Amlodipine Besylate (Amlodipine Besilate) is a type of medicine known as a calcium channel blocker (CCB). It is used alone or together with other medicines to treat hypertension (high blood pressure)and a type of chest pain called angina pectoris. High blood pressure adds to the workload of the heart and arteries. If it continues for a long time, the heart and arteries may not function properly. This can damage the blood vessels of the brain, heart, and kidneys, resulting in a stroke, heart failure, or kidney failure. High blood pressure may also increase the risk of heart attacks. These problems may be less likely to occur if blood pressure is controlled.

Amlodipine Besylate Tablets Dosage

The dosage of Amlodipine Besylate tablets is given below: Swallow Amlodipine tablet whole with a full glass of water. Take your medicine at about the same time each day, either morning or evening. Taking Amlodipine at the same time each day will have the best effect. Generic Amlodipine Besylate tablets can be taken with or without food. For both hypertension and angina, the recommended initial dose is 5 mg Generic Amlodipine once daily which may be increased to a maximum dose of 10 mg depending on the individual patient's response. Small, fragile, or elderly patients, or patients with hepatic insufficiency may be started on 2.5 mg once daily.

Generic Amlodipine Tablets Contraindications

Generic Amlodipine Tablets are contraindicated in patients with a hypersensitivity to Amlodipine Besylate or any inactive component present in the medication. Do not co-administer aliskiren with angiotensin receptor blockers (ARBs), ACE inhibitors, including Amlodipine and Benazepril

Hydrochloride in patients with diabetes.

Additional Contraindications for Amlodipine Besylate and Benazepril

Hydrochloride Combination Tablets

Amlodipine and Benazepril Hydrochloride is contraindicated in patients with a history of angioedema, with or without previous ACE inhibitor treatment, or patients who are hypersensitive to Benazepril, to any other ACE inhibitor, to Amlodipine, or to any of the excipients of Amlodipine and Benazepril Hydrochloride tablets. Amlodipine and Benazepril Hydrochloride is contraindicated in combination with a neprilysin inhibitor (e.g., sacubitril). Do not administer Amlodipine and Benazepril Hydrochloride tablets within 36 hours of switching to or from a neprilysin inhibitor, e.g., Sacubitril/Valsartan

Storage Instructions

Warnings

Symptomatic hypotension can occur, specially in patients with severe aortic stenosis. However, because of the gradual onset of action, acute hypotension is not likely.

Worsening angina and acute myocardial infarction can develop after starting or increasing the dose of Amlodipine tablets, specially in patients with severe obstructive coronary artery disease.

Titrate slowly when administering calcium channel blockers including Amlodipine Besylate Tablets to patients with severe hepatic impairment.

Side Effects

Most Amlodipine side effects are mild or moderate. Inform your doctor or pharmacist in case you experience any of the following common side effects of Amlodipine and they worry you. The most frequently occurring side effects of Generic Amlodipine tablets include swelling of the ankles, feet, face or hands, headache, dizziness, flushing, fast or irregular heart beats, tiredness, drowsiness or sleepiness, stomach pain or nausea. Tell your doctor immediately if you notice any of the following: changes in heart beat either fast or slow chest pain chest pain associated with exertion (angina) that lasts longer, is more severe or occurs more often dizziness or lightheadedness on standing up from a sitting or lying position, eye pain or change in vision changes in mood, feeling anxious or nervous symptoms of liver disease such as itching, yellowing of the skin and eyes, and dark coloured urine. These may be serious side effects that may need urgent medical attention. Serious side effects are rare.

Side Effects

Inform your doctor immediately or go to Accident and Emergency at your nearest hospital, if you notice any of the following serious side effects of Generic Amlodipine Tablets: Shortness of breath, symptoms of allergy such as skin rash and/or itching severe upper stomach pain, often with nausea and vomiting.

Overdose

Amlopres overdose can be expected to cause excessive peripheral vasodilatation with marked hypotension and possibly a reflex tachycardia. Dysrhythmias may occur following overdose with any calcium antagonist including Amlodipine Besylate.

Hypotension and bradycardia are usually observed in patients within 1 to 5 hours following overdose.

Hypotension can continue for more than 24 hours despite treatment.

Cardiac rhythm disturbances have been observed to persist for up to 7 days.

Overdose

If massive Amlodipine overdose should occur, start active cardiac and respiratory monitoring. Frequent blood pressure measurements are essential. Should hypotension occur, provide cardiovascular support including elevation of the extremities and the judicious administration of fluids.

If hypotension remains unresponsive to these conservative measures, consider administration of vasopressors (such as phenylephrine) with particular attention to circulating volume and urine output.

Amlodipine Besylate During Pregnancy

Amlodipine Besylate tablets have been classified by the US FDA as Pregnancy Category There are no adequate and well-controlled studies in pregnant women. In animal reproduction studies, there was no evidence of adverse developmental effects when pregnant rats and rabbits were treated orally with Amlodipine during organogenesis at doses approximately 10 and 20-times the maximum recommended human dose (MRHD), respectively. However for rats, litter size was significantly decreased (by about 50%) and the number of intrauterine deaths was significantly increased. Amlodipine should be used during pregnancy only if the potential benefit justifies the risk to the fetus.

Benazepril Hydrochloride During Pregnancy

Benazepril Hydrochloride tablets have been classified by the US FDA as Pregnancy Category D. Benazepril Hydrochloride tablets can cause fetal harm when administered to a pregnant woman. Use of drugs that act on the renin-angiotensin system during the second and third trimesters of pregnancy reduces fetal renal function and increases fetal and neonatal morbidity and death. When pregnancy is detected, discontinue Amlodipine Besylate and Benazepril Hydrochloride tablets as soon as possible.
