MONOCLONAL ANTIBODY IgM antibody anti HCV NS3 helicase region CRO Aviano researchers have developed a monoclonal antibody presenting a crossreactivity toward both an immunodominant region of the HCV-NS3 helicase (NS31251-1270) and the IgG-Fc region (Fc345-355). The same cross reactivity characterized the IgM component of the cryoprecipitate in a HCV positive patient affected by type II mixed cryoglobulinemia syndrome complicated by a concomitant immunocytoma (De Re V. et al. Leukemia 2006). This antibody has been successfully used in ELISA assays against both the above mentioned peptides (NS31251-1270 and Fc345-355) and also against NS3 and Fc intact antigens. Moreover it presented, in ELISA, a Rheumatoid Factor activity. Ribbon representation of dimeric structure of IgG Fc fragment (pdb code=1fc1). Ribbon representation of HCV NS3 helicase domain structure (pdb code=1a1v). The protein fragments identified by epitope excision method with the IgM of patients 1 (red color), 3 (green) and 5–7 (blue) are localized in region exposed to the solvent. Antibody anti-HCV-NS3 were produced against the HCV-NS3 31251-1270 peptide.
