Exploring the connection between humans and the natural world

Dear Reader

In the last issue, I wrote about the importance of laboratories in South Africa and their connection to conservation projects. I now want to follow on with one such project that I think you will find uplifting and quite beautiful.

The film Pangolin: Kulu’s Journey brought much-needed attention to an animal that is currently known as 'the most poached mammal on earth', when it premiered on Netflix on April 21st.

Directed by Academy Award-winner Pippa Ehrlich (the name behind South Africa’s first Netflix Original documentary, My Octopus Teacher), Pangolin: Kulu's Journey is a uniquely South African narrative of a man and a pangolin pup on a journey together into the wild. This account unfolds over a year and seven months, encapsulated into 90 minutes of mesmerising storytelling.

Filmed at the Lapalala Wilderness Reserve (https:// www.lapalala.com/), in the UNESCO-designated Waterberg Biosphere in Limpopo Province, South Africa, this film has moments of such surreal beauty that it could almost be mistaken for magical realism. This landscape sets the tone for our first glimpse of Kulu, a Temminck’s pangolin, bipedal and prehistoric-looking.

The unique order of Pholidota (comprising eight species of pangolins: four in Africa and four in Asia), evolved with dinosaurs 85 million years ago.

These animals are the totem of indigenous tribes, considered the ‘protector’ and ‘the bringer of rain’. Their scales have been used in traditional medicine cures for thousands of years, now over-harvested with the four Asian species listed as endangered and critically endangered on the IUCN red list.

Now, huge pressure is being brought to bear on the African species, with tons of African pangolin scales exported to Asia as wildlife contraband for use in patented traditional medicine, which may bring an end to all eight species within the next ten years.

In Kulu’s Journey, we are introduced to Gareth Thomas, a rebellious young South African man who leaves school early, lives on the proceeds of poker games, and rides motorbikes.

He has started searching for meaning and a return to a closer relationship with nature, which he remembers from his youth in Zimbabwe. By chance or destiny, he meets Ray Jansen, who is the founder of the African Pangolin Working Group – one of the first four organisations in the world with a singular focus on pangolin conservation, established in 2011 when the illegal trade in pangolins in South Africa was just starting to become a noticeable feature of the illegal wildlife trade. Gareth is with Ray when a 3kg young pangolin is rescued from the illegal trade in a sting operation in Gauteng Province, South Africa. This is the beginning of his journey with Kulu, and it is a privilege to witness.

“The young animal immediately proves himself to be a dynamic central character, and as he overcomes his trauma, we meet a spunky, playful, and stubborn pangolin who will steal your heart quicker than he downs a colony of ants,” writes Shaina Weatherhead (https://collider.com/pangolinkulus-journey-reviewnetflix/).

Pippa Ehrlich, environmental storyteller, conservation journalist, filmmaker, and a member of the Sea Change Project, continues to explore the themes of the relationship between humans and the living world, in Pangolin: Kulu’s Journey. She says, “The relationship between humans and animals is older than time itself. Of course, there are many versions of this story, but at the heart of things is a narrative of connection and interdependence.” Sea Change strives to tell stories that connect humans to the living world, motivating them to become part of the regeneration of our planet.

The African Pangolin Working Group is the organisation responsible for the rehabilitation and release of Kulu, a Temminck’s pangolin who was rescued from the illegal trade in South Africa.

Along with Sea Change Project and the African Pangolin Working Group, the Pangolin Crisis Fund (https:// pangolincrisisfund.org/), a US-based non-profit organisation, will be increasing global awareness around the film, with a goal to eliminate the demand, trafficking, and poaching crisis that puts all eight species of pangolins at risk of extinction. And so, the Pangolarium was built.

The Pangolarium (meaning ‘a place for pangolins') is a collaboration between the reserve, the African Pangolin Working Group (APWG) and Lepogo Lodges, which operates two not-for-profit safari lodges in the reserve.

The Pangolarium will give rescued pangolins the best chance at regaining health before being released back into the wild, which of course brings me full circle –emphasising the importance of laboratories in South Africa and their connection to conservation projects. If you haven’t already seen Pangolin: Kulu’s Journey, please watch it. You won’t be disappointed!

