Armed with a seven-figure grant, Duke researchers hope to better understand how DNA changes can cause cancer.

Brought together by a new research project funded through a four-year, $2 million National Science Foundation grant, four professors of biomedical engineering are looking to look deeper into the structure of DNA and how it impacts cancer development.

Charles Gersbach, Rooney Family Associate professor of biomedical engineering, Brenton Hoffman, assistant professor of biomedical engineering, Michael Rubinstein, professor of mechanical engineering and materials science and professor of biomedical engineering, and Xiling Shen, Hawkins Family Associate professor of biomedical engineering, will all combine their individual knowledge of gene editing, mechanical engineering and computational modeling.

The research is supported by the NSF Directorate for Engineering’s Emerging Frontiers in Research and Innovation program, which aims to address large-scale engineering challenges.

The researchers hope to better understand gene function and how to control DNA structure, as well as the interaction of chromatin with DNA, which can cause cancer development in some conditions. The team is investigating how changes to the cell nucleus’s structure are expressed.

If the three-dimensional structure within the nucleus is perturbed, then the expression level of the various proteins produced is altered, which can impact cell signaling and behavior. This can lead to a disease state.

Using their newly developed technology, the team has discovered that these structures are mutable.

develops imaging tools, Hoffman said.

Combining their skills, they hope to make improved tools to use with Shen’s models to start understanding how epigenetic structure regulates the expression of MIC, a protein found in cancer cells, especially colon cancer.

“There’s a structure, we can change it. We are looking at how and should you do that and then what can it do,” Hoffman said.

This is where Gersbach’s variation of the CRISPR-Cas9 system—engineered to manipulate the epigenome at very specific sites modeled by Rubinstein—comes into play. The team then uses Hoffman’s imaging technology to learn the effects of epigenome changes.

Through this, they will be able to change the structure of the chromatin within DNA and see how different changes can cause cells to become cancerous in certain situations.

“There’s a three-dimensional structure within the genome and that structure effects protein level expression. It’s not just cool how it’s arranged. It has consequences for how much and what proteins are produced,” Hoffman said.

Hoffman explained that the feedback-loop nature of his collaboration with Rubenstein makes this investigation unique.

“He can make predictions, his prediction will hopefully be something that we can directly measure and then by using technology we can see essentially the two chromatin points come close together and then my data will help inform his model. And that’s never been done,” Hoffman said.

Previous research conceptualized chromatin structure as random. Scientists now know that this is not the case and are looking to understand how different formations have different effects on the protein produced from DNA. This can allow a better understanding of which part of DNA expresses proteins and how that can contribute to the development of diseases like cancer.

“From a scientist’s point of view, it’s great to understand that basic structure. From a societal point of view, it’s great to use that understanding to do something,” Hoffman said.

How Duke’s $8.5 billion endowment stacks up against its peers Jeremy Chen | Graphics Editor

A slew of American universities released fiscal year 2018 endowment figures in the past several weeks, including Duke, whose endowment returned a 12.9 percent growth to a record $8.5 billion dollars.

The growth marginally outpaced Duke’s 12.7 percent growth in the previous year. Managed by the Duke University Management Company, known as DUMAC, the Duke Endowment is the 10th largest private college endowment in the United States this year, and Duke’s 2018 growth was tied with the University of Pennsylvania for third among the top 10 endowments.

Michael Schoenfeld, vice president of public affairs and government relations, said that the expanding endowment is the result of DUMAC’s hard work.

“Obviously, DUMAC had a very good year,” Schoenfeld said. “We had very smart and strategic investment managers in DUMAC, and having an endowment that grew at 12.9 percent last year will continue to provide Duke with a level of financial support and infrastructure that will make it possible to continue to do important things like expand financial aid, endow professorships and support programs.”

college endowments this year, according to preliminary data from Cambridge Associates.

Among the top 10 private college endowments, only Princeton and the Massachusetts Institute of Technology had higher returns.

The Duke Endowment’s 2018 returns also beat the 8.4 percent gains that a typical 6040 portfolio of S&P 500 stocks and aggregate bond index investment grade bonds would have returned in fiscal year 2018, according to Markov Processes International. The endowment’s growth also surpassed the S&P 500’s 10.8 percent growth over the same period.

The endowment includes around 5,200 funds as of June 30, according to a snapshot of the endowment released by Duke. Long-term targets for investment are 48 percent invested in equity, 15 percent invested in credit and the remaining percentage doled out among commodity, real estate, rates and more.

“Diversification across investment exposures is central to generating strong investment returns while moderating portfolio risk and volatility,” the report reads.

Spending from the endowment is divided into several pools, with 21 percent devoted to financial aid, 19 percent to professorships and 10 percent to instruction and research. Another 27 percent is devoted to the “unrestricted support of the university or one of its schools or budget centers,” according to the report.

