Canadian excellence building the global bioeconomy BIOTECanada is pleased launch a new look for insights Magazine with our Spring 2008 issue. With enhanced features and new departments, insights has evolved to stay on the cutting edge, much like our industry’s leading technologies. In addition to the new layout, we’ve increased our national and international footprint by growing our distribution list, and we’ve established a strong editorial calendar for 2008.

All these changes set the stage for the theme of this issue: Canada becoming the world’s leading bio-based economy.

This theme of Canada’s potential to lead may appear audacious, but my response to that is simple: Why not us?

The infrastructure of the Canadian biotechnology industry and its commun­ity of partners has never been stronger. Our established leadership in vaccines, oncology, diabetes treatments, food technologies and rapidly expanding industrial biotechnologies offers Canada a unique opportunity. Increasingly, biotechnology offers solutions for modernizing our traditional industries, while affording our nation a new realm of economic and societal advantages Canadians are anxious to benefit from.

In my three years as president and CEO of BIOTECanada, I’ve witnessed changes that are moving Canada

forward in our bid to become a world leader. Our intellectual property environment has improved. The federal government recognized the need to create an internationally-aligned IP regime and extended data protection in Canada to meet global IP protection commitments, thus providing additional incentives for biotechnology investment.

The business environment has improved.

The 2008 federal budget announced changes to Canada’s Scientific Research & Experimental Development tax credit program advocated for by BIOTECanada and our members. Improvements to the program mean more companies will be eligible for refundable tax credits on R&D spending, and companies that previously didn’t qualify for refundable credits can now obtain up to $1 million annually from the federal government.

Canada also passed an amendment to its tax law that removes restrictions to foreign investment by recognizing Limited Liability Companies in Canada. We are making it easier to invest and succeed in Canada.

A number of funding programs and incentives for business exist in Canada, and you’ll read about them throughout this issue. In essence, the foundation is in place for construction to begin on the skyscraper of Canadian biotech.

And dominating the biotech skyline means huge market potential for our innovations. One conservative estimate places the global biotechnology market value in excess of U.S. $700 billion by 2020. Others place it in the trillions.

Canada will remain at the forefront of this global development.

Indeed, by recognizing the value of the industry and the partnerships we are forging around the world, stakeholders from across the board in Canada are aligning to support and promote our biotech industry.

The Canadian government recognizes the importance of growing Canada’s knowledge-based economy and is working to keep pace with technological growth. Industry is committed to building and succeeding in Canada.

Academia is training the next generation of biotech leaders and promising world-leading research. It will take time, but with the right leadership and ongoing commitment, we see no reason why Canada cannot become the world’s leading bio-based economy.

du printemps 2008 d’insights, BIOTECanada a le plaisir de vous faire voir son tout nouveau look. En apportant des améliorations à notre magazine et en y ajoutant des sections supplémentaires, nous l’avons actualisé afin qu’il demeure à jour, tout comme les technologies de pointe de notre industrie. En plus d’une présentation différente, nous avons aussi accru notre portée nationale et internationale en étoffant notre liste de distribution, et nous avons établi un calendrier éditorial ferme pour 2008. Tous ces changements préparent le terrain pour le thème du présent numéro, qui porte sur les efforts du Canada visant à acquérir la plus importante bioéconomie au monde.

Ce thème autour de la possibilité pour le Canada d’occuper le premier rang peut sembler audacieux, mais je vous répondrai simplement ceci : Pourquoi pas nous?

L’infrastructure du secteur canadien de la biotechnologie et de ses partenaires n’a jamais été plus solide. Notre position de chef de file en matière de vaccins, d’oncologie, de traitements du diabète, de technologies alimentaires ainsi que de biotechnologies industrielles en pleine expansion offre au Canada une occasion unique. pour moderniser nos secteurs d’activité traditionnels tout en procurant à notre nation un nouvel éventail d’avantages économiques et sociaux dont les Canadiens souhaitent ardemment profiter.

Au cours de mes trois années à la barre de BIOTECanada, j’ai assisté à des changements qui rapprochent de plus en plus le pays de notre objectif de devenir un chef de file mondial dans notre domaine. Par exemple, la situation de la propriété intellectuelle s’est améliorée. Le gouvernement fédéral a en effet reconnu la nécessité de créer un programme de protection de la propriété intellectuelle aligné sur ceux du monde entier et il a élargi la protection des données au Canada afin de répondre aux engagements internationaux en la matière, ce qui constitue une mesure incitative supplémentaire pour les investissements en biotechnologie.

Le contexte commercial s’est également bonifié. Le budget fédéral de 2008 a notamment annoncé un remaniement du programme canadien d’incitation fiscale Recherche scientifique et développement expérimental (RS&DE), que prônaient BIOTECanada et ses membres. Grâce aux améliorations apportées, un nombre accru d’entreprises auront droit à des crédits d’impôt remboursables pour leurs dépenses en R. D., et certaines sociétés qui n’y étaient pas admissibles jusqu’à aujourd’hui pourront désormais obtenir chaque année jusqu’à un million de dollars du gouvernement fédéral.

Le Canada a en outre adopté une modification à sa loi fiscale, qui élimine certaines contraintes imposées aux investissements étrangers en reconnaissant les sociétés à responsabilité limitée. Nous

contribuons ainsi à faciliter les investissements et la réussite dans le pays. Il existe au Canada plusieurs programmes de financement et mesures incitatives qui permettent aux entreprises de prospérer.

Nous enparlerons tout au long du présent numéro. Essentiellement, les fondations sont en place; il ne reste plus qu’à commencer à construire le gratte-ciel de la biotechnologie canadienne.

Qui plus est, l’énorme potentiel commercial de nos innovations domine actuellement l’horizon de la biotechnologie.

Selon une estimation prudente, le marché mondial de la biotechnologie devrait dépasser les 700 milliards de dollars américains d’ici 2020. D’autres croient même qu’il atteindra le niveau des trillions. Le Canada demeurera à l’avantgarde de ce développement d’envergure internationale.

