Mesalamineenema is used to treat active mild to moderate ulcerative colitis, proctitis or proctosigmoiditis (inflammation of the rectum and bowel). Mesacol Rectal Suppositories Dosage The dosage of Mesacol Rectal Suppositories (Generic Mesalamine Rectal Suppositories) is given below: The recommended dosage of Mesacol Rectal Suppositories is one rectal suppository administered rectally once daily at bedtime for 3 to 6 weeks.

The suppository must be retained for one to three hours or longer, if possible. The usual course of therapy is from three to six weeks depending on symptoms and sigmoidoscopic findings. Safety and effectiveness beyond 6 weeks have not been established. In case a patient misses a dose of Mesacol, it should be administered as soon as possible. However, if it is almost time for your next dose, skip the missed dose and go back to your regular dosing schedule. Do not double doses.

Storage Instructions

Mesacol Rectal Suppositories (Generic Mesalamine Rectal Suppositories) have to be stored at controlled room temperature i.e. from 20°C to 25°C (68°F to 77°F) with excursions permitted between 15° to 30°C (59° to 86°F).

Warnings

Renal Impairment: Evaluate the risks and benefits in patients with known renal impairment or taking nephrotoxic drugs; monitor renal function.

Mesalamine-Induced Acute Intolerance Syndrome: Symptoms may be difficult to distinguish from an exacerbation of ulcerative colitis; monitor for worsening symptoms; discontinue treatment if acute intolerance syndrome is suspected.

Hypersensitivity Reactions, including Myocarditis and Pericarditis: Evaluate patients immediately and discontinue if a hypersensitivity reaction is suspected.

Hepatic Failure: Evaluate the risks and benefits in patients with known liver impairment.

Warnings Photosensitivity: Advise patients with pre-existing skin conditions to avoid sun exposure, wear protective clothing, and use a broad-spectrum sunscreen when outdoors.

Nephrolithiasis: Mesalamine-containing stones are undetectable by standard radiography or computed tomography (CT). Ensure adequate hydration during treatment.

Interaction with Laboratory Test for Urinary Normetanephrine: Spuriously elevated test results may occur with liquid chromatography with electrochemical detection in patients receiving Mesacol Rectal Suppositories; use alternative, selective assay for normetanephrine.

Mesacol Rectal Suppositories Side Effects

The most common Mesalamine Rectal Suppositories side effects with an incidence rate of 1% or greater are: dizziness, rectal pain, fever, rash, acne and colitis.

Tell your doctor immediately in case any of these rare but very serious side effects occur: chest pain, shortness of breath, severe stomach/abdominal pain (especially if spreading to the back), yellowing eyes/skin, dark urine, signs of kidney problems (such as change in the amount of urine).

A very serious allergic reaction to Mesacol is rare. However, seek immediate medical attention if you notice any symptoms of a serious allergic reaction, including: rash, itching/swelling (especially of the face/tongue/throat), severe dizziness, trouble breathing.

Overdose Mesalamine absorption from the colon is limited; however, Mesacol is an aminosalicylate, and symptoms of salicylate toxicity include nausea, vomiting and abdominal pain, tachypnea, hyperpnea, tinnitus, and neurologic symptoms (headache, dizziness, confusion, seizures).

Severe salicylate intoxication may lead to electrolyte and blood pH imbalance and potentially to other organ (e.g., renal and liver) involvement.

There is no specific antidote for Mesalamine overdose.

Correct fluid and electrolyte imbalance by the administration of appropriate intravenous therapy and maintain adequate renal function.

Mesacol Rectal Suppositories During Pregnancy

Mesalamine (Mesacol Rectal Suppositories) is classified by the US FDA as Pregnancy Category B. Limited published data on mesalamine use in pregnant women are insufficient to inform a drug-associated risk. No evidence of teratogenicity was observed in rats or rabbits when treated during gestation with orally administered Mesalamine at doses greater than the recommended human intra-rectal dose.

There are, however, no adequate and well controlled studies in pregnant women. Since animal reproduction studies are not always predictive of human response, this drug should be used in pregnancy only if clearly needed.
