- 7 The Food and Agriculture Organization, because of its interest in the control of foot-and-mouth disease throughout the world felt that the activities of a Central Reference Laboratory should not be con­ fined to European countries but should include all countries in which foot-and-mouth disease occurs. The Pirbright Institute is already in touch with many countries throughout the world.

The European Commission must also interest itself in the disease and the types of virus which are present in countries outside Europe.

While only the three classical strains of the virus have been demon­ strated in outbreaks of foot-and-mouth disease in Europe and in the Americas, from tho work at the Pirbright Institute throe further immunological typos have been shown to exist in Africa and, more recently, another type has been recovered from some parts of Asia, These findings have important implications for tho work of the Commis­ sion for it may happen one day that Europe may be invaded with one or several of those exotic typos. Thus, it is of considerable importance that information may be available on all typos of tho virus throughout the world.

Since the Third Session of tho Commission oorrespondenoo and dis­ cussions havo taken place with some of the United Kingdom authorities and it is hoped that a further statement on this subjoct will be made at tho Fourth Session of the Commission. liutation of tho Viruo of Foot-c.nd-:�10U-t-h Disease Since it was shown in the 1920s that there existed three distinct immunological types of foot-and-mouth disease virus in Europa, tho question of mutation from ono typo to another has often boen tho sub­ ject of discussion. Recently, further consideration has boon given to tho possibility because of the oxpcrionce during tho foot-and-mouth disease outbreaks in Switzorlan4 in May-July 1956, Professor Fluckigcr presented a report to tho Commission's Executive Committee in Soptombor 1956 and this has subsequently been published (Bull. off. intornat. des Epizootios, 1956, Vol. XLV� pp.707-716).

Following this report, tho Executive Committee decided that a Committee composed of roprosontatives of tho Commission and of tho Office International dos Epizootios bo set up to discuss tho subject and, if thought necessary and desirable, to suggest and initiate research work on it.

With the concurrence of OIE, this Committee was formed and mot at the Office of tho OIE on 8 November 1956. The OIE was ropresentod by Dr. G, Ramon, Dr. A. Merlo, Dr. R. Vittoz, Dr. E. Mathieu, Prof. F. Lucam and Prof. G. Fluckigcr: the representatives of tho Com­ mission were Dr. H.S. Frenkel, Dr. I.A. Gallo�ay, Sir Thomas Dalling and Dr. E. Fogedby.

The findings and decisions of the Committee may bo summarized as follows:- Mutation of tho virus of foot-and-mouth disease may be defined as a complete change from one distinct immunological type to

another. Thus, variation within a ty:pe cannot be considered as truo mutation. While it is generally believed that mutations havo taken place in tho field on some rare occasions, the whole question of mutation of tho virus of foot-and-mouth disease has so far not boon scientifically provedg in fact, very littlo is knovm about such mutation.

On the other hand, it is knom1 that strains of tho virus of recognized and proved immunological typos have been kept for many years in laboratories and have maintained their typo specificity, The Swiss exporienco led to tho suggestion that typo C virus had mutated to typo O and that this typo O virus had mutated to typo A, all within a short space of -time. It vms agrood that if this wore so, it must be quite exceptional.

Tho Committee felt that it would be very difficult to plan any ,research which would domonstrato mutation experimentally and that it would be bettor that the different laboratories work on the subject.

If any observations of value emerged from such work, consideration might bo given to a further mooting of this Committee, Dura.tin� of L"J::::1up.ity follo·,--;h-i..� Vo.ccinntio�n nga.in::::t Foct-a.r.J.-lfouth Disease At different sessions of tho Commission, discnnmiuno lravu �o.lr�n place on tho carrying out of experiments to demonstrate the duration of immunity following vaccination against foot-and-mouth disoa.so.

Sovoral plans for such oxperimonts havo b0on drawn up. Thus in 1956 an oxporimont ';,ras discussed. and somo details -,-rnre produced in vvhich groups of cattle in throe difforont European countries would be vac­ cinated with a trivalent culture virus vaccine and challenged at different intervals. It vras hoped that tho three countries concerned ·,-,,rould boar somo of tho costs of tho experiment and that some financial support vvould also be forthcoming from international organizations.