En effet, conscients de la grande valeur de notre industrie et des partenariats que nous établissons partout dans le monde, tous les intervenants partout au Canada s’alignent afin d’ appuyer et promouvoir notre secteur d’activité. Le gouvernement fédéral reconnaît qu’il est primordial de faire croître l’économie du savoir au Canada, et il s’efforce de suivre la cadence de l’évolution technologique.

L’industrie est bien décidée à s’épanouir et à réussir dans le pays. Les universités forment la prochaine génération de chefs de file en biotechnologie et de meilleurs chercheurs prometteurs au monde. The Government of Canada’s long-term economic plan, ­A dvantage Canada, seeks to position Canada as an ­innovation leader. Innovation is crucial to strengthening Canada’s competitiveness and productivity and to improving our quality of life. Canadian innovators and entrepreneurs are doing truly outstanding work in many areas of biotechnology.

Their work is proof that Canadians can compete with the best the world has to offer.

As part of our economic plan, Prime Minister S ­ tephen Harper unveiled our Science and Technology (S&T) Strategy, Mobilizing Science and Technology to Canada’s Advantage. It is a multi-year framework that will: • Encourage companies to invest in research and ­development (R&D); • Support research excellence and align university research with the needs of business; and • Provide opportunities for Canadians and ensure firms have access to highly-skilled workers.

The S&T Strategy emphasizes helping Canada’s ­knowledge-based industries, including biotechnology, to become more globally competitive by strengthening strategic research, talent, private sector R&D and public-private sector partnerships. The strategy ­emphasizes greater investment in four areas, including three with significant interest for biotechnology companies: environmental science and technologies; health and related life sciences and technologies; and natural resources and energy.

are major R&D drivers and therefore key players in the economy of the future. Globally, Canada continues to rank among the top three countries in number of biotechnology firms, fourth in revenues and R&D expenditures, and fifth in biopharmaceutical inventions. Canada is truly one of the world’s leaders in biotech and health R&D. The Canadian biotechnology industry has demonstrated strong growth, with annual increases of 15 percent in R&D spending and 20 percent in revenues. Canadian biopharmaceutical companies have more than 500 products in various stages of R&D, representing one of the strongest product pipelines in the world. Life science companies undertake over 20 percent of Canada’s total private sector R&D, and the products arising from them are an important part of Canada’s innovative economy.

In response to these biotech industry strengths, the Canadian government has made significant investments and fostered partnerships with Canadian biotechnology companies in the areas of research, technology transfer, networks and centres of excellence to private sector R&D. Government of Canada spending on biotechnology has increased over five years from six to nine percent of total S&T expenditures, one of the

highest concentrations of S&T expenditures in biotech in the OECD. Federal health research spending, of which the majority is in biotech, has increased from 17 percent to 23 percent of government R&D expenditures.

The federal government wants to ensure Canadian ­b iotechnology firms can continue to strengthen their ­g lobal competitiveness and attract new global life science investments by building upon our world-class research and ­infrastructure, and ensuring a supply of highly-skilled workers. We have also responded to the innovative strengths of Canadian biotechnology and other life science industries through changes to intellectual property and health regulations, so that Canada is internationally competitive on data protection and regulatory approval times for health products.

Challenges we face The most important role of the government is to ensure a competitive marketplace and to foster an investment climate that encourages private sector innovation. We recognize Canadian biotechnology companies, like their counterparts in other countries, face difficult challenges in terms of commercialization. Over the last two budgets, key commitments that have been well received by the Canadian biotechnology industry include new investments in research infrastructure and genomics research; new Centres of Excellence for Commercialization and Research with the majority focussed on health discoveries; updates of Canada’s Scientific Research and Experimental Development (SR&ED) tax credit program to ensure it continues to encourage private sector R&D; reduction in corporate tax burden; and changes to ensure Canada is a best-in-class regulator. Although Canadian biotechnology has experienced remarkable growth—and many firms have seen improvements in their market opportunities—the primary ­challenge remains the need to attract sufficient capital both domestically and from abroad.

Our firms have demonstrated a remarkable capacity to find new and innovative ways to attract this risk capital. The Government of Canada has made efforts to strengthen the risk capital environment within Canada, including improved flow of capital between Canada and the U.S. and a new investment in the Business Development Bank of Canada for a new, private sector-led late-stage venture capital fund. Part of the government effort also involves getting the message out to the global community, in partnership with the Canadian biotechnology industry, about the competitiveness of the industry here. This is why we had a large delegation attend the industry’s premier annual event, the BIO International Convention 2007, in Boston, and will be out in force at BIO 2008 in San Diego.

Intense global competition is re-shaping how businesses compete and how commerce functions. The prerequisites for economic success, the raw materials of prosperity and the defining measurements of achievement are fundamentally BIOTECanada

En réponse à ces forces de l’industrie de la biotechnologie, le gouvernement du Canada a fait des investissements considérables et il cherche à établir des partenariats avec les entreprises canadiennes de biotechnologie dans les domaines de la recherche, du transfert de technologie, du réseautage et des centres d’excellence pour la R. En cinq ans, les dépenses du gouvernement fédéral pour la biotechnologie sont passées de six à neuf pour cent de l’ensemble des dépenses en sciences et technologie, constituant ainsi l’une des plus grandes concentrations de dépenses en sciences et technologie consacrées à la biotechnologie au sein de l’Organisation de coopération et de développement économiques (OCDE). Par ailleurs, les dépenses fédérales dédiées à la recherche en santé, dont la majeure partie sont investies dans la biotechnologie, ont augmenté de 17 à 23 pour cent de toutes les dépenses de ce gouvernement en R. D. Le gouvernement canadien souhaite s’assurer que les entreprises canadiennes de biotechnologie peuvent continuer à accroître leur compétitivité à l’échelle mondiale de même qu’à attirer de nouveaux investissements étrangers en sciences de la santé. Pour réaliser ce projet, nous comptons miser sur notre infrastructure de recherche de niveau international et veiller à la disponibilité de travailleurs hautement qualifiés.

