What are “Natural Products”?

Chemical characteristics • Naturally-occurring small organic compounds • including heterocyclic compounds, and peptides.

• does not include proteins, carbohydrates, and nucleic acids.

• MW: ~150 ~ <800 amu (“small molecule”) • Methods of extraction and purification are generally similar to the techniques used for organic compounds (e.g., TLC, column chromatography, HPLC, GC) • Methods of structural determination • NMR, MS, IR, X-ray, UV 1. What are “Natural Products”?

Biological characteristics • Compounds are generally characteristic of a particular species or family, i.e., narrow taxonomic distribution (nonubiquitous) • No nutritional or structural function. Functional roles may include: - color (identification) - scent (attraction or repulsion) - sexual attraction - social communication - defense (e.g., plant toxins and antibiotics)

but many still have unknown function in the organism in which they are found.

Why study Natural Products?

• Natural products are the source of the most complex and fascinating chemical structures.

• Natural products represent biological diversity.

• Natural products are expressions of the genome.

• Natural products represent natural biological activity, whether as single compounds or as complex mixtures.

5. Xác định cấu trúc hóa học các hợp chất thiên nhiên

Phổ IR: Phân tử hấp thụ năng lượng bên ngoài dẫn đến quá trình quay, dao động của liên kết. Dùng xác định nhóm chức đặc trưng:

Tần số dao động của một vài liên kết thường gặp

- Phổ UV- vis: Dùng để xác định sự có mặt hay vắng mặt các nhóm riêng biệt trong vùng 200-400 nm.

- Một số nhóm đặc trưng: 

λmax < 150 (nm) liên kết σ



λmax : 200-260 (nm) nhóm benzen hoặc benzen thế



λmax > 280 (nm) hệ liên hợp

Phổ khối lượng: Sử dụng để đo khối lượng phân tử  xác định phân tử đặc biệt hoặc xác định các hợp chất chưa biết.

6. Kỹ thuật thử hoạt tính sinh học Ngày nay, phép thử sinh học rất mạnh, siêu nhạy thậm chí có thể chỉ cần sử dụng khối lượng chất thử rất nhỏ khoảng vài picogam (pg). Phần lớn các phép thử (sử dụng đĩa 96 hoặc 384 giếng) được tiến hành tự động hoặc bán tự động.

7. Tổng hợp hữu cơ Nhiều hợp chất thiên nhiên được sử dụng trực tiếp lâm sàng. Ví dụ: vinblastin (Velban), vincristin (Oncovin), epotosit (VP-16), paclitaxel (Taxol) được sử dụng trong điều trị ung thư; artemisinin, quinin để điều trị sốt rét...

Một số dẫn xuất bán tổng hợp từ hợp chất thiên nhiên được sử dụng trong lâm sàng như: vinorelbin (Navelbin), taxotere (Docetaxel), topotecan (Hycamtin) và irinotecan (Campostar) là các dẫn xuất của vinblastin, paclitaxel và camptothecin ưu việt hơn về hoạt tính, độ tan trong nước cũng như phổ tác dụng trong lâm sàng.
