Role of novel miRNAs in NAFLD

NAFLD is a liver disease in which different types of fats gets accumulated in liver causing NASH (a type of NAFLD). Prevalence of NAFLD in India is 9% in rural population and 32% in urban population. In Asia, 1-2% of the HCC cases are because of NAFLD/NASH. Patients with NAFLD/NASH are at a higher risk of developing hepatocellular carcinoma. From multiple studies, it is evident that microRNAs (miRNAs) play a vital role in pathophysiology of NAFLD.

ABSTRACT Background: NAFLD is a chronic liver disease observed in people who do not abuse alcohol. Accumulation of fats such as triglycerides in the liver lead to NASH, which is the more serious outcome of NAFLD. Rationale: miRNAs play a major role in complex diseases by undertaking post-transcriptional regulation of genes. Strategy: A subset of genes causing NAFLD was selected from literature and in silico analysis carried out using tools like miRDB and DIANA microT for putative miRNA binding sites followed by miRTarBase for miRNA- target interactions. Results: miR-7 & miR-548 family members were found in all 3 programmes and targeted genes involved in liver function. Conclusion: miR-7 & miR-548 family members probably play a vital role in pathophysiology of this disease and can be used as prognostic / diagnostic markers for NAFLD

miR-7 family members are antioncogenic miRNAs and its repression can cause HCC. miR-548 family members are also involved in various pathological processes like cancer/signalling pathways and it promotes tumorigenesis, specially HCC cell proliferation and apoptosis. Various studies have shown association between NAFLD and HCC. Need of the hour in NAFLD and its related complications is the elucidation of new classes of biomarkers. miRNA candidates can serve as a novel approach towards both biomarker discovery as well as understanding NAFLD pathophysiology.
