els smart phones són el futur de les anàlisis d’exposició?

Crec que sí que ho són. Aquesta tecnologia i d’altres com la teledetecció són part del que s’anomena «ubiquitous sensing». Pel que sabem, nosaltres hem estat els primers a fer servir aquesta tecnologia d’smart phones en un estudi real amb voluntaris. Però, si pensem que hi ha bilions de persones que fan servir el telèfon mòbil i que es podrien convertir en voluntaris que recullen dades, el potencial és immens. A més del que hem fet ara, es podrien afegir sensors de soroll, de contaminació, etc., i tot lligat a la localització exacta.

“We are the first to use smartphones in a real study with volunteers” Who is participating in TAPAS and what is its aim? e are the coordinators, but there are research groups from six European countries involved. We want to understand the health impact of policies on active transport, such as walking or cycling, and we are using quantitative models to measure this impact in six cities: Barcelona, Basel, Copenhagen, Paris, Prague, and Warsaw. To do this we use both the existing literature and local data.

The idea is that one can choose a specific policy of active transport, for example the introduction of a network of bike lanes or a congestion charge such as there is in London, and predict the effects on health that come about because of this. We ho-

Shiloh, from the Department of Human Molecular Genetics & Bioche­ mistry of the Sackler School of Medicine at the Tel Aviv University, Israel, investigates the DNA damage response. This research stems from his interest in the human genetic disorder ataxia-telangiectasia (A-T), in which a central axis of the DNA damage response is missing. To investigate this system he uses cell biology methods, gene targeting in mice, and systems biology methods including highthroughput screens, advanced proteomics and bioinformatics.

He has been invited by Juan Valcárcel (CRG).

What kind of effects are we talking about?

Basically, an increase in active transport means an increase in the physical activity of the population. In addition, a reduction of motorised modes of transport means less gases emitted and therefore an improvement in air quality and climate change mitigation. But our studies do not ignore the possible adverse effects such as an increase in accidents or a greater inhalation of contaminants by those who walk or cycle.

What studies have you done so far?

We made a first theoretical model of the Bicing system and undertook experimental studies that allowed us to increase our knowledge. These included a survey of 800 people to understand their beha­ viour with respect to active transport.

We supplemented the survey with a pilot study of 35 volunteers to obtain more objective measurements. For five days they carried around devices which gathered data about their location with a GPS, as well as their level of physical activity using a piece of built-in smartphone software.

What is this kind of information useful for?

We can, for example, analyse the types of routes that people use and what their choice of means of transport depends on.

We can also compare their routes with pollution maps and analyse their exposure.

So smartphones are the future of exposure analysis?

I think so, yes. Both this technology and other developments, such as remote sensing, which are part of what is known as "ubiquitious sensing". As far as we know, we were the first people to use this smartphone technology in a real study with volunteers. But if we think that there are billions of people who use mobile phones, and that they could all become data collection volunteers, the potential is enormous. In addition to what we have already done we could add noise or pollution sensors, for example, with everything pinpointed to its exact location

Caetano Anolles, from the College of Agricultural, Consumer, and Environmental Sciences at the University of Illinois, Urbana-Champaign, USA, is interested in finding creative ways to mine, visua­ lize and integrate data from structural and functional genomic research. In particular, he studies the evolution of macromolecular structures, the function and structure of proteins and ncRNA, the evolution of biological networks, the spontaneous mutation process and the interaction of plants and microbes.

Com s’empaqueta l’ADN en un espermatozou Laia Cendrós

E

n un treball publicat recentment a PLoS Genetics, investigadors del CRG descriuen com el genoma humà aconsegueix empaquetar-se en una cèl·lula d'esperma. En la majoria de les cèl·lules, l’ADN s’enrotlla al voltant de complexos proteics anomenats nucleosomes, que el mantenen compactat i regulen quins gens s’activen i quins es desactiven. En l’esperma, l’ADN ha d’estar encara més compactat per tal de cabre en el cap del petit espermatozoide. Com a resultat, els nucleosomes dels espermatozoides es mantenen tan sols en alguns llocs del genoma, i són substituïts, en la major part, per altres proteïnes capaces d’una major compactació.

Tanya Vavouri, que va fer aquest estudi al CRG i és ara cap de grup a l'IMPPC, i Ben Lehner, cap del grup de Sistemes Genètics del CRG, van trobar que és la mateixa seqüència del genoma, i en concret la proporció de bases G i C respecte a les d’A i T, la que determina on es conserven els nucleosomes en espermatozoides madurs.

