We are so proud that this family entrusted us with the care of their two sons. Ten years ago, Jack (center) lost a kidney to cancer and underwent treatment at Children’s Health. A few years later, his older brother, Matthew, had brain surgery to correct a malformation that was putting pressure on his spinal cord.

AHEAD OF THE GAME In December, 10-year-old Xavier Sanchez was knocked unconscious in a car accident and rushed to Children’s Medical Center Dallas. His mother, Adriana Angel, was deeply worried, but said she could take some solace in the fact that so many doctors were tending to him.

“I am talking about doctors everywhere,” the Dallas mom said. “They were completely prepared.”

Brittany Banks, of Waxahachie, said she had a similar experience when she took her son, Andre, to Children’s Medical Center Dallas after he fell and bumped his head when he was 5 months old. He was not released until four doctors had examined him.

“Their focus was solely on him,” Mrs. Banks said. “I was just really in awe of the service. …

They were very thorough.”

The intense attention that these mothers witnessed is the standard of care at Children’s Health no matter the illness or injury, but particularly so when it comes to managing concussions. Careful examination is important because kids, and even parents, can be too

quick to say everything’s fine, and it’s time to get back in the game.

Concussions have recently received tremendous media attention, much of it spurred by the fact that experts now have much better insights – sobering insights – into just how damaging some of these head injuries can be. And, as more and more parents, coaches and even kids themselves recognize the symptoms, hospitals are seeing a spike in concussion patients. Children’s Health experts predict that they will see more in the coming years, and they are ready with many new initiatives.

Children’s Health Rehabilitation and Therapy Services each have experts and new ways of assessing and treating concussions. This renewed focus on traumatic brain injury has brought Children’s Health its first postdoctoral fellow in headache and concussion, and collaboration with UT Southwestern Medical Center is accelerating concussion research on multiple fronts. The Children’s Health Andrews Institute conducts educational outreach specifically about concussions with thousands of high school coaches and athletic trainers, and it is in the process of acquiring new equipment for concussion patients and recruiting a neuropsychologist to work exclusively with these patients.

“Kids need to be better taught how to tackle correctly,” he said. “I just don’t think kids are physically ready for tackle football until they’re in adolescence.”

The need is great. North Texas’ young population is growing, and Texas celebrates high school football probably more than any other state.

“It’s part of the culture, part of the fabric of Texas quite honestly, and we deal with it as best we can,” said Dr. Robert Rinaldi, who, as division chief of pediatric rehabilitation medicine at Children’s Health, is expanding

But that cannot be mandated – at least not yet. When the junior high school tackle football team was disbanded in Marshall, Texas, two years ago, parents and students who love the sport simply splintered off to form their own league. Nothing is going to stop millions of kids from being active and pursuing sports. That’s where education comes in, and the Children’s Health Andrews Institute is taking the lead.

“I speak at several coaches groups – not just for football but for all sports – several times a week,” said Jeff Baggett, the outreach program manager at the Children’s Health Andrews Institute. He recently presented to hundreds of football coaches with USA Football in Dallas, Houston, Waco and San Antonio, and he presented to athletic trainers at a DFW Coaches Clinic in January.

Mr. Baggett said coaches seem primarily interested in how to return athletes safely to their sport, but there’s no question that almost all of them have accepted concussions as a serious issue.

“No sport is without risk. We’re working on ways to make things safer,” Mr. Baggett said, explaining that it’s not just the high-profile football games that cause concussions but also the unhelmeted sports of basketball, where players are getting elbowed in the head; lacrosse, where midfielders are getting smacked with the ball; and cheerleading where girls are smashing into the hard gym floor.

When the Children’s Health Andrews Institute opened a concussion clinic at the Children’s Health Plano campus in the fall of 2016, doctors saw about 40 concussion patients a week.

That number is expected to double this fall, according to sports medicine pediatrician Dr. Troy Smurawa.

As director of pediatric sports medicine for the Children’s Health Andrews Institute, Dr. Smurawa leads a team that treats concussion patients, along with another physician, a nurse practitioner, physical therapists and athletic trainers. They’re recruiting a neuropsychologist to join the team. A full-court press is needed to treat this growing problem, he said.

“Concussion affects the brain’s ability to function on different levels, anywhere from cognitive levels to emotional levels to balance,” Dr. Smurawa said. His description of what happens in a concussion is even more unsettling: Imagine the brain bashing up against the inside of the skull. With only spinal fluid to cushion it, the brain can even bounce inside the skull and harmfully bang up against the inner cranium more than once. If the body is jarred violently enough, as it often is in sports, the brain can twist, pulling the two hemispheres away from each other.

“Severe cognitive damage could occur. It could last from minutes to a lifetime,” Dr. Smurawa said.

The Childnren's Health Andrews Institute has special equipment to measure concussion patients’ balance, and the clinic recently purchased a cutting-edge machine with sensors that attach to the patient’s head and measure eye movements. The patient’s abilities in areas such as balance and cognition are carefully monitored as doctors try to treat symptoms like headaches and map out a plan for the student athlete to return safely to normal activities.

But concussions are not limited to only athletes. Many children suffer from damaging blows to the head because they have smaller, weaker neck muscles to cushion such a blow, according to Dr. Tonia Sabo, a UT Southwestern assistant professor of pediatrics, anesthesiology and pain management, and neurology and neurotherapeutics.

