EFFECT OF ERBB2 ON GLUTAMINASE EXPRESSION IN BREAST CANCER CELLS
ErbB2isanoncogenethatisoverexpressedincertaintumors,most frequentlyinbreastcancers. ItcanactivatethePI3K/AKTandMAPK signalingpathwaystoenhancetumorgrowthandmetastasis. Glutaminaseis anenzymethatbreaksdownglutaminetoprovideenergy,biosynthetic precursors,andreducingagentsforactivelydividingtumorcells. Our preliminarydatashowsthatErbB2increasesglutaminaseexpression. However,theexactmechanismbywhichErbB2upregulatesglutaminase expressionisnotyetclear. Myresearchgoalistodeterminetheroleofthe PI3K/AKTandMAPK/ERKpathwaysinErbB2-mediatedupregulationof glutaminase.
Toachievethisgoal,IutilizedMCF-10acells(normalhumanmammary cells)andNeuTcells(MCF-10acellswithconstitutivelyactiveratErbB2). I usedMCF-10acellstransientlytransfectedfor24hourswithacontrol plasmid,aplasmidoverexpressingAkt-Myr,andaplasmidoverexpressing pCMV-MEK,respectively. IalsoincludedMCF-10aandNeuTcellsascontrols. IcollectedthewholecelllysateforWesternblotanalysistodeterminethe expressionofErbB2,glutaminase,pAKT,totalAKT,pERK,totalERK,and α-tubulin(aloadingcontrol).