«El genoma humà presenta variacions locals en el contingut de les bases G+C i A+T. El nostre treball suggereix que una raó per a l’existència d’aquesta variació pot ser especificar com l’ADN s’ha d’empaquetar en l’esperma», comenta Vavouri.

«Sorprenentment, els nucleosomes es conserven en els llocs d’inici d’alguns dels gens més importants –afegeix Lehner–. Com que els nucleosomes són modificats a l’esperma, la seva retenció en aquests llocs podria permetre transmetre informació addicional a la generació següent. Aquesta és una idea molt interessant que cal investigar més».

How DNA is packaged into a sperm cell

I

n a work recently published in PLoS Genetics, CRG researchers describe how the human genome is packaged into a sperm cell. In most cells the DNA is wound around protein complexes called nucleosomes that keep it compacted and regulate how genes are turned on or off. In sperm, DNA must be even more tightly compacted to fit into the tiny sperm head. As a result, sperm nucleo­ somes are only retained at a few sites, being replaced across most of the genome by other proteins that lead to higher compaction.

Tanya Vavouri, who did this study at the CRG and has now become group leader at the IMPPC, and Ben Lehner, head of the genetic systems group at the CRG, have found that it is the sequence of the genome itself that determines where nucleosomes are retained. DNA consists of four bases –A,T,C, and G– and it is the number of G and C bases that is important for the retention of nucleosomes in mature spermatozoa.

“The human genome shows local variation in the content of G and C ver­ sus A and T bases. Our work suggests that a reason for this may be to specify how DNA is packaged into sperm”, comments Vavouri. “Strikingly nucleosomes are retained at the start sites of

some of the most important genes –added Lehner–. Nucleosomes are modified in sperm, and so one could imagine that they may be transmitting extra information to the next generation at these locations. This is a really exciting idea that should be investigated further”

Les histones són proteïnes que regulen l’organització del material genètic. Els autors precisen que LSD1 actua evitant que l’expressió dels gens de diferenciació es dispari inadequadament. Això s’aconsegueix mantenint un balanç de marques epigenètiques que ajusten l’activitat dels gens i, per tant, són clau per determinar la identitat cel·lular.

El proper pas serà estudiar la funció de l’enzim LSD1 durant la diferenciació en diferents teixits.

STEFAN DIMITROV, Friday June 17. Dimitrov, from the Department of Cell Differentiation and Transformation of the Institut Albert Bonniot in Grenoble, France, is working on the epigenetic functions of histone variants and the role of aurora kinases in mitosis and cancer development. These problems are addressed by a combination of molecular and cell biology methods and newly developed physical and biophysical techniques, including protein-DNA UV laser crosslinking and footprinting, elasticity measurements of a single mitotic chromosome, optical tweezers for single chromatin fiber stretching, AFM and electron cryo-microscopy or FRAP. He has been invited by Miguel Beato (CRG).

To renew or to differentiate: LSD1 helps stem cells decide

CohenCory, from the Department of Neurobiology and Behavior at the University of California in Irvine, USA, is investigating the molecular and cellular mechanisms that control neuron differentiation and synapse formation, in particular, in the living brain. She is currently focusing on the involvement of neurotrophins during synapse stabilization in actively competing visual inputs or the differential effects of neurotrophins on pre- versus post- synaptic structures. She is also interested in elucidating molecular interactions between neurotrophins and other guidance cues that control synaptogenesis in the developing brain. She has been invited by Berta Alsina (UPF).

Villunger, from the Division of Developmental Immunology at the Biocenter, Innsbruck Medical University in Innsbruck, Austria, works on multiple aspects of cell death signalling. In particular, he focuses on the role and function of pro-apoptotic Bcl-2 family members of the BH3-only subgroup as well as initiator caspases in normal development and diseases of the haematopoietic system. Some of his ongoing projects include the analysis of the relevance of the BH3-only protein PUMA/bbc3 for the effects of anti-cancer therapy and tumour suppression or the identification of caspase substrates. He has been invited by José Yélamos López (IMIM-Hospital del Mar).

Com pot ajudar l’estadística a la recerca?

M

ai en la història de la humanitat s’havien recollit tantes dades sobre el que ens envolta com ara. Estan informatitzades les visites que es fan al metge de capçalera, les altes hospitalàries, els resultats de les ressonàn­cies magnètiques o les característiques dels accidents de trànsit. Les dades donen informació molt útil si s’analitzen correctament.