In her role as medical director of concussion services, Dr. Sabo and her clinical team make up the Neuro-Concussion branch of the comprehensive Children’s Health Concussion Program. Her clinic sees local and statewide concussion referrals – typically the children who may not be recovering well from a concussion or who have specific neurological issues related to a mild traumatic brain injury.

on Transcranial Doppler ultrasound when specifically looking at cerebrovascular reactivity. If the patient’s brain is still in a fragile state, she said, it may not be safe for that patient to resume contact sports. She received partial funding through Children’s Medical Center Foundation, which will enable her and her trainee to begin the certification process to bring this technology to the clinical setting.

Her clinic also uses state-of-the-art equipment for diagnosis and treatment of concussion.

She is specifically interested in radiographic diagnosis of concussion using techniques such as functional magnetic resonance imaging and diffusion tensor imaging, and physiological diagnosis using Transcranial Doppler. Most recently, she presented a paper at the International Brain Injury Association meeting, where she detailed specific cases of post-traumatic headache that have responded to specific treatments for mildly high cerebrospinal fluid pressure.

It is cutting-edge technology such as this that puts Children’s Health and UT Southwestern at the forefront of concussion management and diagnosis, Dr. Sabo said.

She is a co-investigator on two currently enrolling concussion studies looking at these techniques in both high school and college-age students. She said that a patient who may be “asymptomatic” after several weeks following a concussion could still show abnormalities

She’s also involved in a statewide initiative for concussion reporting along with a multidisciplinary team at UT Southwestern.

Board-certified in child neurology and headache medicine, she previously worked for seven years as an assistant professor of pediatrics and neurology at Colorado Children’s Hospital/University of Colorado Health Sciences Center, where she created a comprehensive headache program and treated several Olympic athletes.

“Concussions have always been around,” Dr. Sabo said. “But now we’re realizing how much these smaller injuries can add up to potentially cause significant issues, especially for a child

The clinic is also where Children’s Health’s first pediatric headache and concussion fellow, Dr. Eric Remster, a pediatric neurologist, is studying specific cases of pseudotumor cerebri (which can be seen after concussion) and Transcranial Doppler. The clinic is scheduled to grow fourfold as it accommodates its growing number of patients, many of them concussion patients, and moves into the new Children’s Health Specialty Center Cityville.

for whom brain development and maturity is occurring. Twenty percent of athletes in high-contact sports will have some sort of concussion injury. If you get one, you are three times more likely to have another that season.”

Much education has taken place in recent years, Dr. Sabo said, with schools and athletic governing bodies realizing the importance of watching for the warning signs of a concussion: headaches, neck pain, nausea, sensitivity to light, confusion, slurring of speech and even stuttering.

Dr. Sabo works to provide families with guidance. She loves working with kids – teenagers in particular – because she believes she can make a true difference for them.

“It’s very motivating to see them telling the coach, ‘I want to protect my health.’”

Last year, more than 5,000 patients were seen in the Children’s Health Emergency Department and affiliated clinics for the diagnosis of concussion or related concussion sequela. In addition to working with young athletes, Dr. Sabo also treats children who have been in accidents or experienced other trauma.

One of the patients was Devon Green, a Dallas 10-year-old who suffered a concussion when he fell off a stool and hit the back of his head on the hard floor. What followed next were some of the classic signs of a concussion: He felt dizzy, couldn’t stand and vomited in the school nurse’s office. Two days later he still felt dizzy and was taken to Children’s Medical Center Dallas. The bad news was that Devon would have to sit out the rest of his pee-wee football season. The good news was that

Dr. Sabo had a plan of rest that could – and did – help him reach full recovery.

His recovery was tracked by carefully monitoring important physical and cognitive abilities. A nurse took notes as she asked Devon to recite words and numbers and then recite them backwards. Then she had him stand on a wobbly mat, feet together, then one foot in front of the other, then standing on just one foot with his arms out and eyes closed. His toes griped the mat, and his eyes darted back and forth under his eyelids as he struggled to maintain his balance. He grumbled under his breath that his exclusion from his pee-wee football team cost his team the championship.

The nurse left the exam room and entered data into a huge, growing database of concussion patients that is being administered by Dr. Munro Cullum, who is chief of psychology at UT Southwestern Medical Center and holds the Pam Blumenthal Distinguished Professorship in Clinical Psychology. Dr. Cullum maintains a detailed North Texas database and a statewide database.

“There’s so much we don’t know,” Dr. Cullum said. “We don’t know right now who is going

to be at risk for prolonged recovery from concussion, and that is a huge research area.

Roughly 10 percent to 15 percent still have symptoms weeks later, sometimes months later, and we can’t identify who those are going to be yet.”

He said the growing database will be used to try to answer these questions and identify best practices.

Devon, the would-be pee-wee champ, is in the database. After the nurse typed in his test results, Dr. Sabo came into the exam room and asked him similar questions. She held his head in her hands to examine it and asked if he felt back to his old self. “Yes,” he said.

“Is that the truth?” she asked. “Because sometimes we get kids who want to go back to play, and they don’t always tell us everything.”

Devon said it was true. Dr. Sabo said all signs showed he was right.