Based on the principle of fluorescence quenching. An immobilized Ptbased luminophore is excited by the light of a blue LED and emits a red light.

As oxygen interacts with the luminophore it reduces the intensity and lifetime of the luminescence.

The lifetime of the luminescence is measured by a photodetector and is used to calculate the dissolved oxygen concentration.

The probe is fitted with easy-to-use Smart Caps (HI764113-1), which lock in place and contain pre-loaded calibration coefficients that are automatically transmitted to the probe.

The Smart Cap features an immobilized O2 sensitive luminophore with a rugged insoluble black oxygen permeable protective layer.

Over time, the sensor’s optical components can age but are compensated for by using the reference signal to compensate for the measuring path.

As a result, the sensor provides accurate DO measurements over long periods of time without the need for frequent calibration.

ANALYSING MOISTURE, PROTEIN, AND FAT CONTENT IN FOOD

Three key parameters - moisture, protein and fat content - stand at the forefront of food analytical chemistry, providing crucial data that impacts everything from shelf stability to nutritional labelling and production economics. Determination

of moisture

The moisture content of raw materials, such as grains or finished products, can be an indication of their storage stability. Moreover, moisture is one of the parameters that can generate the highest production margins. If there is a limit on the amount of moisture in a product, it is profitable to keep the water content as close to this limit as possible without crossing it. The water content is traditionally measured with moisture balances or titration. Moisture content is also one of the prime examples for NIR analyses, especially for fast and reliable at- and on-line measurements.

of protein content

Proteins are essential nutrients for both humans and animals and an important quality parameter in food production. The primary reference method for measuring protein content of foods is the Kjeldahl method. In this method, protein is broken down to ammonia, the nitrogen content is measured, and this value is multiplied by a specific factor to obtain the protein content. Besides the Kjeldahl method, NIR is a well-established technology to determine nitrogen/protein.

Determination of fat content

Fat is an important parameter in food products,

such as oilseeds, meat, dairy, and others. Fats have a high energy content, play a vital role as transporters of flavour, and substantially contribute to the mouthfeel of food. Soxhlet extraction is the reference method for the measurement of free or total fat and is preceded by an acid hydrolysis step. The fat is then extracted with an organic solvent. Other methods for fat determination include NIR, NMR, or specific extraction methods depending on the sample type (e.g. Mojonnier for dairy).

CHEMICAL ANALYSES FOR THE PRODUCTION OF LITHIUM-ION BATTERIES

Replacing traditional fuel-powered vehicles with battery-powered options is essential to reduce carbon dioxide (CO2) emissions. This greenhouse gas is a result of fossil fuel combustion - limiting its input into the atmosphere will also influence its effects on global warming. The production of batterypowered electric vehicles (EVs) continues to rise as more governments plan to prohibit the use of combustion engines in the future, and automobile manufacturers commit to the production phase-out of combustion engines. The International Energy Agency predicts that by 2030, 60% of all new car sales will be EVs. At the same time, renewable energy sources like wind and solar require storage capacities for electricity. Batteries are currently the most scalable materials in which to store excess electricity, and this market has been growing steadily as countries begin to invest in energy storage solutions.

Lithium-ion batteries (Li-ion batteries) are the most common rechargeable energy storage options available today. Production of Li-ion batteries needs to follow stringent quality standards. The water content, residual alkali content, or ionic impurities can have a negative impact on the safety and storage capacity of the final battery. Meanwhile, the composition of cathode materials or

electrolytes can influence manufacturing costs and performance qualities of Li-ion batteries. This White Paper elaborates how titration and ion chromatography can be used to monitor various quality parameters during lithium-ion battery production.

• Traces of water can negatively impact the electrochemical performance of lithium-ion batteries, lead to the formation of toxic HF, and change the residual alkali content. Coulometric Karl Fischer titration is ideal for determining water content at trace levels in various Li-ion battery materials and components.