Nous avons également répondu aux forces novatrices de la biotechnologie canadienne et des autres secteurs des sciences de la vie en apportant des modifications aux règlements en matière de propriété intellectuelle et de santé de telle façon que le Canada soit concurrentiel à l’échelle internationale en ce qui concerne la protection des données et les délais d’approbation réglementaire pour les produits de santé.

Les défis à relever Le rôle le plus important du gouvernement consiste à veiller à la compétitivité du marché ainsi qu’à favoriser un climat d’investissement qui encourage l’innovation dans le secteur privé. Nous reconnaissons qu’à l’instar de leurs semblables dans les autres pays, les entreprises canadiennes de biotechnologie font face à certaines difficultés considérables au niveau de la commercialisation. Parmi les principaux ­engagements des deux derniers budgets à avoir reçu un bon accueil de la part de l’industrie canadienne de la biotechnologie figurent les nouveaux investissements dans les infrastructures de recherche et la recherche en génomique; la création des Centres d’excellence en commercialisation et en recherche, dont la majorité sont axés sur les découvertes en santé; la mise à jour du programme d’incitation fiscale Recherche scientifique et développement expérimental (RS&DE) en vue de voir à ce qu’il continue à encourager la R. D. dans le secteur privé; la diminution du fardeau fiscal des entreprises; et les mesures visant à s’assurer que le Canada compte parmi les meilleurs en fait de réglementation.

Supporting R&D in Canada Originally created in 1985, the SR&ED program is designed to provide financial incentives for companies doing R&D in Canada through tax credits that can be used to feed a company’s development pipeline. The government’s 2008 budget announced valuable changes to the program aimed at recognizing the 21st century dynamic of discovery and development. These changes include: • I ncrease the expenditure limit for refundable tax credits by 50 percent, from $2 million to $3 million.

With refundability at 35 percent, this results in an additional $350,000 for eligible companies.

Moreover, the total refundability now increases to over $1 million.

• R aise the upper limit for the taxable capital phase-out range, increasing the amount of companies that can qualify for a tax credit. The limit has been increased from $15 million to $50 million.

• I ncrease the upper limit of the taxable income phase-out range from $600,000 to $700,000.

Companies that previously didn’t qualify for refundable credits can now obtain up to $1 million from the federal government.

• E xtend the SR&ED investment tax credit to certain activities carried on outside of Canada in situations where “A SR&ED performer is required to carry on a portion of its SR&ED activities outside Canada in support of its SR&ED carried on in Canada.” The budget introduced a 10 percent allowance for international research in these credits.

different from what they were even ten or 15 years ago. Global restructuring in many segments of health industries is having an impact on the opportunities for Canada to realize the benefits of public investments in health research.

This restructuring is also having an impact on Canadian biotechnology companies engaged in high-risk and long-term investments. We need to consider a new value proposition that will guide all of us in Canada to ensure our health industries remain globally competitive, and help Canadians realize the full social benefits of biotechnology.

Canadian commitment to biotechnology The Canadian biotechnology industry has demonstrated its ability to innovate, grow and pursue global opportunities.

The Government of Canada sees biotechnology as an important part of Canada’s innovative economy and has made significant investments to enable this industry to realize its full potential.

Change, planned or not, will happen and will challenge us. Within our own personal worlds, continuous change cannot be denied as we engage in the cycle of life. Our professions bring us into a much larger world, one that is shared by many others, each of whom can be secure one moment and threatened the next. Contemporary paradigms may no longer give us the return to which we have grown accustomed, either because they have become inefficient, obsolete or irrelevant.

As we move farther outward, we find ourselves in a global environment that while connecting us through technology, can expose us, rendering a vulnerability to financial, political, social and health issues on a scale unprecedented in our lives or indeed, in the life of human existence on planet Earth.

Although we live in uneasy times, there is a greatness that comes from the challenge of continuous change. Life endangered at one moment can also become a beneficiary of severe change when the imminent threat subsides.

In biology, we know and appreciate nutrients upon which life depends comes from replenishment by the challenging forces of the natural world. Seasonal temperature changes, floods and forest fires all play a role in allowing new life to arise and even flourish.

The value of scientific discovery and innovation is visible when technology is useful and most importantly, when it is widespread and available for the global good. In technology, change is as continuous as in the natural world. But it is the human mind that will challenge, develop, refine and re-define what is possible.

As a graduate of both agriculture and medicine, I can’t imagine a world without biotechnology. I am excited about the reality and the potential of biotechnology to develop new ideas and for the biotechnology industry to move them from innovation to application. For students who are our future researchers, biotechnology is as infinite as are their imaginations in seeking answers to the most challenging questions ever asked of human beings. Change is continuous, and we mark our lives by the way we adapt to change and give the world new life.

Making the deal How a small Canadian biotech company turned a personal battle into an international multi-million dollar deal

On December 17, 2007, BioMS Medical Corp. announced it signed one of the biggest deals in the history of Canadian biotech: to co-develop and market MBP8298 with pharmaceutical giant Eli Lilly & Company.

MBP8298 is the multiple sclerosis (MS) drug that BioMS is currently advancing through pivotal Phase III trials. In the deal, BioMS received an up-front payment of $87 million with potential milestone payments totaling $410 million, plus an undisclosed royalty on future sales of the drug, assuming it is ultimately approved by regulators.

For the team at BioMS, the deal represents one of the biggest milestones in the course of developing MBP8298—but it was not the first, nor will it be the last. “The first big milestone was more personal,”

“ The BioMS deal reflects the increased willingness of pharmaceutical companies to ensure opportunities are not missed.” explains BioMS chairman Clifford Giese. “It was when we became aware of MBP8298, which was being tested at the University of Alberta, and decided to enroll my wife, Robin, who has secondary progressive MS, in a phase I clinical trial. The day we realized some of the brutal effects of her disease were starting to diminish was one of the greatest days in our lives.”