“Does this mean I can go back to sports and stuff?” he asked.

“Yes.” n

BACK IN THE SADDLE AGAIN Following a devastating head injury seven years ago, Baylee Neal is riding high

Few sights are more Texan than an expert rider on horseback galloping through the Texas countryside, so it was impressive to see 18-year-old Baylee Neal take the reins – especially impressive because a riding accident seven years ago nearly cost Baylee her life.

On hand to watch her ride recently was Dr. Darryl Miles, a critical care doctor at Children’s Medical Center Dallas and an assistant professor of pediatrics at UT Southwestern Medical Center, who guided Baylee from being completely unresponsive – unable to even breathe on her own – to making a recovery that enabled her to get back in the saddle and go on to college. Dr. Miles took a day from work to drive out to rural East Texas to reunite with the family with whom he had spent many difficult days to see his patient ride again after a hardfought recovery.

When Baylee returned to her normal routine after her injury, she had to overcome learning

difficulties caused by the accident, but nevertheless wanted to give back to others by participating in Dr. Miles’ research study on head injuries.

She was 11 years old when she suffered one of the worst concussions imaginable. Baylee was barrel racing when the saddle slipped, and her foot came out of the stirrup. She lost her balance and leaned too far over the side.

When her horse ran past a metal post, Baylee’s head smashed into it – hard. She was knocked unconscious.

She was transported to Children’s Medical Center Dallas, which has the state’s first pediatric Level 1 Trauma Center. In the emergency room, Baylee’s father, Jeff Neal, turned her mother, Jennifer, away so she would not see doctors insert a breathing tube. Baylee was then sent to the ICU, where Dr. Miles found her completely unresponsive. He had to deliver the terrible news to Baylee’s parents that she might never open her eyes again.

“He’s a great explainer, but it was not really the things that you want to hear,” Mrs. Neal said. “It was serious. It was a very serious time.”

Pediatric neurosurgeons inserted an external ventricular drain (EVD) to the center of her brain to drain fluid and monitor brain pressure.

“Her first scan showed there was already some mild swelling. Within 12 hours we repeated the scan, and it showed that swelling was significant. Her clinical picture also seemed to be deteriorating,” Dr. Miles recalled. “She didn’t have room to get worse.”

to help others who would suffer injuries similar to hers by participating in Dr. Miles’ study for five years. The study, funded by the Perot Center for Brain and Nerve Injury at Children's Health, required Baylee and other patients in the study to undergo neuropsychological testing of IQ, memory, how quickly they could process information, and other measures.

To Dr. Miles, it is valuable information that he hopes can be used to design better therapies for children who suffer severe concussions and traumatic brain injuries. To Baylee, it was testing that gets to the core of her struggles.

“The hardest thing about having short-term memory loss is figuring out strategies to memorize important facts,” Baylee said. “I was able to take the strategies I had learned from Our Children’s House and speech therapy and apply them to learning the skills I use today.”

Physical, occupational and speech therapy at Our Children’s House taught her simple but effective memory techniques of writing things down and repeating them out loud over and over again. By her senior year, she had a 3.7 GPA. She’s currently studying animal science at Oklahoma State University, and she is back to her favorite activity, riding horses.

“My fear of barrel racing has yet to be let go, but I still believe that everything happens for a reason,” she said. “Because of this, I will continue to race, and I will continue to fight.” n

The medical team temporarily put her in a coma, and the EVD was left in for eight days.

Finally, she made a comeback. She opened her eyes. She responded with nods, smiles and hand squeezing. Then one day she spoke asking, “Where’s Mom?” By the time she left the ICU, she was, incredibly, speaking more than patients with less severe head injuries.

It was an encouraging comeback, but returning to schoolwork was a huge challenge.

“I could only make it through a couple of class periods at a time before having to go home due to exhaustion,” Baylee said. “This pushed me to the point of almost completely giving up.”

No one she spoke to could fully understand what she was going through – but she sought

With the symbolic snip of a giant red ribbon, the new Rees-Jones Center for Foster Care Excellence at Children’s Medical Center Dallas officially opened Dec. 5.

Texas Gov. Greg Abbott joined Christopher J. Durovich, president and CEO of Children’s Health; Dr. Anu Partap, director of the new

Center; Dr. Heidi Roman, ambulatory medical director and assistant professor of pediatrics, and Jan and Trevor Rees-Jones for the ribbon cutting. Only five months earlier, a new foster care clinic had opened at Children’s Medical Center Plano, allowing even more children in foster care to have access to specialized care.

There are approximately 7,000 children in the Dallas-Fort Worth area in foster care, and one in eight has high medical, behavioral, emotional, intellectual and developmental needs. For Gov. Abbott, the opening of the Center offers hope to the 40,000 children in foster care across the state.

"The Rees-Jones Center provides the childcentric care that our families and communities need to meaningfully advance our foster care system," said Gov. Abbott. The new clinic spaces were funded by a $20 million gift from the Rees-Jones Foundation – the largest gift ever from a living donor in the history of Children’s Health – as well as six-figure gifts from the Women’s Auxiliary to Children’s Medical Center, the Meadows Foundation, Rhonda and Charles Koetting, the Lupe Murchison Foundation and the George and Fay Young Foundation.