Sovint, però, les dades presenten problemes. És el cas, per exemple, de la manca de dades (missing) en un estudi perquè els que hi participen no acudeixen a totes les visites programades. També sorgeixen problemes quan en analitzar milions de bases d’ADN es troben relacions entre mutacions i malalties que no són reals, sinó fruit de l’atzar (falsos positius). Els estadístics han de saber mesurar i plasmar l’efecte d’aquests problemes en els resultats de l’estudi.

La modelització de les dades fa que prenguin formes que fan més fàcil la seva

anàlisi. Així doncs, per exemple, l’alçada de les persones segueix una distribució anomenada «normal» (simètrica; la majoria d’individus al voltant de la mitjana) i el nombre d’accidents de trànsit segueix una distribució de Poisson, mentre que el nombre de persones que moren en una unitat de cures intensives segueix una distribució binomial. La majoria de dades s’adapten a una distribució coneguda o a una barreja de distribucions, però no sempre és fàcil trobar la més adequada. Expressar resultats en forma de figura és, de fet, tot un art.

How can statistics help research?

N

ever before in human history has so much data been gathered on the things around us. Visits to the GP, discharged patients, MRI results and characteristics of traffic accidents are all computerised. All these data can provide useful information if properly analysed.

Often, however, the data presents problems. For example, data will be missing if the participants do not attend all scheduled visits. Or relationships between mutations

and diseases may be found which are not real, but rather chance (false positives). Statisticians have to find ways to measure the effect of these problems in the results of the study.

Modelling the data means it assumes forms that make it easier to analyse. For example, people’s heights follow a distribution known as 'normal' (symmetrical, with the majority of individuals grouped

around the average), the number of traffic accidents shows a Poisson distribution (the mean and the variability are equal), while the number of people who die in an intensive care unit has a binomial distribution. Most of the data fits a known distribution or a mixture of distributions, but it is not always easy to find the most appropriate. Expressing results as figures is, indeed, an art

és de dos-cents especialistes internacionals es van aplegar el maig al CRG per tal de celebrar la reunió final del projecte Consolider Epigenètica, on es van presentar els resultats obtinguts pels nou grups de recerca que hi han participat. L’objectiu del projecte era desentrellar el codi epigenètic, basat no en la seqüència d’ADN en si mateixa, sinó en l’estructura i la compactació de l’ADN. «La genètica i la biologia molecular dels propers vint anys se centraran en l’epigenè-

tica», afirma Miguel Beato, organitzador de la reunió i director del CRG. «Queda molta tecnologia per desenvolupar en aquest camp», insisteix.

El projecte ha reunit els millors grups de recerca espanyols en aquest camp i ha donat lloc en quatre anys a la publicació d’uns 150 articles. «Catalunya i Espanya estan jugant un paper important en la recerca epigenètica a Europa i, per això, el projecte Consolider Epigenètica ha estat crucial. És una llàstima que no tingui continuïtat. Seria raonable que els projectes que han donat bons resultats fossin ava­ luats i recolçats», declara Beato.

Epigenetics, an essential field in biology

M

ore than two hundred international specialists gathered on May 12 and 13 at the CRG to celebrate the end of the Consolider Epigenetics project and see the results presented by the nine participating research groups. The aim of the project was to untangle the epigenetic code, based not on the DNA sequence itself but rather on the DNA structure and compaction.

"Over the next twenty years genetics and molecular biology will be focused on

epigenetics", states Miguel Beato, organiser of the meeting and director of the CRG. "There still remains much technology to develop in this field", he insists.

The project has brought together the top Spanish research groups working in the field and has resulted in about 150 papers being published over the four years. "Catalonia and Spain are playing an important role in European epigenetic research, and the Consolider Epigenetics programme has been crucial. It's a shame that it is to be discontinued. It would be reasonable that the projects which have been successful were assessed for continuation", says Beato.

E

l dissabte 14 de maig es va realitzar l’acte de lliurament del VI Premi PRBB, organitzat per la UPF i el PRBB i que s’atorga als millors treballs de recerca d’estudiants de secundària en l’àmbit de les ciències de la salut i de la vida. En aquesta sisena edició es van presentar 123 treballs provinents de 29 comarques de Catalunya.

