TRANSFORMING LIFE SCIENCE AND CONSUMER PRODUCTS BY MIMICKING NATURE’S CELLULAR SCAFFOLD

BioSurfaces is a leader in producing solutions by engineering biology with synthetic cellular matrices using the company’s patented proprietary electrospinning technology (Bio-Spun™). Bio-Spun™ materials have a composition that closely resembles the body’s natural scaffold, which helps cells respond much more naturally than their response to other materials. For our in vitro research tools (IVRT) products, the Bio-Spun™ scaffold provide a more natural 3D environment for cell growth and tissue development without the use of animal products to create partial and full thickness models for skin, alveolar, retina, and more. Our Bio-Spun™ cell chamber provides a delivery vehicle for the customer’s target cells while protecting them from an immune response. The BioSpun™ materials comprising the cell chamber promotes tissue integration and prevents fibrosis/rejection. Lastly, Bio-Spun™ materials allow for easy incorporation of drugs and/or other bioactive agents directly into the nanofibers, demonstrating consistent drug release. Our Bio-Spun™ materials be made into a variety of shapes, including ones that are unusual and difficult-to-manufacture, with vastly improved biocompatibility and physical properties. Our technology has been validated by leading academic, government, medical device and life science organizations.

Gene Tools, LLC (Sharktooth - #7)

Like sequence-specific masking tape binding RNA, Morpholino oligos are versatile tools for controlling gene expression. Gene Tools LLC increasingly supplies pharma and biotech with custom-sequence Morpholinos, with over half of current sales to commercial firms. Have us help you design oligos and make your discovery research oligos. When it is time for preclinical toxicology and clinical trials, Ajinomoto Co. Inc. can make your GMP-grade oligos.

There are four US FDA-approved Morpholino drugs for Duchenne muscular dystrophy (eteplirsen, casimersen, golodirsen, and viltolarsen). The first three listed are from Sarepta Therapeutics Inc., which spun off our company Gene Tools. The last listed, viltolarsen, is from NS Pharma, US subsidiary of Nippon Shinyaku Co., Ltd.

Dr. Jim Summerton, Sarepta’s founder and co-inventor of Morpholinos, formed Gene Tools in 1997 to bring Morpholinos to the research market. In 2000 Gene Tools made its first big splash in developmental biology; the zebrafish and Xenopus research communities started microinjecting Morpholinos into eggs and early zygotes of their model organisms and could assess gene knockdown with less changes of off-target genes than with other available antisense structural types, specificity critical for work in embryos. Over 11,000 research papers report use of Morpholino oligos in animals, plants, protists, fungi, and bacteria.

Morpholinos bind to complementary sequences of RNA and block other things from binding there. Different kinds of RNA targets give different outcomes when blocked: Morpholinos can block translation, modify splicing, inhibit miRNA maturation & activity, block poly-A tailing, block RNA translocation, block RNA methylation, block ribozyme activity, etc.

AcouSort - Automated Cell Handling Rapid Staining and Washing of Precious Cells (South Lobby - #12)

Preparation of cell samples for flow cytometry typically involves staining with fluorescent antibodies. A centrifugation-based wash is often performed, requiring several manual handling steps. This process can dramatically decrease the recovery and viability of the sample and is especially ill-suited for samples with low cell numbers. The AcouTrap (AT) is an automated platform designed to streamline workflows for cell up-concentration, automated staining and washing of cells able to handle 10-500 000 cells with high recovery.

The AcouTrap technology and Cell handling applications

The AT uses acoustic waves to capture cells in a gentle and contact-free way. When cells are entering the AT system they are captured and held in place by the acoustic field levitating in the fluid. When kept in the AT cells can be washed, stained or exerted to other stimuli in a gentle and contact-free manner.

AcouTrap provides improved performance of cell labeling assays

Comparing the results for using the AT for automated staining AT provides:

• 3x faster processing compared to centrifugation-based protocol

• Assay automation with minimized manual handling steps

• Rapid surface marker staining with enhanced binding kinetics resulting in >90% staining efficiency already after 3 minutes

• Efficient washing reducing background fluorescence

• AT staining and wash cycle produces higher relative fluorescence intensity (RFI) than conventional methods

Using the AT for cell handling, binding and staining assays is a novel and unique tool with great potential to improve and streamline cell assays where the standard manual protocols are tedious, cell numbers are low or the cells are fragile to ensure optimal cell sample quality.

Arima Genomics (South Lobby - #3)

Arima Genomics is a leader in exploring 3D genome structure with our Hi-C technology. Arima Hi-C is a proximity ligation method that captures the organizational structure of chromatin in three dimensions, where genomic sequences that are distal to each other in linear distance can be closer to each other in the 3D space. The high-resolution, genome-wide map of interacting genetic loci that is generated from Hi-C data can then be used across multiple genomic applications, including identification of promoter-enhancer interactions for gene regulation studies, detection of structural rearrangements, and scaffolding contigs for genome assemblies to define chromosomes de novo. Using a 3D genomics approach with Arima Hi-C provides information on both the sequence + structure of the genome.

Sekisui Chemical Co.Ltd (FAES Terrace - #6)

Founded in 1947, SEKISUI CHEMICAL is a chemical manufacturer that provides products worldwide in industries such as housing materials, environmental infrastructure, electronics and mobility. We are currently expanding our presence in the life science domain by focusing on cell culture consumables.

We have developed a new state-of-art Chemically-Defined Scaffold (CDS) for iPSCs, ESCs and MSCs, with culturability similar to that of ECM.

Currently, ECM protein-based scaffolds are used for culturing pluripotent stem cells. But these scaffolds face major stability and variability issues and pose problems of safety against viral contamination. In order to overcome these challenges, we have developed a synthetic polymer based scaffold.

Our scaffold (CDS) is ready to use, eliminating the need for the tedious coating process. It is available as pre-coated 6-well plates, individually packaged and sterilized by gamma irradiation, providing much needed stability to the cell culture process. CDS is animal origin free (AOF), has minimal elution, and can be stored at room temperature (no refrigeration needed). CDS could also be easily adapted for large scale and automated culture equipment.

We are looking to collaborate with academic organizations and teams which focus on regenerative medicine research.