“It’s one of the most important goals of our foundation: the defense and healing of children who suffer from abuse, neglect, family violence, or physical or mental disability,” said Trevor Rees-Jones. “Many of these children are in long-term foster care. This new Center gives us hope for the healing of these children and gives us great comfort that they will be receiving the highest quality care available.”

Mr. Rees-Jones called Dr. Partap a “gamechanger.” It was in part her vision that guided the concept and design for the Center.

“The community rallied and did this for our kids, and it is so overwhelming,” said Dr. Partap, who is also an assistant professor of pediatrics at UT Southwestern Medical Center. “It is gratifying to be part of the opening of such an exceptionally beautiful and therapeutic welcoming space for kids. Everyone who had a hand in it went above and beyond.”

Dr. Partap’s passion for serving children in foster care started in medical school when she noticed that if patients were living with family violence, it undermined their health and ability to get better. She said victims of abuse or neglect need to be treated the same as those with other medical conditions, and they need an intensive treatment plan so they are able to live a full life.

The Rees-Jones Center for Foster Care Excellence is the only place in North Texas with an integrated approach. It is designed to become a model of foster care excellence across the state and nation by working to develop best practices for other programs to replicate.

“Children frequently enter foster care with complex medical issues, including drug withdrawal; discomfort from neglected health care; the inability to talk, walk or learn; or brain damage from abuse,” Dr. Partap explained.

None of this would be possible without the incredible generosity of the ReesJones Foundation and the passionate advocacy of each and every one of our team members.”

The Rees-Jones Center brings together experts in pediatric medicine, behavioral health, research and child welfare to deliver rehabilitative, recovery-focused care. In addition to medical, behavioral and mental health care, the ReesJones Center provides on-site Child Protective Services liaisons to coordinate patients’ care with other state agencies as needed.

Children in foster care are on every spectrum of special health care needs, ranging from birthrelated conditions, such as prematurity, fetal alcohol exposure, cerebral palsy and autism, to developmental and learning delays from abuse or neglect. The toxic stress they have been living with often sets off an inflammatory response, including asthma or skin problems.

The children also are living with people they don’t know and moving around, which adds another dimension in a transitional period of life.

Dr. Partap is working to implement a system in Texas in which children who enter foster care are seen by a team of medical professionals within three days. The current standard is 60 days. She has been instrumental in bringing the Texas foster care system to the forefront of legislative focus. In March and April, she testified before the Senate Health and Human Services Committee and the House Select Committee on Mental Health.

It only takes a

SPARC

The Suicide Prevention and Resilience in Children (SPARC) program is one of only two intensive outpatient treatment programs in the nation for suicidal teens, and so far 400 vulnerable patients have accessed the program and found help.

Lily Allen found herself hospitalized in Children’s Medical Center Dallas’ psychiatry program in 2015 after she texted a friend that she wanted to end her life.

“The hardest point was waking up my first day there. I woke up at noon, and everyone was eating lunch,” Lily said. “It was a ton of faces I had never seen before, and it was really scary.

That was probably the hardest because I felt completely alone.”

But she wasn’t alone. The Dallas 14-yearold quickly found out that those strangers – both staff and patients – were people who understood what she was going through.

The staff surprised her by keeping her on a busy schedule of activities and group therapy that was helpful. Progress could have come to a dangerously abrupt halt when she checked out of the hospital after eight days, but because she was at Children’s Health, she was fortunate to have access to one of only two intensive outpatient treatment programs for suicidal teens in the nation: Suicide Prevention and Resilience in Children, or SPARC. The program worked for her as it has for more than 400 other teens in North Texas because it is based on nationally respected research, much of it conducted at Children’s Health.

In 2011, an alarming spike in teens showing up in the emergency room after suicide attempts alerted hospital staff that there was a problem.

population nationwide. Suicide is the secondleading cause of death among people ages 10 to 24, according to the Centers for Disease Control and Prevention. Only accidents take more lives for this wide age demographic.

But pediatric psychiatrists have, until recently, been ill-equipped to deal with the crisis due to lack of research-based treatment; patients under the age of 18 were often excluded from studies using antidepressants.

Children’s Health and UT Southwestern Medical Center joined four other prestigious universities in a National Institute of Mental Health study examining the outcomes of medication and therapy on suicidal teenagers.

The study found that a specific intervention for adolescents at high risk for repeated attempts was effective in treating suicidal

thoughts and behaviors, and that cognitive behavioral therapy with medication was slightly better.

The results were published in a 2009 issue of the Journal of the American Academy of Child & Adolescent Psychiatry. The authors were some of the country’s leading experts, including UT Southwestern professor of psychiatry Dr. Betsy Kennard, who is director of outpatient psychological services at Children’s Health, and Dr. Graham Emslie, division director of pediatric psychiatry at Children’s Health and professor of psychiatry and pediatrics at UT Southwestern. Dr. Emslie holds the Charles E. and Sarah M. Seay Chair in Child Psychiatry at UT Southwestern.

The study was a landmark. Many other researchers took the study’s data set to

produce additional studies. Children’s Health clinicians took the study and put it to work.

There were not enough resources to treat individually the huge number of patients who had attempted suicide, so Dr. Kennard thought innovatively and used what the study found to be successful in one-on-one therapy to build modules for similar sessions in group therapy.