MBP8298 and formed BioMS as a public company with the mission to commercialize MBP8298 globally.

“We realized from the start that at some point we would need a partner to help bring MBP8298 to market,” Kevin Giese adds. “While we’ve had great success at raising capital, the costs of conducting international Phase III clinical trials required to get MBP8298 approved are enormous. If a drug is successful in the clinic, the resources required to commercialize it on a worldwide basis are even greater.” Fortunately, BioMS was able to attract the attention of potential partners early on, and retained consultants from Deloitte to manage the initial unsolicited attention of potential suitors.

Indeed, by mid-2007, BioMS had enrolled the first patient in its pivotal U.S. trial, known as MAESTRO-03, and had completed an equity financing raising more than $40 million. At that point, partnering activity went into high gear, with several major pharmaceutical companies coming to Alberta to conduct due diligence, creating a competitive setting for potential partners—an ideal situation for BioMS. The current environment for public biotechnology companies in Canada has been trying, with valuations falling and sources of capital shying away from the sector. But the BioMS/Eli Lilly deal offers a ray of hope to Canadian biotechnology companies because, despite the challenges they face, the pharmaceutical industry faces an even greater threat: that they may not find

The BioMS deal reflects the increased willingness of pharmaceutical companies to ensure opportunities are not missed. With MBP8298 targeting one of the largest underserved markets in the healthcare sector with blockbuster potential, this was clearly an opportunity that Eli Lilly could not pass by.

In the meantime, Cliff and his wife, who now has been on the drug for twelve years, are enjoying their own personal celebration. “Before MBP8298, MS was steadily and relentlessly damaging Robin. Now that the progression of the disease has been slowed, we are free to live life fully again and look ahead with hope. It has changed everything.”

The route to market in each country is characterized by a unique process and standards that reflect the local conditions and priorities of that country. Canada is no different. The biologics supplier that develops a good understanding of this process is more likely to succeed in getting its product to Canadian patients.

Health Canada administers the regulatory approval system for new pharmaceutical products based on safety and efficacy.

Priority review is available for drugs that show the promise of filling an unmet need or providing significant improvement over existing therapies. Health Canada also allows earlier access to drugs, prior to formal approval, through the Special Access Program (SAP). Under the SAP, manufacturers from either within or outside of Canada may supply products for Canadian patients on a physician’s request. As of 2006, biologics manufacturers are also eligible for data protection.

Long before market approval, a manufacturer should be thinking about establishing a price and a marketing strategy.

For most drugs, there are three key components of the pricing and reimbursement system: • Patented Medicine Prices Review Board (PMPRB): A price review for patented drugs to ensure the price is not excessive; • Common Drug Review: An evidencebased assessment of the clinical and cost effectiveness of the drug for purposes of recommending coverage by the federal, provincial and t­ erritorial public drug plans; and, • Public Drug Plans: A review and final negotiation of price and the terms and conditions of reimbursement.

The PMPRB claims jurisdiction over the price charged by the manufacturer for patented medicines, even if supplied under the SAP. Although its jurisdiction only begins when a patent is issued, the PMPRB also claims retroactive jurisdiction during most of

Since it is not uncommon for a biologic to be sold before a patent is issued, it is important to be aware of the reporting requirements and price guidelines of the ­PMPRB at an early stage.

These guidelines are intended to ensure a price is not excessive.

For most new drugs, prices are limited to those of the most expensive existing treatment in the class. A drug that represents a breakthrough or substantial improvement can have a higher price, provided it does not exceed the median of prices for the same drug in seven other industrialized countries: France, Germany, Italy, Sweden, Switzerland, the United Kingdom and the United States. The PMPRB has powers to enforce price limits after giving the manufacturer an opportunity to be heard, but in its 20-year history, it has achieved a high degree of voluntary compliance, and over 90 percent of patented drugs are priced within its guidelines.

The Common Drug Review (CDR) is a newer process. Since 2003, it has reviewed new drugs on the basis of the manufacturer’s submission, to assess clinical and cost effectiveness of drugs and to make recommendations to the public drug plans for listing purposes. Positive recommendations from the CDR often involve recommendations for specific criteria or restrictions on coverage. The CDR process also prescribes clear timeframes and submission requirements. Its recommendations are published on the CDR website and help provide manufacturers, patients and others with a better understanding of the evidence considered by the CDR, and the relative weight put on that evidence. In response to a recent federal parliamentary committee report, the CDR has begun steps to increase the transparency and openness of its activities.

In recent years, the larger public drug plans have implemented new policies to increase their capacity to negotiate pricing and other conditions when making listing decisions. For the largest plan, in Ontario, the executive officer may negotiate confidential payments or rebates with a manufacturer. This policy is intended to allow the government to exercise its buying power to obtain a better net price than the published price generally available in the market.

Le Programme commun d’évaluation des médicaments (PCEM) constitue une étape plus récente. Depuis 2003, il examine les nouveaux médicaments en fonction du dossier présenté par le fabricant afin d’évaluer l’efficacité clinique et la rentabilité des médicaments et de faire des recommandations aux régimes publics d’assurance-médicaments à des fins d’inscription aux listes. Les recommandations positives du PCEM sont souvent limitées à des critères précis et comprennent des restrictions en ce qui concerne la couverture. Le processus du PCEM prescrit aussi des délais précis et la nécessité de présenter certains documents. Il publie ses recommandations sur son site Web, ce qui aide les fabricants, les patients et les autres intervenants à mieux comprendre les critères objectifs dont il tient compte et le poids relatif de chacun de ces critères. Over the past 200 years, vaccines have been the basis of a systematic attempt to reduce or eliminate ill health and mortality associated with infectious diseases. By stimulating the immune system to provide humans with protection against what were once common and deadly infections, vaccines have done more to reduce human mortality than any other medical intervention—and they have done so in the most cost-effective manner.