• Residual alkali (surface bases) can form when cathode materials are exposed to ambient

SCHOOLS AND NEIGHBOURHOODS ARE THRIVING THANKS TO A UNIQUE DRIVE TO MAKE RECYCLING A WAY OF LIFE

A drive to persuade more South Africans to incorporate recycling into their daily routinesrather than only for observances like Global Recycling Day or Clean-up and Recycle Weekis benefiting schools and neighbourhoods.

Helping drive these positive changes is the country’s longest-standing producer responsibility organisation, Petco, which was established in 2004 with the aim of ensuring post-consumer packaging is recycled and kept out of landfills and the environment.

In 2024, Petco’s work on behalf of its members - brand owners, retailers and packaging importers - equated to over 76 000 cubic metres of landfill space saved, which is equivalent to 30 Olympic-sized swimming pools.

One of Petco’s unique partnerships to help drive recycling at schools and in neighbourhoods involves the African Reclaimers Organisation (ARO).

ARO’s objectives include advocating for the recognition and inclusion of reclaimers as essential contributors to South Africa’s recycling economy. By supporting fair working conditions, ARO aims to provide reclaimers with access to vital resources, promoting their integration into the formal waste value chain, and improving their livelihoods, dignity, and economic prospects.

Together, the organisations celebrate two key projects in Johannesburg, which demonstrate a growing appetite by the public to recycle more - Craighall Primary School and the Bordeaux South Residents’ Association.

A recent video taken at Craighall Primary shows how programmes - run by ARO with

recycling equipment support from Petcoinclude educating pupils about how to separate their recyclable materials from their general waste at home. According to the organisations, an interesting consequence of the campaign is that the pupils are, in turn, putting pressure on their parents to do the same.

“Educating our youth into a more sustainable mindset is really impactful because they haven’t yet developed the bad habits of adulthood, and they have the power to influence recycling behaviours in their own homes and families,” says Petco CEO Telly Chauke.

“On the collections side of things, informal waste reclaimers play a really important role, so Petco is pleased to be working with organisations like ARO to legitimise their efforts and make this work more effective.”

In neighbourhoods like Bordeaux, the organisations have been involved in a yearslong education drive to impart to residents the importance of separating their recyclable materials from their general waste and leaving them out on collection days for reclaimers to gather.

“We started working with the ARO in 2019, following Petco's donation of recycling bins. We saw a need to acknowledge the work that the reclaimers were doing [by collecting recyclable materials and keeping them from ending up in landfills],” says Angela Schaerer, a Bordeaux South resident who helped foster the working relationship between the homeowners in the neighbourhood and the reclaimers with the ARO.

ARO spokesman Luyanda Hlatshwayo says that reclaimers “play a vital yet often overlooked role in recycling. Building a circular, inclusive system requires collaboration among reclaimers, residents, companies, industry organisations such as Petco, and government. Communities and residents are encouraged to get to know the reclaimers' operation in their neighbourhoods, support their efforts, and foster mutual respect. Including reclaimers in waste strategies not only boosts recycling rates but also promotes social and environmental justice.”

NGS workflow stages

NGS is a high-throughput technology used for sequencing DNA or RNA, enabling detailed genetic analysis for genomics, transcriptomics, and personalised medicine.

The typical NGS workflow includes four key stages:

• Sequencing

Step 1: Sample preparation – Aliquoting and nucleic acid extraction

• Aliquoting: Sample types, like blood or tissue, are transferred from primary tubes into formats suitable for nucleic acid extraction. For automation, the ASSIST PLUS pipetting robot paired with the VOYAGER adjustable tip-spacing pipette is ideal for reformatting samples, speeding up workflows with precision.

Step 2: Library preparation - Converting nucleic acids for sequencing Library preparation involves three stages:

• Fragmentation: DNA is broken into smaller pieces

• NGS library generation: Adapters and indexes are added

Size selection: The library is purified, removing unwanted components

Automating this process with the MAG and HEATMAG modules streamlines workflows, reducing human error and improving data reliability.

Step 3: Quality control - Quantification, normalisation, and pooling

Quality control ensures libraries are prepared correctly by quantifying DNA concentration, normalising sample concentrations, and pooling them for sequencing. Automating these steps with the ASSIST PLUS pipetting robot ensures high accuracy and reproducibility in liquid handling.

MAG and HEATMAG Modules