Senior director of psychiatry for Children’s Health Jeanne Nightingale said the hospital administration immediately understood that the need was urgent, and they threw their weight behind it.

“They said, ‘Go ahead and do it, and hire the people you need,’ ” she said. “It was unbelievable. They said, ‘This is an important issue. Let’s see what we can do.’ ” This was the beginning of SPARC, which has suicidal teenagers participating in intensive outpatient group therapy sessions three times weekly with their peers, as well as individual therapy and family therapy. Some of the techniques in the group sessions include problem-solving and steps to prevent relapse, to exercise more control over emotions, and to communicate with family.

“The results are equivalent to or better than the study that we did – and that was better than what we had before. The study was considered a success because that specific suicide treatment showed a decrease in additional suicide attempts,” Dr. Emslie said. “We can do the same treatment in a group format equally effectively, maybe slightly better.”

transition program and to continue to evaluate the program’s results. Other donors have contributed more than $500,000 over the past two years to augment the staffing of the suicide treatment and prevention program.

Notably the Bezalel Fund, a donor advised account administered by the Charles and Lynn Schusterman Family Foundation at the Vanguard Charitable Endowment Program, pledged $5 million in December 2016 toward mental health and suicide prevention.

A smaller, but similarly meaningful, gift came from another source, from Lily, the 14-yearold who woke up in Children’s psychiatric program and didn’t know anyone there.

Now a 15-year-old sophomore at Booker T. Washington High School for the Performing and Visual Arts, Lily took action – as she had learned in SPARC’s group therapy sessions.

She organized 50 of her fellow dance students at the arts magnet high school into a 2016 dance performance benefiting SPARC, and she raised more than $1,200 for the program.

She said it was important to her to give back to a program that will help teens like her, teens who will look out into a sea of strange faces sitting at the lunch table.

“I always thought being depressed was something I was going to have to live with forever, and it was going to basically ruin my life,” she said. “But because of SPARC and because of Children’s, I found out that I don’t have to live this sad life forever. There was hope of getting better.” n

While the numbers show results, Dr. Kennard said she has seen bonding and support take shape among the teens as they share their struggles in the group therapy sessions.

“They give each other good feedback and advice,” she said. “The kids will tell us one of the things they like about SPARC is they realize they are not alone, that there are other people who have the same kind of problem.

Drs. Kennard and Emslie are starting to study the biomarkers of suicide risk with colleagues at UT Southwestern, and the program is also attracting more funding. In 2016, UT Southwestern created a version of SPARC for Hispanic females, and The Children’s Trust awarded SPARC a recent grant to develop a

These milestones from 2016 are just a sampling of innovative, groundbreaking projects and studies taking place at CRI.

RESEARCH REVERSING OSTEOPOROSIS WITH A NEW BONE-FORMING GROWTH FACTOR Osteoporosis, a progressive bone disease characterized by decreased bone mass and an increase in fractures, affects over 200 million people worldwide. Most existing therapies reduce the rate of bone loss, but they do not promote new bone growth.

Research conducted in Dr. Sean Morrison’s lab last year uncovered a new bone-forming growth factor, Osteolectin (Clec11a), which promotes bone growth and reverses bone loss after the onset of osteoporosis in mice.

Deletion of Osteolectin in mice accelerates bone loss during aging, reducing bone strength and delaying fracture healing.

“These results demonstrate the important role Osteolectin plays in new bone formation and maintaining adult bone mass. This study opens up the possibility of using this growth factor to treat diseases like osteoporosis,” said Dr. Sean Morrison, director of the CRI, principal investigator of the Hamon Laboratory for Stem Cell and Cancer Biology, and holder of the Kathryne and Gene Bishop Distinguished Chair in Pediatric Research and the Mary McDermott Cook Chair in Pediatric Genetics. He is also an investigator in the Howard Hughes Medical Institute and a professor in the CRI and of pediatrics.

These early results are encouraging, suggesting Osteolectin might one day be a useful therapeutic option for osteoporosis and in regenerative medicine. These results were published in the scientific journal eLife in December.

MAPPING METABOLIC PATHWAYS IN CANCER TO IMPROVE DIAGNOSIS AND TREATMENT Proper control of metabolism is required for essentially every basic biological process, including cell growth and division. Altered cellular metabolism is a hallmark of cancer, as cancer cells must reprogram their metabolism to support excessive cell growth. Finding and understanding how these growth-promoting metabolic changes occur in cancer is a key step toward developing new anti-cancer therapies.

In 2016, researchers from Dr. Ralph DeBerardinis’ lab made two important discoveries. In February, the DeBerardinis team pioneered techniques to measure cancer metabolism directly in human tumors, providing the most comprehensive and accurate views of tumor metabolism to date.

Traditionally, lung tumors are diagnosed using a clinical form of glucose imaging called FDG-PET scanning because increased glucose (sugar) uptake is a well-known feature of human lung cancer cells. It was unknown, however, how these tumors use glucose to fuel their metabolism.

The DeBerardinis laboratory scientists mapped the fate of glucose in human lung tumors and identified several metabolic differences between tumors and healthy lung tissue. They also showed that individual tumors, and even regions within the same tumor, used strikingly different metabolic activities to generate energy.

