The Skin’s Microbiome

O

ver the last several years, there has been increasing interest in better understanding the skin’s microbiome.

Essentially, the microbiota of skin encompasses the microbial species that inhabit the skin, helping it to ward off foreign pathogens. A number of key activities occur in the microbiome of skin including biofilm formation and quorum sensing. The microbiome of skin serves a vital function in the skin’s innate immune system.

Researchers in academia and industry alike have sought to determine the composition and behavior of the skin’s microbiome. A great deal of research investigating the skin’s microbiome is underway, including efforts to understand its fundamental nature, the effect of topical treatments, and the influence of environmental factors.

The NYSCC will host a symposium on the skin microbiome on Tuesday, June 4th at the Chart House

in Weehawken, New Jersey. Some of the topics to be discussed include topical product development and its effect on the skin’s microbiome; the antimicrobial properties of skin lipids and their influence on the microbiome; the microbiome of normal, dry, and sensitive skin; the beneficial and detrimental roles of certain bacterial species in skin health; the role of the skin’s microbiome in odor biochemistry of the axillae region; and the development of new products aimed at improving the microbiome. Join us for an intellectually stimulating day to learn more about this increasingly important subject.

hope you are enjoying the beautiful blue skies and sunshine that we have been so fortunate to have—finally! With Suppliers’ Day having been a success, we can now start focusing on upcoming summer programs, which are plentiful!

But first, a brief recap of Suppliers’ Day: the expanded show floor was brimming with the latest technologies and new exhibitors. As Happi reported, “Records were broken!” Nearly 540 exhibiting companies occupied 100,000 net square feet of expo space; 107 of these companies were new to the event. Attendance topped last year with 9,981 registered participants! A 40th retrospective and focus on the “evolution of beauty” was the overarching theme. The innovation born of the evolution was evident with the highly successful Future Chemists Workshop showcasing all that’s new to current students, while the newly launched Mentor Program successfully debuted and engaged dozens of mentors and mentees. The Innovation Hub & Theater showcased current exhibitors debuting new products to the North American market, and CEW Supplier Award finalists demonstrated why their product made the cut!

An extensive survey has been sent to both exhibitors and attendees; we will share these results in our next edition of Cosmetiscope and urge you to participate in the survey so that we can continue to meet your needs.

Although the show officially closed on May 8th, activities continued thereafter: ICMAD’s FDA Regulatory Conference took place at the Jacob K. Javits Convention Center on Thursday, May 9th, and the CEW BEAUTY AWARDS marked the official end of the festivities, presenting the Supplier Award: Ingredients & Formulation and celebrating the winner: KOBO Products, Inc. Giorgio Dell’Acqua, Chair of our Scientific Advisory Committee, then jetted off to Brazil for FCE Cosmetique to present on sustainability and, in particular, on recycling food waste for use in cosmetic active ingredients. Giorgio acted as an NYSCC-Suppliers’ Day representative. This is just one example of the types of partnerships formed through Suppliers’ Day. We will also be participating in Cosmoprof in Las Vegas together with ICMAD, as well as at SCC bellwethers: Florida Sunscreen Symposium in Orlando and California Suppliers’ Day in Long Beach to name a few. So, while things are finalized for 2019, we are already looking at improvements, program expansion, and more for the 2020 New York City expo!

What should be next on your calendar? Our Microbiome Symposium, taking place at The Chart House in Weehawken on Tuesday, June 4th. This full-day program will center on the role of the microbiome in cosmetic/personal care product development and will feature expert-led discussions of tactics, tools, and ingredients that are necessary as this becomes integrated into formulations.

The highlight of our social events will take place on Friday, July 19th: the annual NYSCC Golf Outing will be held at the beautiful Crystal Springs Golf Course in Hamburg, NJ. Hopefully, the sun will be shining this year so we can fully enjoy the venue!

Skin Microbiome and Topical Product Development

t has long been acknowledged that the skin serves as a key organ of immunity beyond a physical shield and that its communication with the environment plays a role in overall human health, mood, and the ability to cope with stress. Elements of the skin’s innate immunity shape the nature of the body-acquired humoral immunity based on environmental signals collected from birth onward.

The idea that microorganisms have a profound impact on our well-being is rapidly becoming more widely acknowledged. The scientific and medical communities are working to study these effects, yet the practical means of implementation as well as regulatory landscape are evolving. The skin microbiome, the population of resident and transient microorganisms of our skin, represents a growing area of research and interest. The identification, classification, mapping, study of diversity, environmental implications, and inferences to skin condition are key areas of investigation. Various molecular and in silico methods have been developed and employed in this area of research, but paths to utilizing this knowledge in skin care product development are limited. This presentation provides a general overview of the exciting state of research and development in this field. n Nava

Dayan, Ph.D.

