Competent cells are ready to use bacterial cells that have been chemically treated to allow incorporation of foreign DNA/plasmids. Gentrox has a variety of chemically competent and electrocompetent cells for transformation of DNA with the highest transformation efficiencies. Gentrox's rapid transformation protocol is shorter by more than 60 minutes as compared to a standard protocol while maintaining optimal efficiency. We offer a range of cells which can be used for blue white screening or routine screening. Our cell strains offer blue/white selection, transformation of methylated DNA and T1 phage resistant transformation.

Rapid Protocol for Optimal Transformation

Standard Protocol for Optimal Transformation:

Thaw cells: Place the microcentrifuge tube containing competent cells on ice to thaw the cells. Add plasmid DNA, gently mix the contents and keep at room temperature for 2 minutes. No incubation or heat shock is necessary.

Thaw cells: Place the microcentrifuge tube containing competent cells on ice to thaw the cells. Add plasmid DNA, gently mix the contents and keep the tube on ice for a specified time.

Plating cells: Preferably use warm agar plates with appropriate antibiotics for plating. Incubate for approximately 16 hours at 37°C.

Incubation and Heat shock: For optimum transformation efficiency, incubate the cells with DNA for about 20 to 30 minutes and heat shock for 30 seconds at 42°C or 5-10 minutes at 37°C. Cell Growth: Add growth media (1:4) and incubate at 37°C in a shaker. Supplied growth media should be preferred to other standard media like LB. Plating cells: Preferably use warm agar plates with appropriate antibiotics. Incubate for approximately 16 hours at 37°C.

Satellite Free Ampicillin

• More potent than traditional antibiotics • Perfectly optimized for gene expression and recombinant studies • Suppresses growth of satellite colonies following transformation, making colony counting and picking much easier • Available in pre-packed 1ml, 1,000x concentrate Gentrox introduces Satellite-Free Ampicillin which is a specially formulated antibiotic, optimized for gene expression and cloning. Satellite-Free Ampicillin is an advanced version of frequently used antibiotics such as Ampicillin, Carbenicillin and Methicillin. These frequently used antibiotics are degraded over the time by the enzymes secreted by the bacteria resulting in undesired satellite (non-recombinant) colonies. Gentrox Satellite-Free Ampicillin is uniquely formulated so that it is resistant to bacterial enzyme degradation even after prolonged incubation at 37°C resulting in Satellite-Free colonies.

X-Gal and IPTG are routinely used in screening protocols to identify recombinants and to calculate transformation efficiency. Gentrox offers a validated mixture of XGal and IPTG that is optimized for screening. The product is offered in a convenient mist spray allowing for direct application to your agar plates. Usage: Blue white screening of bacterial colonies. Each bottle is enough for 100 plates (100 x 15mm size). Brief Protocol: Spray the mix 4 times (equivalent to approximately 200µl) on an agar plate while maintaining a distance of 7 - 12 cms between spray bottle and plates. Leave the plates in the hood until all the solution is absorbed. If necessary, use a spreader to facilitate uniform absorption.

Note: Perform the operations in a safety hood, wearing safety glasses and gloves. An LB agar plate showing the result of a blue white screening. Competent cells were transformed with white/blue colony making pUC vectors. Cells were poured on Ampicillin agar plates which were sprayed with mixture of X-Gal/IPTG

TCEP HCl is an alternative for Beta Mercaptoethanol (BME)/dithiothreitol (DTT). Unlike DTT or BME, odorless TCEP can be conveniently used for reductions in a healthier lab environment. It is compatible with all common biological buffers used for protein purification and conjugation. (e.g. 6 X His/GST-tagged protein purification). TCEP selectively and completely reduces most stable water-soluble disulfides bonds in less than 5 minutes even at very low concentration (5 mM).