The team also demonstrated that a magnetic resonance imaging technique could predict metabolic behavior in tumors before surgery, providing a completely new approach to study metabolism in human cancer. This work is expected to lead to better ways to exploit metabolic alterations to improve cancer diagnosis and therapy, especially when applied to childhood cancers.

Dr. DeBerardinis is director of the CRI Genetic and Metabolic Disease Program, a professor in the CRI, in the Eugene McDermott Center for Human Growth and Development, and of pediatrics. He holds the Joel B. Steinberg, M.D., Chair in Pediatrics and is a Sowell Family Scholar in Medical Research.

In April, scientists from the DeBerardinis lab identified a novel metabolic pathway that helps cancer cells thrive in conditions that are lethal to normal cells. Researchers found cancer cells use an alternate version of two well-known metabolic pathways to adapt to

the stress associated with cancer progression and defend against the reactive oxygen species that kill cells via oxidative stress. Most normal cells are killed after detaching from a support structure called extracellular matrix, because attachment to the matrix keeps oxidative stress at manageable levels. The DeBerardinis lab team found that cancer cells have the ability to activate a novel pathway that counteracts oxidative stress after matrix detachment. This allows them to survive and grow under such conditions.

This discovery has the potential to lead to new cancer therapies, particularly therapies aimed at preventing cancer metastasis, which requires that cells survive a period of matrix detachment. Ongoing work will determine if this metabolic pathway, so far characterized in cultured cancer cells, is required for metastasis in animal models of cancer.

IMPROVING OUR UNDERSTANDING OF THE LINK BETWEEN CANCER AND TISSUE INJURY Chronically damaged tissues are more likely to develop cancer, but it is unclear if tissue regeneration itself promotes or inhibits the development of cancer. Understanding the relationship between injury, regeneration, and cancer is key to discovering new treatments that could promote tissue regeneration and prevent or delay cancer.

Research from Dr. Hao Zhu’s lab in 2016 provided new insights into the link between tissue regeneration and cancer in the liver. In humans, a gene called ARID1A is inactivated in several cancers, including liver cancer. Based on this, the researchers hypothesized that mice lacking Arid1a would develop liver damage and, eventually, liver cancer. They were surprised when the opposite proved to be the case – no liver damage occurred. In fact, livers of the Arid1a-deficient mice regenerated faster and appeared to recover better.

“The livers were resistant to tissue damage and healed better, which are two good things – like playing offense and defense at the same time,” said Dr. Zhu, assistant professor in the CRI and of pediatrics and internal medicine.

“These results opened up a whole new avenue of investigation for us, and through that investigation, we found a new function for this gene.” This raised the possibility that there are mechanisms that both promote tissue regeneration and inhibit the development of cancer.

The Zhu laboratory is currently funded by the Cancer Prevention and Research Institute of Texas (CPRIT) to develop a drug that would inhibit the function of Arid1a. The long-term hope is that such a drug will increase liver regeneration and inhibit the development of cancer.

GRANTS AND AWARDS Two CRI faculty members were awarded prestigious grants in 2016. Dr. Hao Zhu was one of 10 researchers in the nation to receive a Stand Up To Cancer grant. This grant is awarded to early-career scientists to support innovative, high-risk, high-reward projects in cancer research. Dr. Zhu is using this grant to fund research that explores the relationship between organ regeneration and cancer formation.

The lab will study mice that have a defect in a gene called Arid1a, which protects against liver cancer and promotes liver regeneration.

In September, Dr. Ralph DeBerardinis was among 84 scientists from 43 U.S. institutions chosen as a Howard Hughes Medical Institute Faculty Scholar. The program, in its first year, awarded grants of $600,000 to $1.8 million over five years to each early-career scientist. The HHMI Faculty Scholars were selected for their potential to make unique contributions to their fields. Dr. DeBerardinis plans to use his grant to further his research in cancer metabolism. n

EVENTS 2016 RED BALLOON RUN & RIDE For the sixth year, North Texas families laced up their running shoes and adjusted their bike chains to participate in the signature fundraising event for Children’s Health. More than 15,000 attendees converged on the Children’s Health Plano campus for a day of exercise, family activities and food. From a 1K fun run to a 72-mile bike ride, there was something for everyone at the event, including activities for children and families. The Run & Ride is produced by Passion for Children’s for the benefit of Children’s Health. Capital One and Denbury Resources were presenting sponsors of the event, which attracted more than 7,000 registered participants and raised $762,245. Participants were encouraged to support Children’s by donating an additional amount beyond the registration fee, or creating or joining an existing team committed to raise funds for a specific cause. Each team – there were 212 – chose a program at Children’s for their fundraising dollars. Teams of grateful patient families, friends and even the Children’s caregivers themselves supported cancer, heart disease, organ transplantation, gastroenterology, allergies, the Children’s Medical Center Research Institute at UT Southwestern, and more. Thank you to all who participated, including Joel T. Williams III and his Passion for Children’s organization, which conceived the event and managed the race details with support from our Children's Health team.

2016 CHILDREN HELPING CHILDREN JUNIOR SINGLES TENNIS TOURNAMENT More than 500 junior singles tennis players served up their best game at the 26th annual “Children Helping Children” Junior Singles Tennis Tournament, presented by GEICO and benefiting Children’s Health.

