letter from the president Friends, It’s hard to believe that it has been over a year since we lost my dad, and my how the world has truly changed in that time. Despite the ongoing pandemic, I know that my father would be proud to know that the mission continues as we accelerate our research in the safest and most expeditious ways possible. As we celebrate our 35th year, there is much to be proud of, but so much MARC BUONICONTI more to do to ensure that all our friends in wheelchairs president get their cure. By now everyone has heard about the promise that my dad made to me to do everything in his power to find a cure for paralysis. I carry that promise forward and will not rest until we succeed. The Miami Project, under the expert leadership of Drs. Barth Green and W. Dalton Dietrich, is well positioned to not only deliver for those who are paralyzed, but for all suffering from neurological injuries, diseases and disorders.

The revolution in neuroscience research that has been underway at The Miami Project these past three and a half decades has literally transformed the practice of medicine. The Miami Project, once a fledgling outfit, is now a Center of Excellence at the University of Miami Miller School of Medicine, and is internationally recognized as the preeminent research center for spinal cord and brain injuries. This achievement would have been thought impossible when I was first injured, but thanks to you and our loyal friends and donors we are closer to a cure than ever. And to add more good news to our story, I am happy to report that the Christine E. Lynn Rehabilitation Center for The Miami Project to Cure Paralysis at UHealth/Jackson Memorial is now open. It is now one of the country’s elite facilities for patients recovering from spinal cord and traumatic brain injury, and other complex conditions.

As you can see, The Miami Project and The Buoniconti Fund remain all-in in the quest to achieve our ultimate goal of curing paralysis. I want to thank everyone for their continued support and belief in our mission including the Great Sports Legends Dinner Chairman Mark Dalton, The Buoniconti Fund Board of Directors, the Mack family, Pepe Badia and Badia Spices, the Great Sports Legends, Honorees, our wonderful donors, tireless volunteers, Dinner attendees, our University of Miami President Dr. Julio Frenk and University of Miami Miller School of Medicine Dean, Dr. Henri Ford.

So as I reflect on these past 35 years, I do so with great pride because of the tremendous strides we’ve made in the field, but I know that the job is not finished yet. That knowledge is what fuels me and the rest of the team at The Miami Project each and every day and we will not rest until a cure is found.

THE MIAMI PROJECT co-founder It seems like yesterday that I was shaking hands with Nick Buoniconti and joining forces to create The Miami Project to Cure Paralysis. It was 35 years ago, shortly after his son Marc was paralyzed on the football field. Our focus was that the concept of “paralysis was forever” needed to be disproven. On that day, we committed to change medical and scientific history. Not by a prayer or dreams or hoping alone, but by creating good evidence-based research that could be translated into the clinics and operating rooms and ultimately become part of the practice of medicine globally. We have seen this happen at The Miami Project to Cure Paralysis.

Changing the practice of medicine or surgery is a goal that few physicians or surgeons have ever achieved. Today I am proud to claim this distinction, not as an individual, but as part of the prestigious Miami Project to Cure Paralysis team of scientists, clinicians, technicians, and graduate students who have worked together every day, 24/7 for the last 35 years to create a better life for people with catastrophic injuries and diseases of the nervous system. Yes, spinal cord injury is still our focus, but as part of that focus, it is natural that once you repair the spinal cord, you can also repair brain and peripheral nerve tissues because they are all anatomically and functionally connected.

Improving function, improving quality of life, improving opportunities, diminishing dependence and amplifying each individual’s independence has become our mantra!

Today we are not just excited, but supercharged with enthusiasm about our new collaborators from the Departments of Engineering and Rehabilitation Medicine, as well as our long-term teammates from numerous other basic and clinical sciences.

It’s truly a new era.

Is Covid-19 a challenge? Yes. Is global warming and our deteriorating environment a challenge? Yes. Is social unrest and racial injustice a challenge? Yes. However, despite all these challenges and difficulties that we all are facing in 2020, we have continued to work together, totally focused and committed to our mission.

Our short-term goal; to improve the quality of life for people with neurological injuries and diseases today.

Our long-term goal; to create treatments and cures for those disabling conditions.

Finally, we must always remember that it “takes a village.”

letter from scientific leaders The Miami Project family would like to first thank all of our friends, volunteers, and the spinal cord injury (SCI) community for their unwavering support for our multidisciplinary programs as we continue to advance our research initiatives. This year has presented many challenges for all of us that had to be overcome to ensure our community’s safety while remaining true to our research and education objectives. We know that COVID-19 has had serious consequences on many of our friends and loved ones. We send our best wishes while looking forward to a future of exciting discoveries, successful translational programs, and meaningful clinical advances.

Over the last several months, The Miami Project to Cure Paralysis, a Center of Excellence at the University of Miami Miller School of Medicine, has continued investigating and testing new treatments for improving outcomes after spinal cord and brain injury. To adhere to safety guidelines, including physical distancing requirements, our scientists and staff have developed new remote programs focused on safety, including virtual training, research, and outreach programs.