It is estimated the introduction of polio immunization in Canada in 1955, and the subsequent eradication of polio in this country, has saved approximately $3.3 billion in healthcare costs. Vaccine manufacturers in Canada expect those savings to increase as the next generation of vaccine technology, targeting both infectious and non-infectious diseases such as cancer and neurological disorders, is introduced.

New biotech approaches to vaccine ­development Biotechnology is a key driver in the development of new understanding about the immune system, opening the door to a host of new vaccines directed at the more difficult communicable diseases and chronic diseases. Throughout the nineteenth and twentieth centuries, most vaccines were developed by the stimulation of the immune system to produce antibodies. Advances in immunology have given us a deeper understanding of cell-mediated immunity, which is essential to the development of vaccines for HIV, tuberculosis and malaria.

Likewise, advanced genetic engineering and molecular biology are enabling the application of new strategies in vaccine development (Table 1). These second-generation vaccines

Proteomic analysis of disease-causing organisms will eventually lead to the construction of synthetic peptide-based vaccines.

Combination vaccines and vaccines that take advantage of new routes of administration (oral, intranasal, transdermal, intradermal) are also playing an important role in the ­development of future vaccines for a host of communicable disease targets (Table 2). New adjuvants will be used to improve immune responses to influenza and other vaccines.

While universities have achieved impressive gains in fundamental life sciences research in recent years, crossing the gulf from basic research to viable economic and health gains has proven a difficult journey. The current “commercialization gap” exists due to the high risk and cost of developing new therapeutics and the incomplete nature of the supporting data packages produced in academia. Canada, however, has a mechanism that bridges that gulf to help companies be successful in bringing products to market and improving the health of Canadians.

Step one: Take a new idea and add collaboration The Centre for Drug Research and Development (CDRD) is a groundbreaking British Columbia-based organization with a unique solution to bridge the commercialization gap. CDRD aims to advance early-stage health-related discoveries through development so they can be quickly brought to the stage where they can attract funding required for clinical trials. It was recently named a Canadian Centre of Excellence for Commercialization and Research and received funding from a federal government program supporting research commercialization in Canada.

Integrating innovation: Canadian leadership in industrial biotechnology Public and private funding commitments have positioned Canada as a leader in developing bio-petro industrial products. The public face of industrial bioproducts is ethanol and biodiesel. However, the real valueadded will come from bio-based fibers, plastics, and resins used in building and construction materials, automotive parts, biomedical devices, textiles and packaging and containers — products that could be developed on a large-scale in Canada.

The University of Western Ontario’s Research Park, Sarnia-Lambton Campus sits on the Ontario-Michigan border at the southern tip of Lake Huron. It received a boost in February when it was named a Centre of Excellence for Commercialization and Research. The federal government $15 million investment will be matched by over $5 million from other sources and will leverage over $30 million in additional funding commitments from industry, institutions, municipalities and provincial governments.

The Bioindustrial Innovation Centre, now under development at The Research Park, will create Canada’s leading centre for the commercialization of large-scale industrial biotechnology. The public-private partnerships plan to attract over $1 billion in new investments by 2014 and position Canada as the world leader in integrating sustainable feedstock into existing products and value chains.

The Pan-Provincial Vaccine Enterprise (PREVENT), a non-profit company, aims to bridge this gap by developing affordable vaccines in this critical niche of public health.

Based at the University of Saskatchewan, PREVENT is one of 11 new Centres of Excellence for Commercialization and Research established through the Networks of Centres of Excellence program. Awarded $15 million and an additional $10.5 million of in-kind support, PREVENT will make use of expertise at: • The Vaccine and Infectious Disease Organization (VIDO), which brings preclinical research capacity (Biosafety Level 2 laboratories and animal facilities) and is leading development of the International Vaccine Centre (InterVac), Canada’s largest BSL 3 laboratory dedicated to vaccine development and capable of vaccine trials in large animals.

• The Canadian Center for Vaccinology (CCfV), a joint initiative of the IWK Health Centre, Capital Health and Dalhousie University. The CCfV brings a 30,000 square foot vaccine research centre for basic translational science, epidemiology and clinical trials (Phases I-III), and implementation and evaluative policy research.

• The BC Centre for Disease Control (BCCDC), which delivers prevention services and conducts research on the epidemiology, pathogenesis and control of infectious diseases and environmental hazards. The BCCDC will contribute to PREVENT through vaccine antigen discovery methods (with secured patents) for respiratory viral diseases and sexually transmitted pathogens.

By shouldering the risk of early-stage vaccine development, PREVENT will strengthen Canada’s vaccine industry, promoting growth, investment and improved global competitiveness. Step two: Mix your expertise for the desired consistency Bringing a drug to commercial viability requires broad resources and expertise.

Dr. Sandra Dunn is a cancer investigator affiliated with the University of British Columbia’s Department of Pediatrics, Experimental Medicine and Medical Genetics. She identified a protein necessary for cancer cell survival, but needed an effective means to block it – an objective that required considerable expertise in chemistry. Without an inhibitor, Dunn could not secure the funding to advance her research to the next level. Her research stalled.

She came to CDRD for help. Head of CDRD’s Division of Drug Design and Synthesis, Dr. Marco Ciufolini, worked with Dunn to design an effective inhibitor based on the location of key amino acids in the newly discovered protein. In a

education is difficult to obtain, but doing so with a chronic illness presents additional challenges. The UCBeyond Crohn’s Disease and Rheumatoid Arthritis Scholarship Program makes education an achievable goal for students living with illnesses, while also building awareness of the diseases.

In 2007, 14 $5,000 scholarships were awarded and because of its success, the program will be offered again in 2008.

