Evolve®D chromatography platform overview
Evolve®D columns are advanced single-use chromatography systems developed to support the evolving demands of modern biopharmaceutical purification processes. Available process formats include column diameters ranging from 7 cm up to 60 cm, with a broad and flexible range of bed heights to support both development-scale and manufacturing-scale applications (Figure 1). The platform is designed to enable seamless scalability while maintaining consistent packed-bed hydrodynamic performance and robust operational reliability.
The platform combines scalable packed-bed performance with a fully non-metallic flow path architecture (Figure 2), reducing the risks associated with corrosion, contamination, and biomolecule inactivation. By eliminating the need for column cleaning and validation, Evolve®D columns streamline manufacturing operations, reduce process downtime, and improve overall operational efficiency.

Column installation and preparation
Installation of the Evolve®D platform has been designed to be simple, efficient, and manufacturing-ready, supporting rapid deployment into downstream processing operations. Columns are removed from protective packaging and positioned on a level surface before sanitary caps are removed and the system is connected to a primed chromatography skid using 25 mm tri-clamp fittings.
Following installation, the column is flushed using equilibration buffer at approximately 50% of the intended operating flow rate until pH and conductivity stabilize. Optional conditioning steps may then be performed using reverse-flow and down-flow equilibration cycles prior to process operation.
The streamlined installation workflow minimizes preparation time, reduces operator intervention, and supports efficient implementation within GMP manufacturing environments while preserving process consistency and operational reliability.

Materials and methods
1. Packing methodology
MiMode PuraBead® CX2 was packed into Evolve®D columns using Astrea Bioseparations’ standard compression packing methodology in an ISO 7 cleanroom suite. The resin was packed using 0.1 M NaCl as the packing solution with a compression factor of 1.2. Slurry concentration was determined using settled bed height analysis within small-scale C10 columns prior to scale-up, with the slurry typically prepared at approximately 50% settled concentration.
The packing methodology was designed to maximize packed-bed stability, hydrodynamic efficiency, scalability, and operational robustness while keeping low operational backpressure across all evaluated process formats.
2. Column performance testing
Following packing, columns underwent standard chromatographic efficiency testing using pulse injection methodology.