At-home exercise and rehabilitation programs are now available to our SCI community, which have enhanced safety, accessibility, and personalization. Our FDA-regulated cellular therapy trials have provided a wealth of new information on how stem cell and Schwann cell transplantation can be successfully conducted while providing a platform for testing new therapeutics. The recent discovery that extracellular vesicles (exosomes) may be able to induce reparative mechanisms has provided a new acellular approach for our clinical trial initiatives.

Innovative approaches that combine multiple treatments, including cell transplantation, neuromodulation, and activitybased therapy, are now being used to maximize recovery and improve whole-body health and function. An FDA-regulated cell-based clinical trial is actively recruiting participants to test a new peripheral nerve bridging strategy that may promote axonal regeneration and functional recovery. An innovative brain-computer interface technology that demonstrated efficacy in improving hand function in a research participant with SCI is launching industry partners. This coming year we plan to combine these various approaches with robotic-assisted motor rehabilitation and pharmacological therapies to augment recovery mechanisms. We continue to strengthen alliances with industry partners as we evaluate a new non-invasive electromagnetic neuromodulation technology to promote upper limb function. Based on preclinical findings, a first-in-man multi-center clinical trial is being proposed to improve function in people with chronic SCI. A critical component of The Miami Project’s mission is to conduct basic and translational research to enable future clinical advancements in neurological disorders. Thus, our expanded mandate incudes testing multidisclipinary strategies to repair the nervous system and restore function not only in SCI and Traumatic Brain Injury patients, but

letter from scientific leaders also for the millions of victims world wide suffering from the consequences of other neurological injuries and diseases. Our scientists utilize state-of-the-art imaging and drug discovery approaches to identify and test compounds that reduce harmful inflammatory mechanisms, improve circuit function and enhance regeneration of nerve fibers. Recently, a single molecule that simultaneously targets two important mechanisms for successful regeneration has been successfully tested in SCI injury models, signifying possible clinical translation. A new family of neuroinflammatory mediators of tissue destruction are being developed in our laboratories, which may be relevant in a range of neurological disorders where successful treatments are lacking. Our research programs also continue to target critical quality of life issues that impact our SCI community, including neuropathic pain, cardiovascular and autonomic dysfunction, muscle spasticity, and fertility.

Exciting new collaborative efforts with the Departments of Physical Medicine and Rehabilitation, Physical Therapy, and Biomedical Engineering are providing unique opportunities for improving the health and wellbeing of our constituents. Miami Project Educational and Outreach Programs reach out to thousands of individuals every year, providing current evidence-based information about SCI research and resources.

During the current COVID-19 pandemic, our remote summer research program inspired undergraduate students to pursue careers in neuroscience. We are in the final stages of planning our move to the new Christine E. Lynn Rehabilitation Center, a state-of-the-art facility where we will be able to provide even more opportunities for recovery and long-term health to our disability community and caregivers. We remain very excited about the future as we continue to build new collaborations with research and industry partners, and develop new treatments and technologies for improving functional recovery.

Green, M.D., world-renowned neurosurgeon, and Nick Buoniconti, NFL Hall of Fame linebacker, joined forces and founded The Miami Project to Cure Paralysis after Nick’s son, Marc, sustained a spinal cord injury during a college football game. Since then, research at The Miami Project has changed the landscape of knowledge and therapeutic strategies for spinal cord injury and traumatic brain injury.

Committed to finding a cure for paralysis resulting from spinal cord injury and to seeing millions worldwide walk again, the Buoniconti family established The Buoniconti Fund to Cure Paralysis in 1992. The Buoniconti Fund is a non-profit organization whose mission is to raise funds and awareness to help The Miami Project achieve its primary focus; to repair and restore function to the injured and diseased nervous system and thereby improving the quality of life of patients throughout the world.

The Miami Project, a Center of Excellence at the University of Miami Miller School of Medicine, is considered one of the premier research programs conducting cutting-edge discovery, translational and clinical investigations targeting traumatic spinal cord and brain injury, and other neurological disorders. The Miami Project’s international team of over 175 scientists, researchers, clinicians and support staff is dedicated to improving the quality of life of patients today and ultimately, finding a cure for paralysis. Miami Project researchers are conducting clinical studies and trials in spinal cord injury, including testing neuroprotective strategies, cellular therapies using Schwann cell and stem cell transplantation, and advanced rehabilitation and neuromodulation approaches including the use of brain machine interface technologies.

Other areas of current research include drug discovery for axonal regeneration and immune modulation, neuropathic pain, male fertility and cardiovascular disorders targeting their neurological impairments and those of numerous other nervous system diseases and injuries.

David “Big Papi” Ortiz is a Dominican-American former professional baseball (MLB) designated hitter (DH) and first baseman who played 20 seasons in Major League Baseball (MLB), primarily with the Boston Red Sox. He also played for the Minnesota Twins.