Un 70% dels participants van declarar que es volen dedicar a la medicina o a la recerca bio­

mèdica, mentre que un 11% encara estaven indecisos.

L’acte va tenir lloc a l’Auditori i va estar obert a tots els participants i els seus amics i familiars. Els més de 80 assistents van poder visitar l’edifici, veure l’exposició de tots els treballs a la plaça del PRBB i escoltar la presentació del treball guanyador, «Lavate­ ra olbia (espècie autòctona en regressió)», de Ferran Cirac, estudiant de l’IES Vilatzara, de Vilassar Mar. El segon premi va ser atorgat a Sergi Casadó, de l’Escola Pia de Sant Antoni, i el tercer, a David Mas, de l’Institut Mare de Déu de les Escoles Pies. Els premis d'enguany han consistit en un iPad i en passar un dia en un grup de recerca del PRBB per al primer premi, un netbook per al segon premi i una col·lecció de llibres de divulgació científica per al tercer premi.

VI Premi PRBB award ceremony

T

he award ceremony of the VI Premi PRBB, organized by the UPF and the PRBB, took place on Saturday May

14. This prize is given each year to the best research work by high school students in the field of health and life sciences. In this sixth edition 123 works were submitted from 29 regions of Catalonia. About 70% of the participants said they would like to work on medicine or biomedical research, whilst 11% were still undecided about what to study.

The ceremony took place at the auditorium and it was open to all participants, friends and family. More than 80 attendees visited the building, looked at all the works exposed at the inner square and listened to the presentation of the winner work, “Lavatera olbia (autoctonous species in regression)”, by Ferran Cirac, a student of the IES Vilatzara, de Vilassar de Mar. The second and third prizes were awarded to Sergi Casadó of the Escola Pia de Sant Antoni and David Mas of the Institut Mare de Déu de les Escoles Pies. This year, the first prize was an iPad and a day with a research group at the PRBB. The second prize was a netbook and the third one a collection of science popularisation books

Els nens i nenes entre 8 i 12 anys ara poden posar-se la bata blanca i explorar el món dels sentits i la percepció durant tot un matí, a partir d’experiments que ells mateixos fan al laboratori. A més, també tenen l’oportunitat de visitar les instal·lacions del centre.

El nou taller amplia el programa educatiu «Ciència al CRG», que fins ara oferia el taller «La cèl·lula, una fàbrica ben organitzada», on l’alumnat es qüestiona diversos aspectes sobre la cèl·lula i el seu funcionament. «L’objectiu del programa és despertar l’interès per la ciència i la descoberta entre els més joves i també donar a conèixer la recerca que s’està fent al centre

Durant el mes de maig s’ha realitzat una campanya informativa sobre aquest tema, la primera de les dotze campanyes mensuals que es realitzaran al PRBB sobre prevenció de riscos laborals.

Chemical product labels are changing.

From December 1, 2010, all labels of hazardous products have started to change with the aim of creating a global standard across all countries in the world. Both the new system and the old one will be valid until December 1, 2012. From that time onwards only the new system will be used. To make sure everyone gets to know the new labels, an information campaign has taken place in May. It is the first of the 12 monthly campaigns that will take place at the PRBB regarding risk prevention at work

The new workshop expands the educational programme “Science at the CRG” which up to now has been offering “The cell, a well-organized factory”, where students look at several aspects of the cell and its function. "The programme aims to arouse interest in science and discovery among young people and publicise the research that is being done both at the centre and in Catalonia", says Miguel Beato, director of the CRG. NOTÍCIA BREU / BRIEF NEWS Nous espais d'incubació UPF Business Shuttle. A partir de l’1 de juny estaran disponibles els nous espais d'incubació de la UPF Business Shuttle, que inclouen projectes de l’àrea de la biomedicina al PRBB. Els 290 m2 estaran gestionats per la Unitat d'Innovació i Parcs de Recerca de la UPF i s’ubicaran a la segona planta de l'edifici de la nova incubadora Almogàvers Business Factory, impulsada per Barcelona Activa. Abans de l’estiu s'hi instal· laran qGenomics, una spin-off recent constituïda per la UPF i el CRG, i Chemotargets, una spin-off de l’IMIM creada el 2006.

New UPF Business Shuttle Incubator. As of June 1 there will be new incubation spaces available which are to include projects from the biomedical groups at the PRBB. The 290 m2 site will be managed by the Unit for Innovation and Research Parks of the UPF and is located on the second floor of the Almogàvers Business Factory building, a new business incubator driven forwards by Barcelona Activa.