This type of oxporimont Yms suggested bocauso tho Italian Veterinary Authorities have strossed that thore is not yet enough information on tho value of culture. virus vaccine nnd that more experimental vvark should be carried out.

On further discussion at an informal mooting of somo members of the Commission's Standing Technical Committee in Paris in May 1956 ? it was agreed that an experimont using a considerable number of cattle should be undertaken at tho Virus Research Institute, Pirbrightg but, as tho results of such an oxporimont would not be available for some con!lidorablo time, it ,;ms further agreed that smallor-ecale experi­ ment� should also bo carried out at institutes in different countries in order that information could be accumulated, Tho latter oxporimonts would. follow tho design referred to in the above paragraph.

In ardor to como to a final decision on tho design and nature of tho experiments a group of workers on foot-and-mouth disease, roproscm­ ting tho Commission and OIE, mot in Paris on 3-4 December 1956. Tho

OIE was r0prosent0d by Prof. F. Lucam 9 Dr. G. Moosbrugger and Dr. R. Willems. Th0 Commission v,as represented by Dr. I.A. Gallo·imy, Dr. H.S. Frenkel and Dr. E. Fogedby.

The problem was thoroughly dis­ cussed and it ryas felt by tho majority of the group that the subject should be approached at a high scientific level. Tho group, thoroforo; made the following proposals g.- the v,hole experiment should be carried out at ono centre; a comparative tost between a "culture" virus vaccine and a "natural" virus vaccine should be carried out; only one type of virus vaccine should be investigated at a time; the number of' animals in the experiment should bo increased from that originally suggested; in order to minimize the risks of infection and so that suitable animals might be readily provided and maintained, tho experi­ ment should be carried out at tho Research Virus Institute, Pirbright; Dr. Frenkel be roquosted to prepare two monovalent vaccines for tho purpose of tho oxporiment (a "culture" virus vaccine and a "nature.l" virus vaccine); tho viruses used for these vaccines should be titratod at tho Pirbright Instituto and each of tho vaccines should contain th,, same quantity of antigen� tho immunity produced by each vaccine should bo challongod at throe, six, nine and tvrnlvo months after vac­ cination, ten vaccinated together with 2-4 control animals being used at each challenge; that 184-192 cattle be provided for tho oxporimont.

It is oxpocted that information on tho financing of tho suggested experiment ·,7ill be available at tho Fourth Session of tho Commission. f.'oot-o.nd-I.Io-1:!ih DiG-On.o·o in L\tropoan Countries The disease position in Europe has boon kept under review since tho last Session of tho Commission and reports have boon made by tho Secretariat to tho Executive Committee. In addition to visits made by tho Secretariat to different countries, tho Executive Committoo received verbal reports from tho Directors of Veterinary Services of Belgium, tho Gorman Federal Republic and Switzerland at a recent moot­ ing. During 1956, tho Socrotariat visited Portugal, Spain, tho Nether­ lands, Franco, Switzerland, Denmark and tho United States of America.

Early in 1957 short visits �oro also made to tho Gorman Federal Republic, the Netherlands, Belgium, Franco and Austria in connection v,ith tho increase in tho incidence of foot-and-mouth disease in Europe, In Portugal, foot-and-mouth disease has occurred fairly froquently in tho past, invasion, as a general rule, having takon place from Spain.

On tho other ha.nd, some frozen moat is imported from South American countries, especially Brazil, and constitutes a further potonticl source of infection.

Tho overall position in Portugal is not con­ sidorod to bo serious and outbreaks aro usually mild in character and tho disoaso has shovm a. tcmdoncy to die out. Bocauso of the risk of introducing exotic typos of foot-and-mouth disease virus, ospocially tho African typos, tho importation of cattle into Portugal from AngolA, has boon discontinued.

Foot-and-e10uth disease opizootics in Spain aro, as a gon0ral rulo, caused by invasion of tho disease from Franco. At tho beginning of 1956 tho diseaso flared up and caused a number of outbreaks but, although opizootics in Spain have of ton boon sovoro, YJith considerable economic loss, tho 1956 outbreaks vrnro of a mildor cha.ractcr.

