Editor’s Note If you’ve ever looked in the mirror, checked your birth year, and thought, “That can’t be right,” this issue is very much for you. From November 30 to December 6, the Andaman Sea once again became the stage for one of Asia’s most storied sailing events as the 37th edition of the Phuket King’s Cup Regatta unfolded against the luminous backdrop of Kata Beach. What if ageing isn’t inevitable, but editable? As technology, wellness, and mindset collide, age is fast becoming less a fact—and more a glitch in the system.

For most of human history, aging has been accepted as an unalterable fact of life — the quiet tax paid for the privilege of being alive. Wrinkles, frailty, cognitive decline and, eventually, death were treated as inevitabilities rather than variables. David Sinclair, Professor of Genetics at Harvard Medical School, rejects that assumption entirely. In his view, aging is not destiny but a condition — a malfunction in the body’s information system that can be slowed, halted and potentially reversed. Sinclair stands at the forefront of longevity science, challenging the most basic assumptions about what it means to grow old. From his early work on resveratrol and sirtuins to his current focus on epigenetic reprogramming, he is outlining a future in which turning ninety could feel far closer to turning forty. His vision suggests nothing less than a new operating system for human life. Central to his thinking is what he calls the Information Theory of Aging. For decades, the dominant explanation for aging was the “damage theory” — the idea that the body wears out like an old machine, gradually succumbing to oxidation, random mutations and accumulated stress. Sinclair argues this explains only part of the story. The deeper problem, he suggests, is not damage to the hardware but corruption of the software.

He often describes the genome as a compact disc. The DNA sequence — the “music” — is written in a digital language that remains remarkably intact throughout life. Aging does not destroy the music. Instead, the disc becomes scratched. These scratches represent epigenetic noise: disruptions in the system that tells cells which genes to activate or silence. Over time, cells lose their ability to read the instructions correctly. Liver cells begin to forget they are liver cells. Neurons lose their youthful precision. This gradual loss of cellular identity is what we experience as aging. The implication is radical. The information has not been lost, only obscured. If the scratches can be smoothed away, the original instructions can be read again. In this framework, youth is not rebuilt — it is remembered. Sinclair’s early scientific work focused on sirtuins, a family of proteins that regulate cellular health and longevity. These proteins respond to stress, energy availability and environmental signals, acting as custodians of the genome. His research brought compounds such as resveratrol into the public eye, alongside NMN, a precursor to NAD+, which supports cellular energy metabolism. The idea was that activating these pathways could mimic some of the biological benefits of exercise and fasting.

More recently, however, Sinclair’s work on epigenetic reprogramming has placed his laboratory among the most closely watched in the world. Building on Nobel Prize–winning research by Shinya Yamanaka, who identified genes capable of reverting adult cells into pluripotent stem cells, Sinclair posed a more restrained question: could this process be partially reversed, rather than fully reset?

A full reset would return cells to an embryonic state — a biological catastrophe. But partial reprogramming, he hypothesised, might restore youthful function without erasing cellular identity. His team’s experiments suggest this is possible. Using a subset of socalled Yamanaka factors, they treated mice with crushed optic nerves — injuries long believed to be irreversible. By reducing epigenetic noise, the neurons regained youthful behaviour, regenerated and restored vision. This work has since been extended to non-human primates, where similar techniques rejuvenated the optic nerves of green monkeys. The results represent a critical bridge between animal research and eventual human therapies. Gene therapies of this kind are powerful but complex and costly. Sinclair now describes his current mission as a Manhattan Project for aging: the search for chemical alternatives that can deliver similar rejuvenation without viral vectors. In 2023 and 2024, his group published data showing that combinations of small molecules could reverse cellular aging markers within days. Artificial intelligence plays a central role, screening vast numbers of molecular combinations before laboratory testing begins. The long-term goal is clear — a safe, affordable pill that could be taken daily to reset the body’s epigenetic clock. Measurement is another pillar of Sinclair’s approach. He co-founded Tally Health to make biological age testing accessible to the public. Using DNA methylation clocks, the company allows individuals to measure how lifestyle, diet and supplements influence their biological aging. The aim is a feedback loop that turns longevity into something measurable rather than abstract.

Sinclair is also a co-founder of Life Biosciences, a company applying partial epigenetic reprogramming to diseases once considered incurable. Its work targets conditions such as optic neuropathy and liver disease, with human clinical trials projected to begin soon. The ambition is not to manage decline but to restore tissues to a younger state by resetting their biological age. In the near term, Sinclair anticipates a shift in how people understand aging. Within five to ten years, he expects epigenetic age testing to become as routine as monitoring weight or blood pressure. At-home kits could reveal not only how old a person is chronologically, but how old their cells perceive themselves to be — and whether that number is accelerating or slowing. Alongside this will come the first generation of age-modifying medicines. These will not promise immortality but targeted rejuvenation, likely beginning with organs such as the eye or liver. Therapies that restore vision or reverse fibrosis by making tissue biologically younger could provide the first tangible proof that aging itself can be treated. For now, lifestyle remains the most accessible intervention. Sinclair is a vocal advocate of hormesis — the concept that controlled stress activates powerful survival mechanisms. Intermittent fasting, heat exposure through saunas, cold exposure and high-intensity exercise all fall into this category. While new therapies advance, these practices remain frontline tools for slowing biological aging. Beyond that lies a more speculative horizon. Sinclair speaks openly about “longevity escape velocity” — the point at which medical advances extend healthy lifespan faster than time passes. In such a scenario, lifespan becomes an open-ended variable. He has suggested publicly that the first person to live to 150 may already be alive.

In this future, aging would be actively managed. Every decade or so, individuals might undergo rejuvenation treatments that reset biological age, allowing life to unfold in cycles rather than a single trajectory toward decline. Time would still pass, but its biological impact would be continually softened. The societal implications are profound. If people spend fewer years debilitated by age-related disease, the economic and social benefits would be immense. A world where ninety-year-olds think, move and contribute like forty-year-olds would fundamentally redefine productivity, creativity and longevity. At its core, Sinclair’s message is disarmingly simple. Aging is not solely the story of systems breaking down. It is also a story of lost information — a software glitch in the operating system of life. And software, unlike hardware, can be debugged, rebooted and upgraded. The cure for death may remain distant. But if Sinclair’s work continues on its current trajectory, the choice to remain biologically young for far longer than any previous generation may soon be a matter of decision rather than chance.

Delegates travelled from across Asia, Africa, Europe, and the Americas, arriving via Suvarnabhumi Airport before beginning a carefully curated journey through the region. From the outset, the programme emphasised connection — between cultures, communities, and the landscapes that define this stretch of Thailand’s coastline. Early activities introduced contestants to Hua Hin’s heritage, including a visit to the town’s historic railway station, one of Thailand’s most photographed landmarks. The group then travelled south to Pranburi, where they checked in for a two-night stay at Pranburi Cabana Resort, a beachfront retreat that became a hub for both relaxation and creativity.