Two companies are set to move in before the summer: qGenomics, a recent spin-off from the UPF and CRG, and Chemotargets, a spinoff from the IMIM, founded in 2006

l «I Curs de formació per a personal experimentador», organitzat pel PRBB i l’Escola Bonanova i acreditat per la Generalitat de Catalunya, va tenir lloc el maig passat. Hi van participar 22 persones provinents d'institucions de recerca, de la indústria farmacèutica i de mòduls de formació professional. El curs, obligatori per als tècnics que treballen amb animals d’investigació, és el primer que es fa a Espanya a través d’una escola de formació professional. «La valoració dels participants ha estat molt positiva. La idea és repetir aquest curs cada any», explica Juan Martín Caballero, director de l’estabulari. D’altra banda, una col·laboració de l’empresa Harlan amb el PRBB ha donat lloc a les primeres Jornades de Formació Continuada per a professionals que treballen amb animals

he "I Training course for experimental personnel", organised by the PRBB and the Escola Bonanova and accredited by the Catalan government, was held last May. There were 22 participants from research institutions, the pharmaceutical industry and professional training programmes. The course, which is compulsory for all of the technicians who work with research animals, is the first carried out in Spain through a professional training school. "The participants’ assessment was very positive.

Now the idea is to repeat this course each year", explained Juan Martín Caballero, director of the animal facility. In addition, a collaboration of the PRBB with the company Harlan has resulted in the first Continued Training Seminars directed at professionals who work with research animals, from caregivers to researchers.

This course consists of six seminars to be held at the PRBB between May and November 2011. The topics covered include, among others, how to obtain transgenic mice or biological safety. The seminars are free, but registration is needed at harlanclientes@harlan.com

Enguany s'ofereix un contracte de dos anys per a la incorporació d'un d’investigador postdoctoral als laboratoris del CRG. «L’aportació de fons privats és molt important per mantenir i millorar la qualitat de la recerca que es duu a terme a Espanya», expressa Miguel Beato, director del CRG. 10th edition of the Novartis postdoctoral fellowships in genomics. These grants seek to contribute to the promotion of genomics research in Spain. This year they offer a two-year contrac for the incorporation of a postdoctoral researcher to the CRG. "The contribution of private funds is very important to maintain and improve the quality of research carried out in Spain", explains Miguel Beato, director of the CRG. Creu Roja atorga la Medalla d’Or a la Fundació Pasqual Maragall. Amb aquest guardó, Creu Roja Espanyola reconeix la tasca de la fundació en la recerca i divulgació de la malaltia d’Alzheimer. L'acte de lliurament va estar presidit per la Reina Sofia el 10 de maig passat a Logronyo, amb motiu de la commemoració del Dia Mundial de la Creu Roja i la Mitja Lluna Roja. En representació de la fundació van recollir el guardó Pasqual Maragall i Diana Garrigosa.

Pasqual Maragall Foundation awarded with the Red Cross Gold Medal. With this award the Spanish Red Cross acknowledges the work carried out by the foundation in promoting research into and knowledge of Alzheimer's disease. The ceremony was presided over by Queen Sofia on May 10 in Logroño, on World Red Cross and Red Crescent commemoration day. The award was received by Pasqual Maragall and Dia­ na Garrigosa on behalf of the foundation.

Nou portal sobre fibromiàlgia i fatiga crònica. Els hospitals del Mar i l’Esperança han posat en marxa un nou portal per tal de fomentar l’autogestió de la salut dels afectats per aquestes malalties: www.parcde salutmar.cat/fibromialgia. La pàgina web conté informació sobre el funcionament de la Unitat de Fibromiàlgia i Síndrome de Fatiga Crònica dels hospitals i continguts d’interès, com ara símptomes de les malalties, així com informació dirigida als professionals sobre el diagnòstic i tractament i sobre els projectes actuals de recerca. A més, s’ha creat un espai de relació metge-pacient.

New portal on fibromyalgia and chronic fatigue. The Hospitals del Mar and l’Esperança have launched a new portal to encourage and improve health self-management of those affected by these diseases: www.parcdesalutmar.cat/fibromialgia. The website contains information about the Fibromyalgia and Chronic Fatigue Syndrome Unit of these hospitals and content on symptoms and disease prevention, as well as information aimed at professionals on diagnosis and treatment and current research projects.