In tho Netherlands, only 47 outbreaks occurred during 1956. A systematic vaccination prograra has boon followed a.nd, combined rlith tho slaughter of affoctod animals (usually pigs and unvaccinatcd young stock), has maintained a remarkably satisfa.o-tory disoaso position.

A policy of slaught or of 2.ffoct cd animals has boon mado. possible because of tho results of □ass vaccination, In Switz or land, aft or the outbreaks in May-July 1956 n:ttri hut.�.hl r: to pigs in transit from Belgium to Italy r.'oro brought under control, only sporadic outbreaks have b0on reported. Tho Swiss system of controlling foot-and-mouth disoaso (combination of slaughter, vaccina­ tion, disinfection and ::1ovomont control) continues to gi vo good results.

In Belgium, tho uso of trivalent vaccine is being extensively practised. Tho vaccine consists of culture virus of O and C typos and natural virus of A type. Thero is clear ovidonco that this vaccine confers a considor.1blo dogroo of immunity>. 1:1ost of tho outbreaks v,hich are recorded occur anong pigs •,·vhich are not vaccinated, or in young, unvaccinatod co:ttle. Vaccination is being encouraged.

Observations in Belgium indicate that� although tho vaocina.tion of pigs may bo followed by an irnmuni ty of short duration, tho poriod of pro­ tection may be of considorablo value in preventing spread of tho infection, In tho Gorman Federal Republic, vaccination is being practised and is giving satisfactory rooults. Vaccination hc1s boon usod along frontiers throatenod by invasion and in localized outbreaks to prevent spread of tho disease. Slaughter of affected animals has been carried out in a proportion of outbreaks but is consid0rod to ·bo impractical in largo herds, Tho prosont position of foot-and-mouth disease in tho Gorman Fodoral Republic would appoar to be well in hand.

In Denmark, only few outbreaks have boon rocontly recorded, Vaccination is proceeding in tho southern part of tho country Yrhoro it is appreciated that tho method of control usod in Germany has provontod tho usual mass infection.

In tho Unit0d Kingdor:i, there -,,ere many introductions of foot-and­ mouth disease virus in 1956. Satisfactory control has been exorcised by the usuaJ. stamping-out policy ·irith all it involv0s by way of slaughter of stock, mover.ient control, disinfection of premises, general sanitary 1i1easures, etc. On circuostantial GVidGnce, sources of infec­ tion are fro□ ioported South Ar.ierican r,rnat and by indirect means including birds fro□ continental Europe.

All restrictions had boon removed by tho end of F0bruary 1957 and th0 country ·;ms once r.1oro froo froc foot-and-mouth disease.

In Italy tho disease position is probably unaltered, In Franco there is apparently no improvooont in the recent posi­ tion.

It is unde.rstood that consideration is being givon to tho adoption of a slaughter policy for a particular region and that there arc efforts to oncourago further vaccination.

There appear, hoYrevor, to be insufficient supplies of vaccine and, even if adequate, thoro aro too few veterinarians to undertake an effective vaccina.tion progran.

Norway, Sweden, Iceland, Finland and Ireland continue to bo free froi:1 tho disease.

Yugoslavia remains free from foot-and-□outh disease and so far as information is available, tho position in Eastern Europe continues to be satisfactory.

In Greece 9 sor;1e few outbree.ks occurred during 1956g inforraation is that tho position is now satisfactory.

Austria, after a long period of freedom, became infeotod in D0cer.1ber 1956, close to tho Bavarian border. After tho r;.iddlo of January whon 42 outbreaks wore rocordod in one week, the incidence docroasod and no further outbreaks have boon recorded since 7 Fobruary.

The Austrian authorities adopted rigorous r.10asuros of slaughter and vaccination. An urgent request to tho Cor.1oission for supplies of vaccine for tho purpose of intensifying tho control oeasur0s was dis­ cussed at a special r.�ooting of tho E:x:ocutivo Cor.1rnittoe in Dublin in ,January 1957 nhen it -:ms docid.:Jd to release 500 frori1 the Special Account of tho CoLmission for tho purchase of tho vaccine, which ·,;as nado available 0,t a specially reduced cost by tho Netherlands Govern­ □ont. This vaccine m.1s used to comploto, as :i:ar as possible, vaccina­ tion of anir,mls in tho Upper Austrian and Salzburg area where tho infection vm.s hoaviost and to broaden tho zone of vaccination.