Watch out, MIT—here come the Butanerds Last November, a team of students from the University of Alberta who call themselves the “Butanerds” took the top prize in the Energy and Environment Category at the fourth annual International Genetically Engineered Machine (iGEM) competition, held at Boston’s MIT. The 10-member team bioengineered E. Coli to produce butanol by introducing the genes responsible for butanol production from Clostridium acetobutylicum (endogenous butanoate pathway) into E. Coli. The butanol can be used as an alternative biofuel, eventually replacing gasoline. The team is still perfecting their technology to make production more efficient by working with computer models to increase production levels.

Attention MIT : voici les Butanerds En novembre dernier, une équipe d’étudiants de l’University of Alberta, qui s’est donné le nom de « Butanerds » (littéralement, les « petits génies du butanol »), a récolté le plus prestigieux prix de la catégorie Énergie et environnement lors du quatrième concours annuel International Genetically Engineered Machine (iGEM) qui se tient au MIT, à Boston.

L’équipe formée de 10 membres a modifié l’E. coli par voie de bio-ingénierie afin qu’il produise du butanol : elle a transféré les gènes responsables de la production de butanol du Clostridium acetobutylicum (vecteur endogène du butyrate) dans l’E. coli. Le butanol est un biocarburant qui pourrait remplacer l’essence. L’équipe continue de perfectionner sa technologie afin de rendre sa production plus efficace à l’aide de modèles informatiques.

Educating the educators Renseigner les enseignants Teachers spend every day in front of a class—but occasionally, they get to be students. Over 100 educators from across Canada and the United States attended the Second Annual National Bioscience Educators Conference (NBEC) at Toronto’s Seneca College on February 28 and 29, 2008. Conferences such as the NBEC are instrumental in equipping teachers with current information to take back to the classroom to inspire future biotech leaders.

Organized by the Canadian Biotechnology Education Resource Centre (CBERC), the conference brings leading scientific and commercial experts together to provide informative sessions on genomics and bioethics, agri-foods, bar-coding biodiversity, high school science curriculum, water pollution and aquatic ecosystems, and much more. It also gives teachers hands-on experience at interactive workshops.

The event is supported by the Ontario Ministry of Research and Innovation, Monsanto, Seneca College, AstraZeneca, GlaxoSmithKline, Borden Ladner Gervais, BIOTECanada, Bio Business, Ontario Genomics Institute and UCB Pharma. Plans are underway for 2009 conferences to be hosted at different locations across Canada, in partnership with regional biotechnology organizations.

Plus de 100 professeurs de partout au Canada et aux États-Unis ont assisté à la deuxième conférence nationale des enseignants en biosciences, une activité annuelle qui a eu lieu au Seneca College de Toronto les 28 et 29 février 2008. Les conférences telles que celle-là jouent un rôle-clé dans la mise à jour des connaissances des professeurs, qui les partagent ensuite en classe avec la prochaine génération de chefs de file en biotechnologie.

Organisée par le Canadian Biotechnology Education Resource Centre (CBERC), la conférence réunit les plus grands experts scientifiques et commerciaux, qui offrent des séances d’information sur la génomique, la bioéthique, l’agroalimentaire, le catalogage de la biodiversité, les programmes scientifiques au secondaire, la pollution de l’eau et les écosystèmes aquatiques, etc. Cette activité donne aussi aux enseignants une expérience pratique lors d’ateliers interactifs.

Two awards will be granted: $5,000 to an individual, and $10,000 to a not-for-profit organization award.

The nomination deadline is Friday, August 15, 2008 and the award will be

Biofuel technology will continue to play a key role in a diversified energy future. Canada is rich in advanced technologies and diverse feedstocks that can be leveraged for biofuels production, positioning this country as a leader in the bio-economy. Tomorrow’s biofuels, however, will be derived from a diverse set of feedstocks and the processes by which they are made will also produce valuable co-products such as bio-chemicals.

The evolution of biofuels technology Biofuels are not a single technology, but rather a family of related applications that contribute to a sustainable energy future. Like any new technology, biofuels continue to advance through several cycles of development—introducing new manufacturing processes, efficiencies and economic opportunities with each new discovery.

Canadian companies are contributing to the advancement of biofuels along the innovation value chain.

Canadians are familiar with traditional biofuels derived from grain-based feedstocks including corn and grains-based ethanol, soya and canola-based biodiesel. These biofuels are noticeably and technically different from their fossil fuel counterparts

Comme toute nouvelle technologie, celle des biocarburants continue de passer par plusieurs cycles de développement, et chaque découverte amène de nouveaux procédés de fabrication, une nouvelle rentabilité et de nouvelles possibilités. Des entreprises canadiennes contribuent à l’évolution des biocarburants tout au long de la chaîne de valeur de l’innovation.

Les Canadiens connaissent bien les biocarburants traditionnels issus de matières premières céréalières, tels que l’éthanol à base de maïs ou d’autres céréales et le biodiesel à base de soya ou de canola. Ces biocarburants sont nettement différents, entre autres du point de vue technique, de leurs équivalents fossiles, et on les utilise couramment dans des mélanges avec de l’essence et des carburants diesel traditionnels.

Cellulosic biofuels make use of residual (waste) feedstocks from the agricultural and/or forestry sectors and from post-consumer sources that would otherwise go unused.

Dedicated biofuel crops such as Miscanthus and hybrid poplar are being developed as biomass sources for cellulosic biofuels. These fuels are also blended with gasoline for transportation fuel and will be key to meeting renewable fuel targets for the transportation sector well into the future.

Green hydrocarbons are also derived from non-food based residual feedstocks and are designed to be indistinguishable from their fossil fuel counterparts. Technologies such as pyrolysis and the genetic modification of algae, bacteria and plants to produce new biofuel precursors are currently being commercialized in Canada. This third generation of biofuels will fit seamlessly into the existing fuel refining and transportation systems and will be used in specialized applications such as jet fuel, where the fuel source must be identical to traditional hydrocarbon fuels.

Canadian leadership in biofuels innovation Performance Plants of Kingston, Ontario uses biotechnology to develop new drought and heat tolerant varieties of corn, soybean, rice and canola crops that will allow farmers to reap greater yield per acre, meaning more food and more biofuel.