Dr. Nava Dayan has nearly 30 years of experience in the skin care segment, and has produced more than 150 publication credits in numerous, widely respected industry journals and four books. She has been awarded the In-Cosmetics Gold Award for innovation and commensurate recognition from the NYSCC and the Controlled Release Society for excellence. Dr. Dayan is the owner of Dr. Nava Dayan LLC, a skin science and research consultancy serving the pharmaceutical, cosmetic, and personal care industries. The firm focuses on product feasibility in skin care; composition of R&D plans covering efficacy and toxicology; planning, executing, and interpreting data into claims, formulations, delivery for improved efficacy, and attenuated toxicity. Areas of expertise include drug-skin interactions, bio-markers (genomics and proteomics), skin-/age-related sensitivities, inflammatory skin disorders, innate immunity, and biota. Dr. Dayan is the co-editor of the book Innate Immune System of Skin and Oral Mucosa: Properties and Impact in Pharmaceutics, Cosmetics, and Personal Care Products published by John Wiley & Sons. Antimicrobial Lipids at the Skin Surface—Implications for the Microbiome – Philip W. Wertz (University of Iowa)

O

ne of the factors that determines which microorganisms can live on the human skin is the presence of certain lipids at the skin surface. It has been recognized since the 1940s that lipids from the skin surface are antibacterial and contribute to the innate immunity of the skin; however, this area has not received much attention until recent years. Antimicrobial fatty acids are derived from sebaceous triglycerides through the action of microbial lipases. Two of the major contributors of lipase for this hydrolysis are Cutibacterium acnes (formerly Propionibacterium acnes) and Masassezia spp (formerly Pityrosporum spp)—both can use sebaceous fatty acids as a carbon source. The sebaceous fatty acids include saturated species ranging from 7 to 22 carbons in length and some monounsaturated species. The major monounsaturates have been identified as C16:1Δ6 (sapienic acid) and C18:1Δ8. It turns out that the short, odd-carbon saturated fatty acids (C7:0, C9:0, C11:0, and C13:0) have antifungal activity. Lauric acid (C12:0) and sapienic acid use these potent antibacterial properties to fight against a range of Gram-negative and Gram-positive bacteria, including Staphylococcus aureus. Some of the other sebaceous fatty acids have bacteriostatic activity.

The other class of antimicrobial lipids at the skin surface are free long-chain bases including sphingosine, (Continued on page 6)

dihydrosphingosine, and 6-hydroxysphingosine. These are liberated from both free and covalently bound ceramides by epidermal ceramidases. The long-chain bases are also active against a range of Gram-negative and Gram-positive bacteria, including Staphylococcus aureus. Under circumstances where production of these innate antimicrobial lipids is reduced, opportunistic pathogens may colonize the skin and potentially produce infections. For example, in atopic dermatitis, it has been shown that levels of sphingosine and sapienic acid are both below normal, and, under these circumstances, the skin becomes colonized by Staphylococcus aureus. n Philip W. Ph.D.

Dr. Wertz holds the title of Professor Emeritus in the Department of Oral Pathology, Radiology, and Medicine at the University of Iowa and the Dows Institute. He earned his A.B. in Biochemistry at Rutgers University (1971) and received a Ph.D. in Biochemistry at the University of Wisconsin (1976). His thesis research focused on chemical kinetics, catalysis, and computer simulation. After post-doctoral training at McArdle Laboratory for Cancer Research, Dr. Wertz held the position of Research Scientist at Marshall Dermatology Research Laboratories at the University of Iowa.

In 1990, he was appointed Associate Professor at the University of Iowa College of Dentistry. Dr. Wertz’s research focuses on lipids and barrier function in skin and oral mucosa. He has been a member of the Society for Investigative Dermatology, American Oil Chemists Society, American Society for Biochemistry and Molecular Biology, American Association for Dental Research, American Association of Pharmaceutical Scientists, and the Society of Cosmetic Chemists. Dr. Wertz is currently a member of the editorial boards of International Journal of Cosmetic Science, Skin Pharmacology and Physiology, and Journal of Lipids. _____________________________________________

