Apoptosis through Death Receptors from BioLegend Apoptosis Apoptosis is also known as programmed cell death, a mechanism by which metazoans can control their tissue development and eliminate individual cells that may harm the animal’s survival. A number of cell surface receptors that respond to external death signals can initiate apoptosis in mammalian cells. One such group of receptors is the TNF (Tumor Necrosis Factor) superfamily, otherwise known as Death Receptors. Currently, the members include TNFR1 (Tumor Necrosis Factor Receptor-1), Fas (also known as DR2, APO1, and CD95), DR3 (Death Receptor-3), TRAILR1 (TNF-Related ApoptosisInducing Ligand Receptor-1, also known as DR4 and APO2), TRAILR2, DR6, EDAR (Ectodysplasin-A Receptor) and NGF (Nerve cytoplasmic region termed the DD (Death Domain) that serves as the recruitment site for intracellular ligands upon receptor activation. A parallel set of proteins that inhibit apoptosis through death receptors soluble form, can compete for binding to death ligands, but do not have the DD, and thus inhibit death signals to the cell. Some of the known DcRs include TRAILR3 (also known as DcR1), TRAILR4 (also known as DcR2), and DcR3. FAS is the prototypical death receptor expressed widely on leukocytes. Intracellular proteins including FAF-1 (FAS-Associated Factor-1), FADD (FAS-Associated Death Domain), DAXX, FAP-1 (FASAssociated Protein- Tyrosine Phosphatase-1), and RIP (ReceptorInteracting Protein) directly interact with the FAS cytoplasmic DD. Signaling complexes formed by FAS, TRAILR1, and TRAILR2, capable of inducing death, are known as the DISC (Death-Inducing that recruits Procaspase-8. Cleavage of Procaspase-8 produces active Caspase-8, which initiates a cascade ultimately leading to the release of CytoC (Cytochrome C), a factor that interacts with APAF-1 and Caspase-9. Active Caspase-9 triggers Caspase-3 cleavage of a variety of proteins, ultimately leading to apoptosis. (TNFR-Associated Death Domain), FADD, or ASK1 (Apoptosis Signal-Regulating Kinase 1). It can also trigger a variety of downstream cascades including the Caspase-8 pathway like that of FAS, or the NF-κB and the JNK1 (c-Jun kinase) pathways. Understanding Death Receptor signaling provides potential for cures Alzheimer’s. Our understanding of selectively activating DRs could lead to alternative therapies for tumors, particularly those that have lost p53 function.
