* Please note that the above table represents approximate indicators only. All expected detection times depend on the analyte, including: metabolic process (blood/saliva/sweat/urine), exposure to chemical, biological or physical stress (invisible traces/surfaces) and food/drink consumption (urine).

Test Procedure:

Evaluation of Results:

The test should be performed immediately after opening the protective pouch, because the test is humidity-sensitive. Refrigerated cassettes, controls and any refrigerated patients’ samples should be brought to room temperature (15-30°C) before opening and prior to testing to avoid condensation of moisture on the test. Ensure that a sufficient quantity of specimen has been collected.

Negative: Both control (C) and test (T) lines appear. The test line (T) is the drug line; the line at (C) is the control line, which is used to indicate proper performance of the strip. The colour intensity of the test line may be weaker or stronger than that of the control line.

1

Open the pouch and remove the cassette. Once opened, the cassette must be used immediately.

2

Draw the urine sample to the line marked on the pipette.

3

Dispense 3 drops into the sample well. Use a separate pipette and device for each sample or control. Start the timer. Caution: If the urine directly touches the reaction zone, the test becomes invalid!

4

Note: A very faint line in the test zone indicates that the drug in the sample is near the cut-off level of the test. These samples should be re-tested or confirmed with a more specific method before a positive determination is made.

Read the results at the end of 5 minutes. Do not interpret the result later than 15 minutes after starting the assay.

T: +44 (0)1730 829 202

Positive: Only the control line (C) appears. The absence of the test line (T) indicates a positive result. Keep the sample if confirmation testing is required.

Invalid: No line appears in the control zone (C). Under no circumstances should a positive sample be identified until the control line forms. Repeat the assay with a new test device, paying special attention to the required amount of liquid and reaction time. If problems persist, please contact our client services team.

Multiline Cassette Tests

Test Procedure:

Test device, patient’s samples, and controls should be brought to room temperature (15-30°C) prior to testing to avoid condensation of moisture on the membrane. Do not open pouches until ready to perform the assay. 1

Remove the test device from its protective pouch. Label the device with patient or control identification information.

2

Dispense 3 to 4 drops into each sample well. Be aware that no urine comes into direct contact with the reaction zone, as this will invalidate the test.

Evaluation of Results:

Negative: If you have analysed a drug-free sample for each drug, a test line together with the control line (C) appears at each reaction zone. The line in each test region (1, 2 & 3) is the drug probe line; the line in the control region (C) is the control line, which is used to indicate proper performance of the device. The colour intensity of the test line may be weaker or stronger than that of the control line. Figure B illustrates negative results for all drugs tested, as both control and test lines (1, 2 & 3) are visible in each reaction zone. This is irrespective of how faint the lines appear.

Figure A shows a cassette with two strips (reaction zone) and two sample wells. 3

Read results after 5 minutes, when the control line has appeared at the reaction zone of each strip. The control line should appear after 5 minutes at the latest.

4

Do not read results after 15 minutes.

Important note: Do not quantify the intensity of the test lines. Positive: Only the control line (C) is visible and the test lines (1, 2 & 3) are invisible. Figure D below shows that the reaction zone “a” is positive for the drugs “1”, “2” and “3”. Some reaction zones may not have all test lines available for testing. Figure D shows in reaction zone “b” the testing of drugs “1” and “3”, obtaining positive results. Each test line should be evaluated separately. That means, not all parameters will be positive at the same time in one test. An invisible test line indicates a positive result for the corresponding drug only. The control line must be visible in all cases. Mark the positive results after evaluation time (after approx. 5 minutes). A very faint line in the test region indicates that the respective drug in the sample is near the cut-off level of the test. These samples should be re-tested or confirmed with a more specific method (e.g. GC/MS) before a positive determination is made. Invalid: If no line appears in the control region (C) after approx. 5 minutes, the test or rather the respective parameters are invalid and the test should be repeated. Important: the test must not be evaluated if the control line is absent! Figure E illustrates invalid results for the drugs “1”, “2” and “3”, because no control line (C) in this reaction zone is visible.

Alcohol Testing

Alcohol can be detected in breath, saliva, urine, hair and blood. The easiest method is taking a breath sample which needs to take place whilst following strict Chain of Custody procedures and completing specialist testing forms. A fully calibrated breath testing instrument can be defensible in Court, for a quick indication, an instant screening device can also be used. Breath Alcohol Testing

The quickest and easiest method of alcohol testing is via a breath sample. Chain of Custody procedures including the completion of a breath alcohol testing form are essential. The findings can be defensible in Court when using a calibrated testing instrument.

Limitations with breath samples are the need to use a calibrated testing device and the short detection window. However, impairment at work can be substantiated. Alcohol detection in urine and hair is far longer and can give you information on alcohol abuse.

Oral Fluid Test

Rapid Saliva Alcohol Test Strip is intended for use as a rapid method to detect the presence of alcohol in saliva for blood alcohol concentration (BAC) greater than 0.02%. It has been published that the concentration of alcohol in saliva is almost equal to that in blood. The rapid test is intended for the semi-quantization of ethyl alcohol in human saliva. To confirm the concentration of positive specimens, a calibrated breath alcohol instrument or urine Laboratory test should be used.

Chain of Custody

The Chain of Custody defines a set of procedures from when a sample is provided, during the analytical process and until its final destruction.

If a screened sample is positive, a Chain of Custody sample must be taken and sent to an accredited (e.g. UKAS) Laboratory for expert GC-MS or LC-MS analysis. This process is defensible in employment hearings and known as the ‘Gold Standard’ for use in Court. Medical Review

Medical Review is an integral part of any confirmation testing and laboratory service. Beacon has its own independent Medical Review Officer who discusses any medication taken by the donor which could impact upon the final result. Reporting of Results

Screening results are available from the laboratory within 24hrs and Confirmations take a further 2-5 working days. Results can be sent securely via email, telephone on online via the secure customer specific portal.

Anti-HCV (HCV-Ab) Test is a rapid chromatographic immunoassay for the qualitative detection of antibodies generated against proteins that are encoded by conserved sequences of core, NS3, NS4, NS5 parts of HCV genome in human whole blood / serum / plasma. Anti-HCV Test can detect antibodies generated against proteins that are encoded by conserved sequences of CORE, NS3, NS4, NS5 parts of HCV genome. Anti-HCV Test uses solid-phase immunochromatographic technology for the qualitative detection of antibodies against HCV antigens in human whole blood / serum / plasma. The test is a two-site immunometric assay in which a combination of monoclonal antibodies and recombinant HCV antigens (CORE, NS3, NS4, NS5) are used to selectively detect antibodies against HCV antigens in samples with a high degree of sensitivity.

HIV

Anti-HIV 1/2/O Test is a rapid qualitative immunoassay for the detection of antibodies (IgG, IgA and IgM) generated against all subtypes of Human Immunodeficiency Virus Type 1 (HIV-1) (including Group O) and Type 2 (HIV-2) in human whole blood / serum / plasma. Anti-HIV 1/2/O Test can detect antibodies (IgG, IgA and IgM) generated against all subtypes of Human Immunodeficiency Virus Type 1 (HIV-1) (including Group O) and Type 2 (HIV-2). Anti-HIV 1/2/O Test uses solid-phase immunochromatographic technology for the qualitative detection of anti-HIV in human whole blood / serum / plasma. The test is a two-site immunometric assay in which a combination of monoclonal antibodies and recombinant antigens (include gp120, gp41, gp36, p24) are used to selectively detect anti-HIV in samples with a high degree of sensitivity.
