If the cell line has been exposed to other cells it may mutate into another form and the experiment is no longer prevalent. It is important if establishing a tissue culture and it helps to keep the cells healthy and actively growing. At this point surface to surface contact with other cells causes them to inhibit growth. Subculturing is important at this juncture to ensure that the individual cells are receiving the proper media innoculant. Confluency lets the scientist know when it is time to subculture because the cells have proliferated enough to become a detriment to one another.

Trypsinization - The enzymatic dissociation of cells from their culture vessels during subculture. In order to remove the cells from the walls of the container or slide, trypsin must be used. It can be supplemented with enzymes, EDTA, or chelating agents. The ideal situation is to dissociate and separate cells in the shortest amount of time with the least harmful method possible.

Wipe hood and all surfaces with 70% ethanol. Make sure the work place is cleared of clutter under the hood. The work area should be far from drafts and traffic flow.
