ABSTRACTBODY:
Objectives:Endodontictreatmentfailuresdueto Enterococcus faecalis andotherbacteriaremainsa significantchallenge.Calciumhydroxide,acommonlyusedintracanalmedicament,hasmanylimitations, includinglowflowability,difficultyinfullretrieval,andlimitedefficacyagainst E. faecalis Thisstudyaimsto assessamixtureofcalciumhydroxideandsalicylicacid(CASA),encapsulatedinthermo-reversibleF127-DMA hydrogel,asaninjectable,retrievableintracanalmedicamentwithantibacterialactivityagainstfibrinogen modified E. faecalis and Streptococcus mutans
Methods:CASAwaspreparedbymixingcalciumhydroxideandsalicylicacidina6:1ratio,respectively,and incorporatedintoF127-DMAatconcentrationsof5%,10%,and20%(w/w).ThemixturewasUV-crosslinked usinglithiumphenyl(2,4,6-trimethylbenzoyl)phosphonate(LAP).Chemicalpropertieswereanalyzedusing XRD,Ramanspectroscopy,FTIR,andXRF,whileinjectabilityandretrievabilitywereassessedusingaplastic rootcanalmodelandrheology In vitro antibacterialefficacyagainst E. faecalis and Streptococcus mutans biofilmswastestedusingbovineextractedteeth.Additionally,Cytotoxicityanddoublingtimeofdentalpulp stemcells(DPSCs)weremeasuredinresponsetoleakedCASAfromthehydrogel. In vivo modelusingbeagle dogswithinducedapicalperiodontitiswillassessbacterialreduction(CFU),periapicallesionchanges(3DCT scans)ofF127-DMA/CASA.
Results:EncapsulationofCASAinF127-DMAreducedtheviscositybythreeordersofmagnitudecompared tothepasteform,facilitatinginjectionandretrieval.Furthermore,XRD,Raman,andFTIRanalysesconfirmed theformationofcalciumsalicylateandsuccessfulintegrationofCASAintoF127-DMA.Nosignificant cytotoxicitywasobservedinDPSCs,anddoublingtimewasnotsignificantlydifferenttocontrol.F127-
DMA/CASAexhibitedcompleteeradicationof E. faecalis inplanktonicformandthebiofilmmodelshoweda three-logreductioninbacterialcountandnosignificantdifferencetotheendodonticgoldstandardofCa(OH)
Conclusions:F127-DMA/CASAexhibitssuperiorflowability,retrievability,optimalpH,biocompatibilityand antibacterialefficacywhichhighlightsitspotentialasasignificantimprovementovercalciumhydroxidefor endodontictherapy
