Sexcel™ Sexed Genetics - Straw Handling Guidelines At ABS Global, the experience when using our genetics is a priority. Every semen collection from ABS Global sires undergoes a series of rigorous quality control evaluations to ensure that every straw sold meets our high standards.

To get the best possible results from using Sexcel™ Sexed Genetics, please follow our heifer rearing protocols, and use the following straw handling protocols:

1.

Straw identification and transfer

• Use the tank inventory card to identify the goblet location containing the straw.

• Never lift the goblet above the frost line in the neck of the tank.

• If the straw is not identifiable within 5 seconds, lower the goblet back into the liquid nitrogen for at least 10 seconds before recommencing identification.

• Use the appropriate tweezers for 0.25cc straws, never fingers, to pick up straws.

2.

Thawing procedure

• ABS Global recommends thawing only one straw of Sexcel at a time as the objective is to thaw

the number of straws that can be inseminated within 5 minutes of thawing.

• Replace the thaw bath water daily and ensure the temperature is between 35C to 37C (95F to

98F) using a thermometer.

• Thaw the straw in the water bath for 30 seconds.

• Remove the straw from the water using tweezers and protect from the environment using a clean, dry paper towel. Use the paper towel to dry the straw.

3.

Move the air bubble

• Move the air bubble upwards toward the crimped end of the straw. If the bubble is not moved, 1-5% of the sperm will be lost.

• Ensure that there is minimal time between removing the straw from the thawing bath and loading it into the AI gun, and that you protect the straw from cold wind and sunlight.

4.

Load the AI gun

• Place the thawed straw into the pre-warmed AI gun with the plugged end first. An electronic AI gun is recommended.

• Cut the straw at 90 degrees just below the crimped end, with clean scissors or a straw cutter.

• Place the plastic sheath over the loaded AI rod. Ensure that the straw enters and adapts well inside the green or blue plastic piece of the sheath.

• The use of a plastic ‘chemise sanitaire’ is recommended on top of the AI gun sheath.

5.

Number of straws per heifer

• Inseminate one complete straw per heifer, do not split straws.

Note: Sexcel may be used solely for single artificial insemination of one female bovine during natural ovulation with the intent to produce a single offspring.

Sexcel™ Success - Heifer Rearing To maximise the best possible chance of success when using Sexcel, heifers should be settled, stress free and reared successfully to an optimal weight and body condition score.

1.

Rearing management goals

• Complete any veterinary recommended preventative treatments such as vaccinations disease or anthelmintic treatments at least 4 weeks prior to breeding.

• Keep the heifer group in the same group and environment (breeding pen) for 6 weeks prior to and following breeding.

• Keep heifers on the same diet for the 6 weeks prior to and following breeding;

i.

Increasing the nutritional value of the ration prior to breeding is advisable under the

guidance of a nutritional consultant.

ii.

Special attention to levels of copper, zinc, selenium, iodine and vitamins A and E are

needed over the 12 week period before service.

• Avoid new heifers entering the group during the breeding period where possible.

• Reach 55-60% of target mature weight at start of planned breeding (330-360 kg for an animal expected to calve in at 600kg).

• At least 12 months of age.

• Target body condition score of 2.5-3.0 on the 1-5 BCS sale at service.

• Growth rates of 0.75-0.8kg/day are recommended for Holstein Friesians from birth to calving.

• Target 90% of mature weight at calving.

2.

Breeding period

• Observe heifers for at least 3 weeks prior to breeding to ensure normal oestrous events are occurring.

i.

Observing 2 natural heat events is best.

• If no oestrous is expressed prior to start of breeding or the heifer has not expressed oestrus within 4 weeks of eligibility for insemination, present to a veterinary for examination.

• Accurately identify heifers that are eligible for service.

• Best results are obtained when inseminating heifers following excellent heat detection accuracy.

• Use a safe and low stress handling facilities for restraining heifers for insemination.

• Retain heifers in the breeding group until 6 weeks post breeding and when confirmed pregnant.

• Accurately record the identification of the heifer inseminated and Sexcel product used.