Schoenfeld declined to comment on specific asset allocations. One of the most significant uses of college endowments is financial aid, and colleges have been increasingly vying to provide better financial aid packages to attract the best students from around the world.

“Over the years, Duke has continued to increase and expand opportunities for financial aid,” Schoenfeld said. “Affordability and access are very important to this institution.”

Many universities with smaller endowments—such as Amherst College, Brown University and Davidson College—only provide no-loan financial aid packages. Rice University, with a $5.5 billion endowment, announced in September that it will begin offering fulltuition scholarships to undergraduates whose families earn less than $130,000 a year, with students whose family incomes are lower than $65,000 also receiving grants covering the total costs of room, board and fees.

“How [financial aid] gets done is really highly specific to every institution, but in recent years, we have significantly reduced the amount of loans that are in financial aid packages,” Schoenfeld said.

Regarding the potential for increased endowment funds towards financial aid, Schoenfeld noted that Duke cannot repurpose donations towards financial aid if they were initially endowed for other purposes. In fact, even the investment return on a gift “has to be used for the purpose for which [the gift] is designated.”

Therefore, Duke’s fundraising priority has been to raise money for financial aid endowments to lessen the burden of financial aid on the operating budget, he said.

Although the endowment’s 2018 performance is encouraging for the University, plans to improve Duke’s financial aid with endowment funds may be hindered by the 1.4 percent endowment tax legislated this year in the Bipartisan Budget Act of 2018.

“We expect we will be impacted eventually, but it’s unclear because the rules have not been confirmed yet,” Schoenfeld said. “There’s going to be less money that goes to financial aid, there’s going to be less money that goes to scholarships, there’s going to be less money that goes to maintaining the infrastructure, and that’s just contrary to everything that both an endowment and public policy towards education should stand for.”

Pair of new dorms planned for Duke job applicants no West Campus, Larry Moneta says longer have to disclose criminal history

As Central Campus retires at the end of this school year, West Campus is about to embrace more residents in upcoming years.

Speaking to the Duke Student Government Senate, Larry Moneta, vice president for student affairs, discussed plans to build two new residential halls on West Campus to accommodate students living in the 300 Swift apartments.

“Our goal is to get two more buildings on West Campus as soon as we can,” Moneta said.

Speaking to The Chronicle after his talk, Moneta pointed to two areas on West Campus for possible development. One is located near the Duke Wellness Center, at the intersection of Towerview Road and Science Drive. Several sites between the Hollows and Edens Quad are also under consideration.

The source of funding is still undetermined and there is therefore no formal timetable for the construction at the moment, Moneta added. However, the hope is to accomplish the project within five years.

In the meantime, undergraduates are being housed in 300 Swift, which Duke purchased for $50 million in late 2016 to house students displaced by renovations to Crowell and Craven Quads. Moneta added that when the new dorms are ready, the 300 Swift apartments are likely to be converted to either graduate apartments or market housing, as originally intended.

This year also marks the last year for students to live on Central Campus. A plan to redevelop Central is currently developing, Moneta said.

“I’m grateful that I will be here long enough to truly see the end of Central Campus,” said Moneta, who is stepping down as vice president for student affairs after this academic year.

Moneta pointed to the opportunity to redevelop the 58 acres of Central Campus as an exciting next step.

“I personally hope to be able to take a sledgehammer to something,” Moneta said, joking about the demolition of Central Campus.

The closing of Central will officially take place the day after commencement at the end of the 2018-2019 academic year. According to Moneta, Duke’s long-term housing plan is geared towards moving all students to West Campus.

“West Campus is the place to be,” he said. “We’ve made this incredible investment.”

The developments of plans for the new buildings, including living arrangements and locations, will be spearheaded by the Next Generation Living and Learning Experience Task Force, which was established by the Board of Trustees this May. It has not yet been determined whether the new dorms will be apartments, suites or traditional doubles, Moneta said.

Applicants to jobs at Duke will no longer have to say whether or not they have been convicted of a crime, according to a news release.

“While responses to the question are not currently shared with hiring managers, asking for this information in an application can lead to a misperception among applicants that the responses are used to filter out those with a criminal record from employment opportunities at Duke,” said Kyle Cavanaugh, vice president for administration, in the release.

In an email to The Chronicle, Cavanaugh wrote that administration had two meetings with subgroups of students about the change.

“This has been something under discussion and review for some time,” he wrote.

The change, which is in line with the national push to “ban the box,” was one of the demands issued by the People’s State of the University last semester. This is the first of PSOTU’s demands to be met by administrators.

Cavanaugh explained that other schools have made the change to comply with local laws on the subject.

“I certainly don’t think we can [say] we are first, but I do think you can say that is rare that a institution makes this move when not also being required to do so by local law,” he wrote.

The new policy, effective Nov. 1, will not stop Duke from conducting background checks. Duke will still do background checks for government sanctions, criminal records and driving record history— when relevant. These checks will be performed during the offer stage for finalists for positions that have been “recommended for hire,” according to the release.