New genes to boost biomass yield and quality in dedicated cellulosic biofuel crops are also in their development pipeline with the goal of ensuring that both food and fuel needs can be met.

Agrisoma of Burnaby, British Columbia is also working on green hydrocarbon biofuels in oilseed crops. The company is focused on the development of dedicated biofuel crop varieties through its patented Engineered Trait Loci technology to efficiently re-program the synthesis and accumulation of plant oils ideally suited for energy or bio-based manufacturing. These oils will be capable of being used to directly displace traditional petroleum feedstocks.

Lignol Energy Corporation, based in Burnaby, British Columbia is constructing biorefineries for the production of fuel-grade ethanol and biochemicals predominantly based on

biomass from Canadian forests. Using an enzyme-based technology originally commercialized in the Canadian pulp industry, Lignol has produced ethanol and other high-value bio-chemicals from hardwood and softwood forest species.

The cellulose-to-ethanol process provides new ways to meet global demands for fuel, and further lessons demands on traditional grain crops to meet our biofuel needs.

Ensyn Technologies of Renfrew, Ontario has developed the world’s only fast pyrolysis technology that has operated on a long-term commercial basis since 1989 to convert residual solid biomass into liquid bio-oil. Bio-oil as it is currently deployed is a type of “second generation” biofuel used in thermal energy applications. Bio-oil produced through pyrolysis is emerging as a key ingredient in the production of the next wave of biofuels, which will be indistinguishable from their fossil counterparts.

Stimulating biofuel development Not only is Canada leading the world in unique innovations, but the country is also creating a supportive business environment for green technology development. Canada’s federal government established a goal of reaching an average of 5 percent renewable content in gasoline by 2010 and 2 percent renewable content in diesel fuel and heating oil by 2012 in an effort to made greener fuels for our future a reality.

Agriculture and Agri-Food Canada launched the ecoAgriculture Biofuels Capital Initiative (ecoABC), is a $200 million, four-year program ending in 2011 that provides repayable contributions for the construction or expansion of transportation biofuel production facilities. Funding is provided for projects that use agricultural feedstocks to produce biofuels and that have new agricultural producer equity investments in the projects equal to, at minimum, 5 percent of the total eligible project costs.

Private Health Care Opportunities Grow in Canada

Provincial government decisions about which health services (defined broadly as physician services, technology, procedures, and pharmaceuticals) to insure are complex determinations that consider medical necessity, the public policy complexities of balancing unlimited demand and scarce funding, and the sustainability of the public health care system. As a result, a number of medically necessary health services are not insured. There are also some health services that may be insured for one patient but not for another, depending on the patient’s particular circum-

stances, e.g., drug-eluting stents (diabetic vs. non diabetic) or live liver transplants.

There is a growing gap between health services that are insured by the provincial health insurance plans and uninsured health services that, although perhaps more expensive, provide better clinical outcomes. Examples include endovascular surgery, intravenous cancer drugs, bone morphogenic proteins, MRI guided focused ultrasound ablation, natural orifice transluminal endoscopic surgery (NOTES) and PET scans or other “enhanced”

Having an idea is one thing, but as all researchers know, even the best idea isn’t worth much without the time to see it through.

Just ask Canada Research Chair Dr. Robert Hancock (University of British Columbia). His big idea was to harness one of our body’s natural defenses—the inflammation response— and turn it into a powerful therapeutic tool capable of stopping infections that regularly defeat the most powerful antibiotics. Given the lack of new broad-spectrum antibiotics since the 1960s, Hancock’s research was timely.

But to reach his ambitious goal, he’d need to log countless working hours.

In 2000, the federal government introduced the Canada Research Chairs Program to drive Canadian research and development excellence by creating world-class centres of research in Canadian

Today, there are close to 2,000 Canada Research Chairs (CRCs), making the program one of the largest of its kind in the world.

The results of this $300-million-a-year program have been impressive. In Hancock’s case, he was able to learn how to selectively stimulate the body’s bacteria-fighting system, the same system that protects us from the thousands of bacteria we are exposed to daily. By supercharging our immune system without triggering harmful side effects such as septic shock, Hancock’s

research responded to the problem of antibiotic-resistant bacteria and the thousands of deaths that result from it.

The research also offered new hope to the millions of people who depend on a shrinking number of effective antibiotics during surgery and other medical procedures. Along the way, Hancock also generated important spin-offs for the biotechnology industry, including adapting a revolutionary array of technology to allow researchers to create unprecedented numbers of peptides.

Hancock is joined by many other CRCs working in biomedical research across the country. They know the most pressing medical challenges of our time may only be solved when we understand how to treat conditions at the deepest cellular, molecular and genetic levels.

“ CRCs within the same hosting university complement each other because each is selected based on the university’s overall research priorities as outlined in its strategic research plan.”

Dr. Samuel Benchimol at York University in Toronto, for example, is now deep into one of the most thorough studies of p53, the so-called tumour-suppressing gene. He’s working to understand why this gene functions perfectly in some people, but not at all in others. Meanwhile, the University of Waterloo’s Dr. Marianna Foldvari is merging biotechnology with nanotechnology as the CRC in Bionanotechnology, creating tiny robots capable of delivering therapeutic agents, including genetic therapies, with perfect accuracy and without degradation.

As well, Dr. Eric Cohen, the CRC in Human Retrovirology at the Université de Montréal, is working with leading pharmaceutical vaccine manufacturers to gain new understanding of how HIV-1 depletes T-cells.

Of course, it’s not just biomedical research that is advancing.

CRC research is ongoing in a wide range of biotech fields.

Examples include McGill University’s Dr. Lyle G. Whyte and his research in the Canadian Arctic on cold-adapted bacteria that break down toxic pollutants; and McMaster University’s Dr. Robert H. What Canadians Need to Know about SubsequentEntry Biologics

by Cate McCready and Dr. Philip Schwab, BIOTECanada Biotechnology research has opened up promising new worlds of opportunity in the treatment of human disease.