During his 14 seasons with the Red Sox, he was a ten-time All-Star, a three-time World Series champion, and a seven-time Silver Slugger winner. Ortiz also holds the Red Sox single-season record for home runs with 54, which he set during the 2006 season.

Ed Reed is an American former professional football player who was a safety in the National Football League (NFL), spending the majority of his career with the Baltimore Ravens. He was elected into the NFL Hall of Fame in 2019. He played college football for the University of Miami, where he was a two-time consensus All-American and National Champion. He was drafted by the Ravens in the first round of the 2002 NFL Draft and played 11 seasons for Baltimore before playing with the Houston Texans and New York Jets in 2013.

During his playing career, Reed was selected to nine total Pro Bowls, was the 2004 NFL Defensive Player of the Year Award winner, and has an NFL record for the two longest interception returns (106 yards in 2004 and 107 yards in 2008). After his retirement, Reed became a coach, serving as assistant defensive backs coach for the Buffalo Bills in 2016. In 2020, he became the Chief of Staff for the Miami Hurricanes football team.

MARIANO RIVERA Mariano Rivera is a Panamanian-American former professional baseball pitcher who played 19 seasons in Major League Baseball (MLB) for the New York Yankees, from 1995 to 2013. Nicknamed “Mo” and “Sandman”, he spent most of his career as a relief pitcher and served as the Yankees’ closer for 17 seasons. A thirteen-time AllStar and five-time World Series champion, he is MLB’s career leader in saves (652) and games finished (952).

Rivera won five American League (AL) Rolaids Relief Man Awards and three Delivery Man of the Year Awards, and he finished in the top three in voting for the AL Cy Young Award four times. He was inducted into the Baseball Hall of Fame as part of its class of 2019 in his first year of eligibility, and was the first player ever to be elected unanimously by the Baseball Writers’ Association of America (BBWAA).

TARA LIPINSKI Tara Lipinski is an American former competitive figure skater, actor, and sports commentator. A former competitor in ladies’ singles, she is the 1998 Olympic champion, the 1997 World champion, a two-time Champions Series Final champion (1997–1998), and the 1997 U.S. national champion.

She was, until 2019, the youngest skater to win a U.S. Nationals. She was also the youngest skater to win a World Figure Skating title and the youngest to win an Olympic gold medal. She is the first woman to complete a triple loop-triple loop combination, her signature jump, in competition.

ALEX RODRIGUEZ Alex “A-Rod” Rodriguez is an American former professional baseball shortstop and third baseman who played 22 seasons in Major League Baseball (MLB), as well as a businessman and philanthropist.

He played seven seasons with Seattle Mariners, three seasons with the Texas Rangers and twelve seasons with the New York Yankees.

Rodriguez is the chairman and CEO of A-Rod Corp as well as the chairman of Presidente beer.

Rodriguez began his professional career as one of the sport’s most highly touted prospects, and is considered to be one of the greatest baseball players of all time. Rodriguez amassed a .295 batting average, over 600 home runs (696), over 2,000 runs batted in (RBI), over 2,000 runs scored, over 3,000 hits, and over 300 stolen bases, the only player in MLB history to achieve all of those feats. He was also a World Series Champion, a 14-time All-Star, winning three American League (AL) Most Valuable Player (MVP) Awards, ten Silver Slugger Awards, and two Gold Glove Awards. Rodríguez is also the career record holder for grand slams with 25. He signed two of the most lucrative sports contracts in baseball.

Dr. Eugene J. Sayfie Some say no one is perfect, but the multitudes who knew Dr. Eugene J. Sayfie would argue otherwise.

An old-fashioned doctor with a cutting-edge mind, Dr. Sayfie left every place he visited and every person he met better than he found them. He carried his doctor’s black bag with him everywhere and was rarely seen without a stethoscope around his neck. Every patient had his personal cell phone number and he never knew the meaning of ‘working hours’ because he was always on call, for anyone in need, meeting each person on his path with enthusiasm, optimism and a readiness to help in any way he could.

He often quoted Khalil Gibran’s The Prophet, “Work is love made visible” and he was renowned for his steadfast commitment to his patients and his incomparable work ethic. “My view has always been that we are here to serve the patient first and foremost,” said Dr. Sayfie at the dedication of The Eugene J. Sayfie, M.D., Pavilion for Excellence in Patient Care at the University of Miami Miller School of Medicine.

Dr. Sayfie measured life by two standards: the first- whether you enjoyed the journey, and the second- did you make a difference to others along the way. Dr. Sayfie exceeded all measures on both. Everyone left his side feeling better about the world and better about themselves. His optimism and enthusiasm were infectious and will be an enormous part of his legacy.

Dr. Sayfie’s greatest passions were his loving family, his practice of medicine, his devoted faith, humanitarianism, excellence in education and philanthropy.