Duke gains one spot in U.S. News rankings Staff Reports The Chronicle

Duke regained its eighth position in the U.S. News and World Report rankings this year.

The University, which fell behind the University of Pennsylvania to ninth place last year, is tied with Penn for the 2019 rankings. The three years previously, Duke was in eighth.

Princeton maintained its position at the top of the rankings for the eighth year in a row, beating out Harvard, which again came in second. This year, there was a four-way tie for third place between Columbia, Massachusetts Institute of Technology, University of Chicago and Yale.

The four-way horse race was followed by Stanford, then Duke. Johns Hopkins moved back into the top-10 in a tie with Northwestern, bumping out the California Institute of Technology.

As Duke gained one spot in the national university rankings, Williams maintained its hold on the top of the list for best liberal arts colleges.

Aside from the best national university and liberal arts colleges rankings, U.S. News and World Report also publishes more specific rankings for a wide swath of college features.

In the “Best Values School” category, Duke dropped from its 10th position last year to 13th this year.

The Pratt School of Engineering moved up from 20th place to a tie for 18th place this year, compared to 18th in 2017.

Duke dropped from 10th last year to a tie for 16th this year in the “Most Innovative Schools Category.” For “Undergraduate Teaching,” Duke maintained its hold on 10th place, compared to ranking 14th in 2017.

The Blue Devils, however, remain firmly ahead of the University of North Carolina Chapel Hill, which moved into a tie for 30th from its ranking of

The U.S. News and World Report rankings are based on statistical and qualitative measures to parse out the rankings, and the rankings take into account a wide variety of factors. In order of their weight, the factors they consider are graduation and retention rates, undergraduate academic reputation, faculty resources, student selectivity, financial resources, graduation rate performance and alumni giving rate.

In recent years, Duke has hovered safely inside of the top-10 bubble. Its highest ranking in the last five years was a tie with the MIT in 2014 for seventh place.

The rankings for the top-10 universities are

somewhat stagnant from year to year. From 2013 to 2018, the same 12 schools—Princeton, Harvard, Yale, Columbia, University of Chicago, MIT, Stanford, Duke, Penn, Dartmouth, Johns Hopkins and CalTech—have floated in and out of the top-10 spots. This year, Northwestern entered the mix.

For the last seven years, Princeton has held the top spot, beating out its repeated runner-up Harvard each year since the schools tied in 2013.

Last week, the Wall Street Journal/Times Higher Education rankings Duke slipped to seventh place position for U.S. schools from fifth.

Senior Trey Walk, an organizer in PSOTU, noted that the policy change is important because Duke is the largest employer in Durham and it sets an example for other businesses in the area.

He said he hopes it has a cascading effect on the area, but noted that “second-chance hiring” goes beyond removing the initial disclosure. Going forward, the student organization and community partners are pushing for targeted hiring and skills training.

Senior Sydney Roberts, an organizer for PSOTU and a co-chair of The Chronicle’s independent editorial board, said these can have positive economic impacts on local families.

“There are no downsides to these changes,” Roberts said.

The change is in line with the direction in which Duke has been moving on the issue, Cavanaugh wrote to The Chronicle.

“Duke transitioned to the required [Equal Employment Opportunity Commission] guidelines several years ago, and the move to eliminating the self-disclosure section on applications will not be a huge lift,” Cavanaugh wrote.

What does it mean to be a cultural anthropologist? Professor Charles Piot will share his ongoing research in a remote area of West Africa, an area that has all the earmarks of tradition and yet, Piot suggests, is modern through and through.

Utilizing the lens of photography, he will also examine the projects of DukeEngage students he has taken to this area over the last ten years: the health insurance system they have set up in a village clinic, the two cyber cafés they have established in villages without electricity, the literary club they run each summer, and the computer classes for elementary school children who have never seen a computer before.

Founded in 1979, the Pitchforks are the oldest a cappella group at Duke University. They perform on campus and across the country throughout the year, appearing in venues ranging from dorm common rooms to professional sports arenas. They have sung the national anthem at Chicago Bulls and Duke Basketball games, showcased themselves before the Queen of Jordan, and even opened for Ben Folds in 2009 and The Band Perry in 2015. In 2014 and 2018, the ‘Forks crossed the Atlantic to tour with some of Britain’s best collegiate singers. They have a rich history of recording and have twice won the CARA Award for Best Male Collegiate A Cappella Album. Their annual Fall Classic show has been a staple of Family Weekend for a long time, and this year’s shows will occur on both Friday, October 19 and Saturday, October 20! Lady Blue was founded in 1991 and is one of Duke’s premier all-female a cappella groups. This exhibition presents work by 39 artists in a group exhibition exploring the striking cross-currents of photographic work happening in the Piedmont region of North Carolina.