Tho E:x:ocutivo Coomittoc have noted with ouch interest that Y1here reliable vaccine ·;,as properly or systoraatically a.pplied, outbreaks of tho disease arc either prevented or, whon they occur, arc quickly brought unclor control. Frenkel, sorno experiments ·carried out in tho Netherlands on the duration of immunity follovdng tho vaccination of cattle 11ith 11 culturo11 virus vaccine.

Systematic vaccination of cattlo in tho Netherlands was begun in February, 1953, a trivalent culture virus vaccine, produced .;d th viruses typos O, A and C being usocl. In tho Netherlands all cattle aro vaccinated and annual rovaccination is carried out g thus, there arc cattlo in the country ·.--rhich have boon vaccinated from one to four timos. During 1954-56 vaccinated cattle have bocn tostod for immunity and thoro is 0, considorablo numbor con­ cerning v1hich data aro available. Tho rosults of the challenge experi­ ments showed that tho cattle possessed a satisfactory dcgroo of immunity up to ono year aft or vaccination with, of course, n clocroaso of immunity towards tho ond of that period.

Ono of tho visits to Franco was, in addition to attending and participating in tho annual meeting of tho OIE, to visit a private veterinary institute in Tours in order to obtain detailed inforr.iation on tho production of foot-and-mouth disease viruses for vaccine pro­ duction. Tho method used in this institute consists in the simultaneous production of foot-and-mouth virus and vaccinia virus on tho skin of adult cattle.

In July and August 1956 9 Dr. Fogedby worked for eight wooks at tho Danish Veterinary Institute for Virus Research. Ho devoted this time mainly to work on problems of immunity to foot-and-mouth disease and tho typing of viruses.

In Soptombor 1956, by special invitation, Dr, Fogedby attondod tho official dedication of the new Animal Disease Laboratory, Plum Island, United States of America and also took part in a symposiur.i on vesicular disoasos of livestock, Tho major emphasis of tho future research at this lc:boratory i,-1ill bo on foot-and-mouth disoaso: other vosi-cula-r typos of diseaso, o. g, vesicular stotJ.atitis and vosicular exanthoma ·.-rill also be studied. The Government of Belgium expressed

its willingness to receive the group and suggested that :Dr. G. Ramon, Director, OIE, should join the group.

Unfortunat0ly, after full arrangements had been completed for tho visit, neither Prof. Fluckiger nor Dr. Ramon found it possible to make the visit.

The representatives of tho Commission were warmly received by the Minister of Agriculture and the veterinary officials. The Minister summarized tho foot-and-mouth disease position in Belgium and expressed his confidence, shared by practicnlly every Belgian farmer, in vac,cina­ t±on as a means to control tho disoo.se. For the tir;ie being, a slaughter policy could not be introduced in Belgium. It ua.s, however, evident that the number of outbreaks of foot-and-mouth diseas0 was closely collated to tho extent of vaccination and; thorcforo, as much vaccina­ tion as possible is being encouraged throughout tho country.

Proposal to amend the Constitution of tho Commission At tho Second Session of the Commission, the observer from Portugal statcxl that his govornmont intended to become a Member of ·tho Commission, but that his government strongly felt that the annual con­ tribution for Portugal fixod in tho Constitution was excossive ancl suggested that it should be halved.

In 1955, Portugal became a Hor.:ibor of tho Commission. In a lotter of 23 Novombor 1956, tho Portuguese Govornmcmt informed tho Chairman of tho Cor.iuission and the Diroctor-Goneral of the Food and Agriculture Organization that tho Portuguese roprosontativo had boon instructed to propose at tho next session of tho .Commission that th0 Portugu·eso· con­ tribution b0 roducod to �n50 per annum and asked that this subject be included in tho Agenda. In accordanco with Article XIV of tho Consti­ tution of tho Commission, tho Diroctor-Gcmoral of tho Organization informed all Members of tho Commj_ssion 11bout this request for an amond­ ment of tho Constitution and that an appropriate i tom would: bo incJ.uilod �n the Agenda for tho forthcoming session of tho Commission. In tho course of tho discuasion, th'? Secretariat reported that it was hoped to finalize at an early dato tho arrangements that the Virus Research Instituto� Pirbright, Unitod Kingdom, would be the C antral Roforonco Laboratory' for foot-and-nouth disoaso and that unclor FAO arrangements it was intended also to use this institute as a World Roforonce Laboratory.

