Precis provided architecture, process architecture and mechanical, electrical, plumbing, fire protection, and process engineering design services for the interior renovations and facility expansion of Piramal’s Lexington, KY facility. The facility is situated on a 12-acre site, giving it sufficient land area to expand the existing building’s footprint and site features to meet the current need for expanded fill/finish capacity. The 16,000 SF expansion included a Grade C Fill/Finish suite with associated Grade C support areas and Grade D equipment staging and CIP/SIP rooms. Renovations to the existing building included the 2,600 SF microbiology lab suite with CNC laboratory and support areas, Grade D gowning rooms, and a Grade C Sterility Testing Room.

411 Innovations Lab Complex

Precis was tasked with the programming of the facility, which involved multiple buildings within the 411 Innovation Labs Complex. The programming included the following manufacturing and testing areas: Viral Vector Manufacturing, Cell Therapy Manufacturing, Filling Manufacturing, Plasmid and MRNA Manufacturing, Testing Labs (Myco, Molecular, QC, NGS, Virology, Flow Cytometry, Environmental Monitoring), and Warehouse and Support Areas. Precis was responsible for defining the requirements from the users to be able to develop a conceptual floor plan and equipment layout.

Expansion
Nashville, TN
Precis provided MEP/FP and process architecture services for a 20,000 SF expansion to the existing facility in Nashville, TN. The expansion included the addition of two filling suites with automatic filling lines that use full isolator technology. One of the filling suites is fully equipped with a combination vial/syringe filling line and lyophilizers. The second filling suite was designed to accommodate one combination filling without lyophilizer capability. The expansion also included multiple process support areas and a GMP warehouse. The facility design accommodates potential future expansion plans.
IFrontage Laboratories
Sierra Drive Fit-Out Exton, PA
Precis provided architecture and mechanical, electrical, plumbing, fire protection, and process engineering design services for the relocation and expansion of Frontage’s existing pilot scale production facility into a new building. The expansion was designed to support Phase I and Phase II Clinical Products. The fit-out covered 46,000 SF and provided a sterile area consisting of two sterile filling suites for multiproduct processing of sterile products into vials, syringes, and bottles. Additional ISO 7 spaces included sterile equipment staging and MAL + PAL airlocks. Each filling suite is ISO 7 with an interior ISO 5 area for filling. Also included was a potent compound area consisting of two potent suites for OEB category 3 – OEL 50 to 100 g/ m3 compounds. Support areas include analytical and microbiology labs, equipment wash, clean equipment staging, GMP warehouse, mechanical rooms, and offices.

Augusta USG Aseptic Production
Upgrades Lenoir, NC
Precis performed a peer review of the interior cleanroom build-out within the existing high-bay structure. Based on cGMP/Annex 1 guideline standards, extensive design modifications to address aseptic personnel and material flows, and process room segregations were recommended. Precis is presently undertaking the design and documentation of necessary modifications in a phased delivery to provide required airlocks and exit corridors addressing cGMP planning recommendations. Phases 1 and 2 include improvements to two compounding and isolator-based vial filling lines. Phases 3 and 4 will focus on planning one existing and two syringe filling lines, terminal sterilization suite, and manufacturing support areas.

Sterile Manufacturing Facility Central, NJ
The design for this project consisted of partial demolition of an existing structure and creation of a new multi-level building to provide space for process equipment and new central utilities. The facility incorporates all functions necessary for sterile production, including warehousing, filling, and packaging. The processing areas are comprised of pre-engineered cleanroom modular panels with walkable ceilings. The process rooms house multiple compounding systems which are followed by a fill line isolator and lyophilizers. All new central utilities, including new electrical service, backup generator, steam, chilled water, and clean utilities were added as part of the project scope.

Sterile Production Area Design Lakewood, NJ
Precis developed the master plan for a new 20,000 SF Compounding and Sterile Filling GMP Multi-Suite Core. Stage one of the master plan implementation entailed the fit-out of the first of three Grade B Small Volume Parenteral (SVP) Sterile Filling Suites with a central aseptic corridor, and Grade D Core perimeter. The Sterile SVP Suite consisted of a 700 SF Grade B sterile fill and primary packaging room, and a 900 SF Grade D assembly, and a vial load and tray off space. A new catwalk and mezzanine to support above ceiling utility and maintenance needs for MEP and process utilities was entirely coordinated using state of the art BIM technology.

Grade C Compounding + Filling Suite
Lakewood, NJ
This project consisted of the expansion of the existing Grade C filling capabilities to support a manufacturing contract for compounding, filling, and packaging of a bio-dosage nasal product with a potent active pharmaceutical ingredient (API). The project renovated the existing process areas to create two new filling suites and one compounding suite, and their associated technical support spaces. In addition, an existing secondary packaging area was enclosed to control access to new secondary packaging equipment. This project utilized pre-engineered modular wall and ceiling panels with honeycomb core and PVC coating and epoxy urethane floor system with an integral cove base integrated with the modular wall panels. An elevated steel dunnage platform extension above the existing roof was provided to house the new chilled water, process chilled water, and air systems infrastructure.

Precis provided mechanical, electrical, and process utilities engineering design to construct an 11,000 SF mixed-use facility that included new sterile fill/finish manufacturing areas focused on the contract manufacturing business model. Key components of the project included chemistry laboratory areas, component prep area, sterile fill/ finish rooms, office and administration areas, new WFI generation and distribution, new clean steam generation and distribution, autoclaves and dry heat ovens, and utility and HVAC for new fill/finish equipment.

Precis provided mechanical, electrical, plumbing, fire protection and life safety, and process engineering services for a new building addition and renovation of 40,000 SF of GMP multi-product biological pilot plant space. Renovations were required to upgrade the suites to comply with quality and regulatory agency requirements. The design included a new second story locker room addition, fire water containment, aseptic isolator, airlock and suite reconfiguration, and renovation of existing utilities such as WFI, clean filtered gasses, HVAC, and electrical.

The key to a project’s success starts with strategic planning, and a welldesigned and thoughtful site plan analysis is an important element in our project service approach.
Civil Engineering does not account for all the critical aspects of a thorough site analysis — it is the architect’s role to consider elements such as solar, wind, views and other site features that help to properly orient the building on a given site. We often get involved early to help our clients with site evaluations prior to final land acquisitions. What might look like a viable building site on paper may not be ideal for the many other considerations we explore.