The games began Sept. 24-25 at High Point Park Tennis Center in Plano. The players and their families raised $100,000 to support the Pauline Allen Gill Center for Cancer and Blood Disorders, where more than 1,000 new patients are cared for each year, including 1 in 5 children in Texas with cancer. Since its inception, the tournament has raised more than $3.1 million for pediatric cancer. With 24 pediatric hematologists and oncologists, the Gill Center is a world-class pediatric cancer treatment center recognized nationally for exceptional clinical care, leadership in pediatric cancer research and academic excellence.

2016 RADIOTHON The 2016 Christmas is for Children Radiothon, which supports Children’s Health, raised its biggest-ever total of $1,262,704. One hundred percent of the funds raised over the two-day period supports programs at Children’s Health. In partnership with Children’s Miracle Network (CMN) Hospitals, CBS Radio DFW’s 98.7 KLUV, La GRANDE 107.5 FM and NewsRadio 1080 KRLD produced live broadcasts from the hospital. Jody Dean and El Chiquilin hosted a marathon 26 hours each over the two-day period. La GRANDE 107.5 FM alone raised a record-breaking $558,199, making it the No. 1 CMN Hispanic Radiothon in the country. Twenty-six patients and families shared their heartwarming stories via pre-recorded interviews or live appearances with on-air personalities from all three stations and inspired listeners to support Children’s Health. The Christmas is for Children Radiothon was made possible by the generous support of local sponsors, including FairLease (presenting sponsor), Credit Union of Texas (phone line sponsor), Neighborhood Credit Union (child champion sponsor), Albertsons-Tom Thumb, The Children’s Courtyard, Granite Properties, Padrino Foods, Skanska, ReTrak, Dallas Fort Worth Acura Dealers, Blue Cross Blue Shield of Texas, ADT Security, Cisco, Marquis Group, UBC and El Rio Supermercados.

WOMEN’S AUXILIARY TO CHILDREN’S MEDICAL CENTER The Women’s Auxiliary to Children’s Medical Center continued its rich history of service and tradition in 2016.

More than 850 members strong, the dedicated and committed members of the Auxiliary serve as advocates in the community, volunteers in the hospital, and raise funds through annual events and programs.

Throughout the year, the Auxiliary coordinates a variety of projects and programs to support the patients, families and staff at Children’s Health. Auxiliary members volunteer and serve dinner to patient families staying in the hospital during the holidays, deliver monthly snack baskets to critical care waiting areas, and provide much needed comfort through the distribution of necessity items during an unexpected hospital stay. The Auxiliary also recognizes and celebrates the staff at Children’s Health with seasonal treats and special recognition. Through holiday events such as Breakfast with Santa and Santa’s Workshop, and signature spring events – Family Night at Six Flags and Legacy of Love Benefit & Fashion Show – the Auxiliary raises funds for priority areas at Children’s Health.

During its 55 years of service, the Auxiliary has supported crucial initiatives at Children’s Health, including Family Support Services, research, intensive care, critical training programs and lifesaving equipment acquisition, as well as establishing endowments to sustain and enhance essential programs and services for patients and families for years to come. Since its inception, the Auxiliary has contributed $19 million to Children’s Health.

The Women’s Auxiliary was established in 1962 by combining the auxiliaries from Bradford Memorial Hospital for Babies, Children’s Hospital of Texas and Richmond Freeman Memorial Clinic. In the very early days, Auxiliary volunteers assisted hospital staff in all aspects of running the hospital, from helping with admitting patients and running samples to the lab to assisting physicians. Today, the impact and contributions of the Auxiliary can be felt across Children’s Health in countless ways.

THE CHILDREN’S TRUST In 2016 The Children’s Trust raised $91,900 for Children’s Medical Center Foundation, bringing the total raised since its establishment in 2002 to more than $1.5 million. A unique aspect of The Trust is that members vote on which programs to support each year. Called the Big Give, this annual event provides members the opportunity to personalize their giving by learning about areas in need. This year, The Trust provided funding for the Suicide Prevention and Resilience in Children (SPARC) program, a telehealth cart for the School-Based Telehealth program and transcranial Doppler equipment for the Neuro-Concussion program.

The Trust was founded by two dedicated couples, Sheila and Dave Beuerlein and Betty and Brian Schultz. Both had a history of supporting the Children’s Health mission and wanted to encourage their friends, colleagues and community members who had a strong interest in children’s health care to become involved with the hospital through philanthropic, educational and social activities. It has grown to include nearly 200 members. In addition to the Big Give vote reception, members are invited to attend educational and social events. You can join The Children’s Trust at the Individual ($250), Couple ($500) or Red Wagon ($1,000 or above) level. Your entire membership fee is a tax-deductible gift to Children’s Medical Center Foundation.

