Peptidomics NanoEngineering Core The Peptidomics NanoEngineering Core provides customized designs, fabricates NanoTraps, and

Targeted Peptide Biomarker Extraction and Quantification (e.g., Hepcidin-25) Our customized NanoTrap design allows us to capture specific peptides in biological serum and then quantify the peptides using an internal standard. For example, we have successfully quantified hepcidin-25 from human serum, plasma, and urine with a high degree of accuracy and reproducibility. We can quantify any peptide biomarker given the appropriate internal standard. The internal standard is typically an isotope-labeled version of the desired peptide.

Our Thermo Scientific™ Velos Pro is a sensitive, high resolution, and fast ion trap mass spectrometer that can analyze the most complex proteomic and bioanalytical samples. The high resolution (60,000), accurate data quality (<1 ppm), and sensitivity of the instrument, make it ideal for the identification of low-level proteins in complex samples.

NanoTrap Fabrication The well-characterized NanoTraps can be fabricated to have different pore sizes (3-10 nm), pore structures (2D-, 3D-, or random), and surface modification (positive/negative charge, metal ions). This platform is inexpensive, high-throughput, and user-friendly. Only 10 ul of serum/plasma sample are required for a single assay, and 150 samples (i.e., 50 samples in triplicate) can be processed processed on a single NanoTrap chip.

1. The NanoTrap chip is used in combination with a MALDI-TOF mass spectrometer (Bruker, MicroFlex) or an ion trap mass spectrometer (Thermo Scientific™ Velos Pro). The samples (e.g., serum, plasma, urine, or cerebrospinal fluid) are analyzed by mass spectrometry after they have been processed on a NanoTrap chip.