In connection with tho duration of j,mmunity oxporiments, tho Con­ mission decided strongly to support tho carrying 01-1-t of thC largo exporiraont which was agreed w:i.th tho joint OIE/Comrllission group in Paris.

It should !i1ak.e this known to OE.EC and invite financial support for this experiment and to a lessor extent for tho smaller cxporimcnt proposed to bo done in Belgium. Furthor 9 in order to explain tho need for and tho value of this major oxporimont, it was docidoa to tak.o advantage of tho offer of FAO to ask Sir Thomas Dalling to attend tho mooting of tho Ministers' Doputios of OEEC.

- 14 Supplementary statements were made in c·onnection with the reJ)ort on the disease position in certain countries. Tho delegate from Deninark and the delegate from the United Kingdom made statei;1ents.

The delegate from tho United Kingdora reported that there had boon four primary outbreaks of disease during the wonth of March 1957. The delegate from Denmark -reported that there had been olevon outbreaks in the southern part of Denmark, three of which wore in Jutland, Most of thoso outbreaks were of As and o2 typo virus, consistent with tho typos being found in outbreaks in the neighbouring part of Western Goroany.

However, rocontly a C type virus had been encountered and supplies of vuccinc against this typo \7ero not entirely adequate and it might be .. that this virus typo would dominate the situation, ·.-,hich tho dologato considored ,night d.etoriorat o in the next few 'l'!ocks. · The st.:i.nping-out policy v1as being used in supplor.1ont to vaccination, but tho delegate v1as not confident that this could bo uai.nt{l.inod· if tho outbreaks became too numorous, because of financial difficulties.

Supply of Vaccines The question of sup.plies of vaccine was also discussed, particu­ larly tho part which tho Commission may play in arranging for supplies for countries in which the amounts available for any special purpose The Committee considers that the Commission should were insuffic"ient. satisfy itself that the tests for innocuity and potency carried out i_n the centres from which it obtains vaccine are adequate and that consideration should be given to the desirability of supplementing those by periodic-check tests. It was decided that these points shouJ.d be discussed in detail at an on.rly mooting of tho Committee and to facilitato thoao discussions the Secretariat should obtain inforr;mtion on tho methods of production and tho tests for innocuity and potency currently in use in supplying laboratories 'in Member countries.

Rocont Progress in Virus Cultivation Tho Cammittoo discussed tho progress :j.n cultivatio.n of virus for vaccine production. Cultivation on tissue has so many advantages f'or tho production of vaccine that further work should continue to be done on tho subject. Thero is avid.once of improvement ih tho immunizing value of the vaccine following the roaninalization of culture virus, It is roalized. that various strains behave differently in culture and tho advantages of roanimalization probably apply more particularly to certain strains. Novurtholoss, tho immunogonic cho.ractor of strains passaged continuously in culture should be oxarained periodically.

In view of tho practical difficulties v7hich ariso in certain countries in obtaining sufficient tongue tissue for largo-scale produc­ �iorr, consideration of tho possibility for using o�her tissues for vaccine production should c-1ntinuo to be given and particularly it would be inportant to oxporinent with tissues which would bo readily available ,for largo-scalo production.

Following discussion in which members of tho Technical Committee took part, tho report was adopted.

The budget estimates have been drawn up in the form established in the Ftnancial Regulations.

No expenditures have so far boe� made under this Chapter and in tho absence of any experience to guide mo I recommend that an arbitrary sum of $5,000 be provided hero for 1958; this is the same amount as was allowed in tho approved 1957 budget.

It will be noted that tho proposed Annual Administrative Budget for 1958 totals j�50,ooo, i.e. the maximum potential contributions from Member Governments that may be received by the Commission.
