Friends, Today is the very essence of a bittersweet day for our family... In 2007, October 28th was the day we earned the titles “Mommy” and “Daddy” . . . it was also the day, just six years later, we learned the little girl that gave us those titles would likely not make it to seven. She didn’t. And today, four years after her diagnosis, we are here to celebrate her life and fight against the monster that stole her from us. We were gifted so much hope and promise when she was placed in our arms . . . and then given none with her cancer diagnosis. Tonight, we fight to change that. We fight for research that will change the terminal prognosis of DIPG. And tonight, we will have fun doing it! Jennifer’s Story Jennifer (JLK) was a beautiful 6 year old who was born with glitter in her veins. She sparkled from the inside out and brought out the sparkle in everybody she met. Jennifer was diagnosed with DIPG on her 6th birthday. DIPG is a type of brain tumor that has no real treatment options and no cure. It is terminal upon diagnosis. The average life span for someone with this tumor is 9 months from diagnosis. Jennifer lived 3½ months. It was never a fair fight. Jennifer’s tumors were donated and a cell line has been created from them. Jennifer will be part of finding a cure and making it possible for other children to celebrate many, many more birthdays. Because her friends and family want to do their part, too, after Jennifer’s death, Libby and Tony founded Unravel Pediatric Cancer. Through Unravel, we’re spreading knowledge and raising funds. Because knowledge is like glitter… it sticks with you and multiplies, and once we know better, we can do better.


About Unravel Pediatric Cancer Unravel is a nonprofit organization working to spread knowledge about the grim realities of pediatric cancer and the devastating impact of a lack of funding. We supply the information, tools and support to individuals and communities that want to help create change – because good people want to do good things but sometimes they need help getting started. In addition, Unravel partners with other nonprofits to strengthen our efforts. We do all of this with one goal in mind: we will unravel pediatric cancer. Tonight, at our first annual Glitter Gala we invite you to join Jennifer’s friends and family and become members of the Unravel family. All monies raised by and for Unravel will go directly to the fight against pediatric cancer. And, because of the generosity of a private donor, every dollar raised this evening will be matched up to $250,000! Dr. Michelle Monje During Jennifer’s illness, Libby and Tony Kranz heard about the work of Stanford pediatric neurooncologist Michelle Monje, MD, PhD, who studies donated DIPG tumor tissue to understand how its biology might be targeted with new treatments. In their final appointment at Lucile Packard Children’s Hospital Stanford, the Kranzes asked if they could donate their daughter’s tumor for this research after her death. Libby asked Dr. Monje to try to figure out how Jennifer’s tumor had spread so fast. Dr. Monje’s lab is taking a multiple-pronged approach to exploring new DIPG treatments, including studying the molecular and cellular vulnerabilities of DIPG cells, determining the mechanisms by which normal cells become cancerous, and finding ways to lessen the effects of cancer treatment on normal cells. Dr. Monje and her colleagues are in the process of publishing a scientific paper that provides an important piece of the puzzle and, she says, Jennifer’s tumor tissue was “the key tool to figuring this all out.”

The new paper, appearing in Cell, asks why highgrade gliomas — cancers like DIPG and glioblastoma — often metastasize to a specific part of the brain. As Jennifer’s did, these tumors tend to spread to a brain region called the lateral ventricle subventricular zone, an area where new nerve cells can grow throughout life.

The researchers’ work revealed that some of the molecular signals promoting neural growth also attract tumor cells. A protein called pleiotrophin — secreted by neural precursor cells in the subventricular zone — was especially important in this regard. Interrupting pleiotrophin signaling dramatically reduced the spread of high-grade gliomas in mice, the researchers found. Researchers have already begun developing drugs that inhibit the actions of pleiotrophin. Unfortunately, these don’t yet penetrate the brain well enough to treat high-grade gliomas. But Dr. Monje’s team showed that, in principle, such drugs should make a significant dent in gliomas’ ability to grow and spread. This is why, at Unravel, we feel it’s critical that scientists are able to make quick decisions regarding the direction of their research projects without seeking approval mid-project from their funding partners. We think that scientists’ time is much better spent doing research than writing grant proposals. For these reasons, we do not fund research using the traditional grant method – we provide funding that is generally not restricted to a specific purpose. In this way, we can allow our beneficiaries the flexibility and agility to quickly follow up on exciting new discoveries.

Mama’s Night Out The best thing about Mama’s Night Out is that it’s an amazing event for an amazing cause. The worst thing about Mama’s Night Out is that there is a cause. Women across the country take breaks from their lives all the time. They go out to eat, shop, dance, and just have some quality girl time.