A New Approach to Better Understand Specific Skin Conditions

O

ver the last several decades, huge progress has been made in understanding the essential role of the skin barrier in maintaining healthy skin. Recently, further knowledge has been acquired— particularly after the start of the Human Microbiome Project in 2008—about millions of microorganisms that collectively exist on our skin, which is referred to as the human skin microbiome. This microbial community exists in a state of permeant exchange with its host (i.e., the skin). Although many functional details are still inadequately understood, these interactions make the microbiota an interesting target for cosmetic research and for new applications. This presentation provides food for thought on how these interactions are associated with various skin conditions such as normal, dry, and sensitive skin. Our clinical data demonstrate how topically applied ingredients can modulate these interactions. n Mathias

Dr. Gempeler holds a Ph.D. in Pharmacy and has also undertaken post-graduate studies in marketing. Until July 2017, he was employed as Head of Global Marketing for Skin Care at DSM and was responsible for creating and executing the company’s global growth strategy. The Jeckyll and Hyde Skin Microbiome: Good Bugs Behaving Badly

n 1886, two years after the Scottish surgeon Sir Alexander Ogston discovered Staphylococcus aureus in the pus of surgical abscises, fellow Scotsman Robert Louis Stevenson published his famous gothic novel, The Strange Case of Dr. Jekyll and Mr. Hyde. The well-respected and community-minded Dr. Henry Jekyll fought with his evil alter ego Mr. Edward Hyde. Like Jekyll and Hyde, we know that our microbiome

can also take on dual personalities. A so-called “good” skin bug, such as the universally commensal Staphylococcus epidermidis, can also be an opportunistic pathogen and cause serious, life-threatening infections.

Conversely, a “bad” skin bug such as Staphylococcus aureus—noted for its role in impetigo, boils, and atopic dermatitis—colonizes 30% of the U.S. population, most often without issue. Exactly what causes “Jekyll bacteria” to transform into “Hyde bacteria” is multifactorial, with strain, host immunity, environment, and other factors at play. The transformation involves quorum sensing, biofilm formation, and a plethora of metabolic changes that ultimately results in acute and chronic skin inflammation. Preventing, inhibiting, or even reversing the pathways involved in this transformation are attractive new therapeutic strategies. This talk will examine this fascinating area of bacterial biology as it relates to the skin microbiome, furthermore discussing how all skin and personal care products should help keep our “Dr. Jekyll” bacteria from becoming “Mr. Hyde.”

Hillebrand, Ph.D.

Dr. Greg Hillebrand received a B.S. in Biochemistry from Michigan State University and a Ph.D. in Biochemistry from Baylor College of Medicine. He completed his postdoctoral training at the University of Illinois. Greg is currently Senior Principal Research Scientist at Amway Global Discovery, where he helps lead new technology for the Beauty and Nutritional categories. Prior to joining Amway, Greg was Principal Scientist at Procter & Gamble for 27 years. He has a broad expertise in skin and hair science, especially skin health, aging, photo-aging, epidemiology, biophysical and imaging clinical methods, and product performance testing. He has over 40 publications and 10 patents, and several of his inventions have been successfully commercialized, most notably the technology platform for the VISIA Complexion Analysis System by Canfield Scientific. He lived and worked in Japan for 7 years on assignment for Procter & Gamble, gaining direct experience and insight into Asian consumer needs and the Asian beauty market. Greg is a member of the International Society for Biophysics and Imaging of the Skin and Society for Investigative Dermatology, and is a member and past president of the Society of Cosmetic Chemists. On the side, Greg is a decent jazz musician on the alto and soprano sax, but not yet good enough to leave his day job. _____________________________________________

Skin Microbiome Exploration for Active Ingredients Innovation – Denis Wahler (Givaudan)

T

he exploration of the skin’s microbiome is an extraordinary journey. The many different environments imposed on the skin—humidity, temperature, and solar radiation exposure—as well as the density of hair follicles and sebaceous glands, essentially contribute to its characteristic patchwork network of complex eco-systems. Anatomical location—scalp, toes, face, arms, axillae, etc.—also contributes to these various ecological niches.

Givaudan Active Beauty’s explorers toolbox is made up of classical microbiology and bacteriology methods, metagenome sequencing techniques, and biogenetic technologies. The use of state-of-the-art equipment, such as Illumina, PacBio, and Nanopore sequencers, is combined with a deep expertise in gene data treatment and mining, and fundamental knowledge of skin microbiota biology. Our experts team up with specialists in clinical studies, skin biology, and marketing, allowing them to discover new active ingredients.

This presentation will serve as a guide for the exploration of the skin microbiome on various body areas, using cutting-edge technologies and new tools to mine and interpret metagenomic information. The resulting data will then be compared with previous knowledge in the field gathered from different methodologies and techniques.