JUNIOR LEADERSHIP BOARD The Junior Leadership Board (JLB) was established in 2010 by Caroline Cooper, a teen who saw an opportunity for high school students to make a difference for Children’s Health. The JLB is a junior board to the Children’s Medical Center Foundation Board of Directors with a mission to connect high school students with Children’s Health through a variety of activities, including volunteering, fundraising, learning about careers in the hospital system, and much more. Now in its seventh year, the JLB has grown from a board of 40 members serving both the Dallas and Plano campuses to two boards, one for each hospital with more than 30 board members on each. The boards comprise high school students from a variety of North Texas schools. Board terms run from September through May, with meetings held throughout the course of the year. Board members, who must be at least 16 when they start their service term, are expected to volunteer at special events for the hospital, help fundraise, and inspire others to become involved. The JLB’s key fundraising activities include an annual charity mum ribbon sold through The Mum Shop in Plano, and the JLB’s Red Balloon Run & Ride team, Teens for Children’s Health. Since the board’s inception in 2010, members have raised more than $90,000 for Children’s Medical Center Foundation.

Children’s Health welcomes five new board members appointed in 2016

Children’s Medical Center Foundation

GEORGE BILLINGSLEY George Billingsley began his career with the family-owned Billingsley Company in 2007.

He started with the Dallas-based land development and property management company as a leasing agent at The Austin Ranch Apartment Development and proceeded to move to the corporate office, where he served as multifamily financial analyst and, later, asset manager.

In 2010, Mr. Billingsley moved to the industrial asset class, where he served as vice president of industrial leasing. He was promoted to partner in 2012 and now serves in an administrative role in which he is responsible for portfolio management, development, leasing and financing.

Mr. Billingsley earned his Bachelor of Arts from Dallas Baptist University and his Master of Science in management from Southern Methodist University. He serves as a member of the Urban Land Institute.

“I am excited to serve on the Foundation board because I admire the many skilled professionals who work at the hospital, and I appreciate the great difference this organization has made in the lives of many children and their families. I look forward to bringing my real estate background to the board and looking for ways to contribute.”

JOHN CARONA A lifelong Texan and graduate of the University of Texas at Austin, John Carona is the chairman and CEO of Dallas-based Associa, a company he founded in 1979. Associa is North America’s largest community association management company, with more than 180 offices in the U.S., Canada and Mexico and more than 10,000 employees. In addition to his real estate business, Mr. Carona has been involved in the banking industry for several decades. He acquired First National Bank of Kemp in Kemp, Texas, in 2015 and currently serves as a member of the board of Pacific Premier Bank of Irvine, Calif.

Mr. Carona served nearly 25 years in both the Texas House and Senate, where his leadership roles included serving as chairman of the Senate Transportation and Homeland Security Committee and chairman of the Senate Business and Commerce Committee. He is a former president pro tempore of the Texas Senate, author or chief sponsor of more than 700 bills that became law, and the recipient of over 200 public service awards.

“As a resident of Dallas for more than 50 years, I’ve long seen the positive impact Children’s Medical Center has on our community, and as parent of five and grandparent of seven, I have experienced firsthand the talent and compassion of its doctors and nurses. It is truly a privilege for me to give back to this fine institution by serving on its Foundation board.”

MICHAEL K. PICKENS Michael K. Pickens is the president and CEO of Pickens Energy Corp. While focused on the energy industry for 90 years, the family company has diverse holdings in real estate, timber, and early-stage technologies, ranging from next generation batteries and remote IOT monitoring and control to wireless towers. Mr. Pickens is a Dallas native, with a Bachelor of Arts from Duke University, and a Master of Business Administration from Southern Methodist University. Michael and his wife, Heidi, have three boys under the age of 6.

ANNE BAVIER Having spent her career in the health care industry, Anne Bavier’s leadership and dedication began years ago.

Dr. Bavier is the dean of the College of Nursing and Health Innovation, the largest academic unit at the University of Texas at Arlington. A leading voice on health care, Dr. Bavier is the president of the National League for Nursing, an organization of nearly 40,000 faculty nurses and leaders in nursing education. Earlier this year, Modern Healthcare nominated her for its 100 Most Influential People in Healthcare list. Dr. Bavier holds an undergraduate degree in nursing from Duke University, a Master of Nursing from Emory University and a doctoral degree from Duquesne University.

“Children’s Health and UTA’s College of Nursing and Health Innovation have a longstanding relationship. Over the years, many of our nursing students have done their clinical rotations at Children’s Health and taken full-time jobs there upon graduation. Serving on the board will further strengthen this relationship. I am honored to contribute further to our partnership by sharing my perspectives on nursing and nursing education, as well as bring my extensive experience on boards to the benefit of Children’s.”

HIAWATHA WILLIAMS Hiawatha Williams is the founder and CEO of Williams Chicken. The Dallas-based fast casual chain has over 40 stores, including 20 franchises, and is responsible for more than 500 employees. Mr. Williams’ popular mantra – dedicated to serve, grow and give back to the community – resonates with the company’s work ethic and unmatched “give-back spirit” supporting major community outreach in the area of education, youth programs and corporate philanthropy. Mr. Williams earned his degree from Paul Quinn College.

“When I received the invitation to serve Children’s Health, it didn’t take long for me to realize that this organization’s mission – to make life better for children – aligns with my life’s mission: to serve others, especially children and youth. I humbly accepted this appointment and am committed to do my part to help Children’s Health become the premier provider for children in our country.”

PHILANTHROPY HIGHLIGHTS The opportunity to provide hope and healing to a sick child is profound. We are deeply grateful for the organizations and individuals who took that opportunity and supported our mission in 2016.