Biological therapies offer new hope to Canadian patients to treat diseases such as diabetes, rheumatoid arthritis, anaemia, multiple sclerosis and cancer that were once beyond the scope of medical science and traditional chemical medicines. In addition to the 150 biotech drugs and vaccines already available, more than 350 new therapies are currently undergoing tests with the goal of offering patients new treatment options.

As patents expire on some of the earliest biological medicines, copies of these medicines known as Subsequent-Entry Biologics (SEBs) or “biosimilars” are being developed. Canadian authorities are beginning to consider how to regulate these products, and Canadians need to understand the complexities behind the development of innovative biologics and SEBs. Unlike traditional chemical medicines or “drugs,” biological medicines are complex proteins produced using living cells or organisms under carefully developed proprietary manufacturing processes.

The protein molecules of a biological medicine are much larger, have more complicated structures, and are much more diverse than the small molecules that make up drugs derived from traditional chemistry.

Sometimes, very small process changes, such as slight variations in temperature, cell culture conditions or transport and storage conditions, can result in significant differences in the efficacy and safety of a biological medicine. Even long-established biotech companies making slight changes to the manufacture of their own medicines have experienced challenges in reproducing the exact manufacturing processes.

Recreating the same biological medicine is an even greater challenge and risk for a subsequent-entry manufacturer.

Biological therapies cannot be exactly replicated; therefore, scientists and regulatory bodies have concluded that

Because of the complexity of these products and their manufacturers’ commitment to the safety of patients, biological medicines undergo numerous tests before receiving market approval— many more, in fact, than chemical drugs. Consider that a typical manufacturing process for a chemical medicine consists of 40 to 50 clinical tests, while a biological medicine typically undergoes 250 tests or more to ensure a new biologic medicine is effective in treating disease and safe for patients to use. An SEB product must also meet the same level of efficacy and safety as the original product through substantial pre-clinical and clinical trials.

Following substantial public consultation, the European Union established a policy framework for the approval of biosimilar therapies. The European Medicines Evaluation Agency (EMEA) considers applications for biosimilar approvals based on the similarity to an existing marketed biological medicine and carefully developed clinical trial requirements for specific product classes. The European Union also ensures incentives for continued innovation in biotechnology through a policy of 10 years of data protection for innovative medicines.

The United States has begun a legislative process aimed at achieving a comprehensive policy to set the rules for biosimilar products entering the U.S. health system. Throughout this process, legislators in Congress and officials at the Food and Drug Administration have recognized the need to protect and

Media coverage has engendered a public perception that drug prices are always significantly lower in Canada than in the U.S., but this is not always the case, especially for newer, innovative products. Créé en 1985, le programme RS&DE a pour but d’offrir des incitations financières aux entreprises qui font de la R.D. au Canada sous la forme de crédits d’impôt pouvant être utilisés pour alimenter leurs filières de développement. Le budget 2008 du gouvernement apporte de précieux changements à ce programme, changements qui constituent une reconnaissance de la dynamique des découvertes et du développement du 21e siècle. Les voici :

• U ne augmentation de 50 pour cent du plafond de dépenses donnant droit à des crédits d’impôt remboursables, de 2 millions de dollars à 3 millions de dollars. Le pourcentage remboursable étant de 35 pour cent, les entreprises admissibles disposeront de 350 000 de dollars supplémentaires.

De plus, le remboursement total potentiel s’élève maintenant à plus d’un million.

• U ne extension des crédits d’impôt à l’investissement du programme RS&DE à certaines activités tenues à l’extérieur du Canada dans les situations où « un exécutant de travaux de RS&DE [a] à mener une partie de ses activités de RS&DE à l’étranger, à l’appui des travaux qu’il exécute au Canada ».

Les dépenses admissibles seront de 10 pour cent des frais engagés pour la recherche internationale.

• U ne augmentation du plafond de la fourchette d’élimination progressive du capital imposable qui rend plus d’entreprises

As co-founder and former CEO of a Vancouver-based company, Neuromed Pharmaceuticals, Natalie Dakers took part in building a successful company from idea to clinical stages and raising over $70 million in financing. She also headed the life sciences section of the UBC University Industry Liaison Office and was involved in the creation of several spinoff companies during a very high-growth period of the biotechnology industry in British Columbia.

L’industrie appuie également ce modèle. Au début de l’année 2008, le géant pharmaceutique Pfizer a octroyé un million de dollars au CDRD afin qu’il crée un fonds d’innovation qui servira à offrir des subventions de recherche afin d’accélérer la mise au point des composés thérapeutiques prometteurs. Pfizer considère le modèle du CDRD essentiel pour faire progresser les occasions de commercialisation et elle reconnaît qu’un partenariat avec cet organisme pourrait aussi accroître son champ de d écouvertes.

Le CDRD est le seul organisme du genre au Canada. Son unique objectif consiste à combler l’écart qui existe entre les découvertes prometteuses et les produits commerciaux de grande valeur qui constituent la base des entreprises dérivées, des homologations ou des nouveaux composés utilisés par les compagnies pharmaceutiques. Un bassin d’expertise de même que l’accès à des plateformes et à des technologies de commercialisation clés devraient considérablement améliorer la situation de l’industrie, pour faire du Canada un chef de file de la bioéconomie.

En sa qualité de cofondatrice et d’ancienne directrice de Neuromed Pharmaceuticals, une entreprise de Vancouver, Mme Natalie Dakers a collaboré à l’établissement d’une entreprise prospère en partant d’une idée pour l’apporter aux étapes cliniques, de même qu’à l’obtention de plus de 70 millions de dollars de financement. Elle a en outre dirigé la section des sciences de la vie du bureau de liaison avec l’industrie de l’University of British Columbia et elle a participé à la création de plusieurs entreprises dérivées au cours d’une période de très grande croissance du secteur de la biotechnologie en Colombie-Britannique.