Denis graduated with a Ph.D. in Chemistry and Biochemistry from the University of Bern, Switzerland. He specialized in white biotechnology and green chemistry at PCAS Biosolution, where he served as Head of R&D and Scientific Manager for many years.

he human skin is colonized by a diverse microbial community, which differs considerably between various human subjects and body sites. Undoubtedly, the skin microorganisms fulfill several beneficial functions for their host. However, from a cosmetic point of view, they are also responsible for several negatively perceived skin phenomena, such as the formation of body odor, impure skin, or dandruff. For many years, it has been well known that the composition and activity of the microbial community of the human skin microbiota play a key role in these processes.

In the case of body odor, the formation of a complex mixture of several classes of compounds—among them steroids, carboxylic acids, and volatile sulfur compounds—causes malodor. In several cases, these smelly compounds are excreted onto the skin as water-soluble, non-volatile precursors from which the volatile parts are subsequently released by microbial enzyme activities. In this context, aminoacylase and cystathionine-ß-lyase probably represent the most well characterized malodor releasing microbial enzymes.

From the consumer’s point of view, human underarm perspiration and body odor formation are important topics. Hence, a better understanding of the composition and the activity of the human axillae microbiota is closely linked to a better understanding of the formation of human body odor.

In this presentation, an overview will be provided of the human skin microbiota and their enzyme activities, with special focus on (but not limited to) the armpit microbiota. Strategies and technologies to influence this microbial ecosystem and to better support the development of innovative and more effective cosmetic products will also be discussed. n Rainer

Simmering, Ph.D.

Dr. Rainer Simmering is a Senior Scientist in the Corporate Microbiology group of Henkel AG & Co. KGaA in Düsseldorf. After finishing his study of Biology at the WWU in Münster, he earned a Ph.D. at the German Institute of Human Nutrition in Potsdam, where he stayed an additional year to carry out postdoctoral research. In 2000, Rainer started at the Nestlé Research Center in Lausanne, where he worked on the detection and function of intestinal microbiota. In 2003, he joined the Hygiene Research group at Henkel, working on different research projects such as the elucidation of the skin microbiota with molecular means. Since 2009, Dr. Simmering has acted as an internal consultant at Henkel for the global beauty care business regarding all microbiological topics. _____________________________________________

R

ecent studies, backed by improving methods and tools with higher sensitivity, have unveiled the unexpected complexity of skin microflora. Consumer awareness and expectations have also dramatically increased. Cosmetic science has no choice but to integrate new data and knowledge on the skin microbiota ecosystem into product development.

To further explore the role of microbiota in a healthy skin microbiotic barrier, we established an interdisciplinary innovation platform on skin microbiota to: • Better understand the role of skin microorganisms in skin beauty.

• Study the complexity of the skin microflora and skin ecosystem to help design a set of more specific solutions adapted to each and every skin condition.

• Develop in vitro models and clinical protocols to monitor skin microflora activity.

In the coming years, and based on an ecosystemic approach, we intend to launch several active ingredients for cosmetic products that harness the innovation platform’s findings on skin microbiota. Taking into account the origin of skin disorders and microbiota dysbiosis, these ingredients will be capable of rebalancing skin microflora or taking advantage of it and thereby contributing to skin beauty.

T

he Cosmetiscope editorial committee invites all interested parties to submit feature technical articles for publication in the NYSCC monthly newsletter.

Authors of feature articles are eligible to win the prestigous NYSCC Literature Award ($1,000) for the best front-page article published during the calendar year.

Authors also receive $200 reimbursement to attend a theatrical performance of their choice. Writing an article for your peers is a very rewarding experience, both personally and professionally, and will ensure your place in NYSCC history. You may choose whatever topic you feel would be interesting to fellow colleagues in our industry.

We also welcome any other types of commentaries or articles that may be published in the Career Corner, Technical Tidbit section, or as a Letter to the Editor.

his is a one-day course in phytochemistry, which will cover the general chemistry of natural products from plants for skin health applications.

This course will be of great interest to cosmetic scientists, formulators, analytical scientists, chemists, and biochemists who want to enhance their knowledge and capabilities using botanical ingredients in product development.

It is assumed that the students attending this course will already have some basic knowledge of organic chemistry and biochemistry.

Course Outline

n Introduction to plant materials and their compounds • Overview of natural products • Acetogenins: flavonoids, lipids, lignans, and quinones • Carbohydrates: mono-, oligo-, and polysaccharides • Isoprenoids: carotenoids, steroids, and terpenoids • Nitrogen compounds: alkaloids, amino acids, nucleic acids, peptides, and proteins n World of polyphenolic/flavonoid compounds in nature • What are polyphenolics/flavonoids?
